Tetracycline-inducible protein expression in pancreatic cancer cells: effects of CapG overexpression

AIM: To establish stable tetracycline-inducible pancreatic cancer cell lines. MeTHODS: Suit-2, MiaPaca-2, and Panc-1 cells were transfected with a second generation reverse tetracycline-controlled transactivator protein (rtTA2S-M2), under the control of either a cytomegalovirus (CMV) or a chicken β-actin promoter, and the resulting clones were ***: Use of the chicken (β-actin) promoter proved superior for both the production and maintenance of doxycycline-inducible cell lines. The system proved versatile, enabling transient inducible expression of a variety of genes, including GST-P, CYP2e1, S100A6, and the actin capping protein, CapG. To determine the physiological utility of this system in pancreatic cancer cells, stable inducible CapG expressors were established. Overexpressed CapG was localised to the cytoplasm and the nuclear membrane, but was not observed in the nucleus. High CapG levels were associated with enhanced motility, but not with changes to the cell cycle, or cellular proliferation. In CapG-overexpressing cells, the levels and phosphorylation status of other actin-moduating proteins (Cofilin and ezrin/Radixin) were not altered. However, preliminary analyses suggest that the levels of other cellular proteins, such as ornithine aminotransferase and enolase, are altered upon CapG induction. CONCLUSION: We have generated pancreatic-cancer derived cell lines in which gene expression is fully controllable.

The history of these two biologically active components, controls on their concentrations, and implications for the complexity of the biosphere and habitability of earth have been hotly debated, but are generally considered independently.

Durability Properties of Materials and Mixture Composition of Bitumen Stabilized Materials

Bitumen Stabilized Materials (BSMs) are widely recognized as environmentally friendly through significantly reduced energy consumption,reduced emissions and reuse of high percentage of RAP and recycled pavement materials. In order for these materials to be cost effective,their life-cycle performance needs to be proven. Durability of BSMs is a long-term phenomena that requires consideration during mix design. Time is very important variable for performance consideration under field conditions. The factors that influence durability and long-term performance of BSMs include the mineral aggregates,binder and mixture compositions. In this study,the important,but less understood aspects of durability properties of BSMs were investigated. These include wear and tear of mineral aggregates,age-hardening of binder (foamed bitumen and bitumen emulsion) and moisture susceptibility of typical mix composition. The wear and tear was investigated using Durability Mill Index (DMI) test,binder ageing was investigated for short and long-term effect,and moisture susceptibility was investigated by pulsing water pressures using new device i.e. moisture induction simulation test (MIST). The accelerated moisture conditioning simulates field conditions whereafter retained shear properties are evaluated. The results found that durability of mineral aggregates is significantly influences by the type and nature of mineral composition. The ageing of binder occurred in both short and long-term conditions,with short-term effects being dominating the foamed bitumen properties during laboratory production. Moisture susceptibility is significantly influenced by the mix compositions of aggregates blends with and without RAP,with different bitumen binder types (foamed bitumen or bitumen emulsion) and active filler type (cement or lime) and contents. The relative importance of economics and environment in debate may soon be reversed due to the influence of three fac- tors. Firstly, in the global economy it is hard to hide the unwanted products of economic processes. Secondly, huge advances in sci- ence will reduce the imperfect knowledge of markets, making some monitoring and analyzing tools show the design of sensible and equitable livelihood in communities, which is more important than the motivation of maximising profits for some individuals or firms. Thirdly, China, as the last major player on the planet to take on economic growth, comes from traditions fundamentally differ- ent from those economies that have experienced the Industrial Revolution previously. Its challenges with sustainability and en- vironmental conservation predate Western economics by millennia, and it is implementing policies domestically and starting to work on the world stage that acknowledges that the surroundings are the host for any economic and socio-political system.

Factors Affecting Internet Banking Adoption: An Application of Adaptive LASSO

This research investigates a broad range of possible factors affecting the adoption of new technology in the banking industry using adaptive LASSO and a standard logit *** research integrated the adoption of the innovation framework and the technology acceptance theory to develop a conceptual framework for the *** data was collected from 400 bank customers in North *** perception and other customerspecific factors such as perceived risk index and negative attitude toward new technologies index were formulated for the proposed conceptual *** findings indicated that individuals with a negative attitude toward new technology are least likely to adopt internet *** addition,the logit model suggested that age,education level,and general(innate)innovativeness significantly impact the adoption of internet ***,gender,income,occupation,perceived risk,familiarity with the internet,and social inclusion have no significant impact on internet banking adoption in North Cyprus.

The larval stages of the cestode parasites belonging to the genus echinococcus grow within internal organs of humans and a range of animal *** resulting diseases,collectively termed echinococcoses,include major neglected tropical diseases of humans and *** larvae are outwardly protected by the laminated layer(LL),an acellular structure that is unique to this *** LL is based on a fibrillar meshwork made up of mucins,which are decorated by galactose-rich *** addition,in the species cluster termed *** sensu lato,the LL features nano-deposits of the calcium salt of myo-inositol hexakisphosphate(Insp_(6)).The main purpose of our article is to update the immunobiology of the *** recent advances in this area are(i)the demonstration of LL“debris”at the infection site and draining lymph nodes,(ii)the characterization of the decoy activity of calcium Inp6 with respect to complement,(iii)the evidence that the LL mucin carbohydrates interact specifically with a lectin receptor expressed in Kupffer cells(Clec4F),and(iv)the characterization of what appear to be receptor-independent effects of LL particles on dendritic cells and *** information is missing on the immunology of this intriguing structure:we discuss gaps in knowledge and propose possible avenues for research.

Steroidal glycoalkaloids(SGAs)are specialized metabolites produced by hundreds of Solanum species,including important vegetable crops such as tomato,potato,and *** it has been known that SGAs play important roles in defense in plants and“anti-nutritional"effects(e.g.,toxicity and bitterness)to humans,many of these molecules have documented anti-cancer,anti-microbial,antiinflammatory,anti-viral,and anti-pyretic *** these,α-solasonine andα-solamargine isolated from black nightshade(Solanum nigrum)are reported to have potent anti-tumor,anti-proliferative,and anti-inflammatory ***,α-solasonine andα-solamargine,along with the core steroidal aglycone solasodine,are the most widespread SGAs produced among the Solanum ***,it is still unknown how plants synthesize these bioactive steroidal *** comparative metabolomictranscriptome-guided approach,biosynthetic logic,combinatorial expression in Nicotiana benthamiana,and functional recombinant enzyme assays,here we report the discovery of 12 enzymes from *** that converts the starting cholesterol precursorto solasodine aglycone,and the downstreamα-solasonine,α-solamargine,and malonyl-solamargine SGA *** further identified six enzymes from cultivated eggplant that catalyze the production ofα-solasonine,α-solamargine,and malonyl-solamargine SGAs from solasodine aglycone via glycosylation and atypical malonylation *** work provides the gene tool box and platform for engineering the production of high-value,steroidal bioactive molecules inheterologoushosts usingsyntheticbiology.

Using a 3-D multicellular simulation of spinal cord injury with live cell imaging to study the neural immune barrier to nanoparticle uptake

Development of nanoparticle （NP） based therapies to promote regeneration in sites of central nervous system （CNS; i.e, brain and spinal cord） pathology relies critically on the availability of experimental models that offer biologically valid predictions of NP fate in vivo. However, there is a major lack of biological models that mimic the pathological complexity of target neural sites in vivo, particularly the responses of resident neural immune cells to NPs. Here, we have utilised a previously developed in vitro model of traumatic spinal cord injury （based on 3-D organotypic slice arrays） with dynamic time lapse imaging to reveal in real-time the acute cellular fate of NPs within injury foci. We demonstrate the utility of our model in revealing the well documented phenomenon of avid NP sequestration by the intrinsic immune cells of the CNS （the microglia）. Such immune sequestration is a known translational barrier to the use of NP-based therapeutics for neurological injury. Accordingly, we suggest that the utility of our model in mimicking microglial sequestration behaviours offers a valuable investigative tool to evaluate strategies to overcome this cellular response within a simple and biologically relevant experimental system, whilst reducing the use of live animal neurological injury models for such studies.

The wild population of the African lion Panthera leo continues to decline, requiring alternate conservation programs to be considered. One such program is ex situ reintroduction. Prior to release, longterm monitoring and assessment of behavior is required to determine whether prides and coalitions behave naturally and are sufficiently adapted to a wild environment. Social network analysis （SNA） can be used to provide insight into how the pride as a whole and individuals within it, function. Our study was conducted upon 2 captive-origin prides who are part of an ex situ reintroduction program, and 1 wild pride of African lion. Social interactions were collected at all occurrence for each pride and categorized into greet, social grooming, play, and aggression. Betweenness centrality showed that offspring in each pride were central to the play network, whereas degree indicated that adults received （indegree） the greatest number of overall social interactions, and the adult males of each pride were least likely to initiate （outdegree） any interactions. Through the assessment of individual centrality and degree values, a social keystone adult female was identified for each pride. Social network results indicated that the 2 captive-origin prides had formed cohesive social units and possessed relationships and behaviors comparable with the wild pride for the studied behaviors. This study provided the first SNA comparison between captive-bred origin and a wild pride of lions, providing valuable information on individual and pride sociality, critical for determining the success of prides within an ex situ reintroduction program.
