Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT03950154)assessed the efficacy and adverse events(AEs)of the combination of PD-1 blockade-activated DC-CIK(PD1-T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 202 participants were enrolled and randomly assigned in a 1:1 ratio to receive either first-line XELOX plus bevacizumab(the control group,n=102)or the same regimen plus autologous PD1-T cell immunotherapy(the immunotherapy group,n=100)every 21 days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 19.5 *** PFS was 14.8 months(95%CI,11.6-18.0)for the immunotherapy group compared with 9.9 months(8.0-11.8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 25.6 months(95%CI,18.3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 20.0%of patients in the immunotherapy group and 23.5%in the control groups,with no toxicity-associated deaths *** addition of PD1-T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

Isothermal microcalorimetry provides thermodynamic and kinetic information on various reactions and processes and is thereby a powerful tool to elucidate their mechanisms. Certain improvement in isothermal microcalorimetry with regard to the studies on soil and environmental sciences is briefly described. This review mainly focuses on the use of microcalorimetry in the determination of soil microbial activity, monitoring the toxicity and biodegradation of soil organic pollutants, the risk evaluation of metals and metalloids, the heat effect of ion exchange and adsorption in soil, and environmental researches. Promising prospects for the applications of the technique in the field are also discussed.

The Nesterov accelerated dynamical approach serves as an essential tool for addressing convex optimization problems with accelerated convergence *** previous studies in this field have primarily concentrated on unconstrained smooth con-vex optimization *** this paper,on the basis of primal-dual dynamical approach,Nesterov accelerated dynamical approach,projection operator and directional gradient,we present two accelerated primal-dual projection neurodynamic approaches with time scaling to address convex optimization problems with smooth and nonsmooth objective functions subject to linear and set constraints,which consist of a second-order ODE(ordinary differential equation)or differential conclusion system for the primal variables and a first-order ODE for the dual *** satisfying specific conditions for time scaling,we demonstrate that the proposed approaches have a faster conver-gence *** only requires assuming convexity of the objective *** validate the effectiveness of our proposed two accel-erated primal-dual projection neurodynamic approaches through numerical experiments.

Effect of Tanshinone IIA on High Glucose-induced Apoptosis and Oxidative Stress Damage in Vascular Endothelial Cells

Objective: The primary cause of microvascular disease in diabetic complications is long-term hyperglycemia, wherein the damage and apoptosis of vascular endothelial cells play a significant role. Sodium tanshinone IIA sulfonate(STS) has been found to have beneficial effects on cardiovascular health. This study aimed to investigate the impact of STS on high glucose-induced apoptosis and oxidative stress damage in vascular endothelial cells, as well as its potential protective mechanisms. Methods:Human umbilical vein endothelial cells(HUVECs) were divided into five groups:low-glucose group, high-glucose group, and three STS groups(STS-a, STS-b, and STS-c). The low-glucose group was incubated with DMEM low-sugar medium containing 5.5 mmol·L-1glucose, while the high-glucose group was treated with 33.3mmol·L-1glucose. The STS groups were exposed to 10, 30, and 50 μg·m L-1of STS,respectively. Each group was cultured for 72 h, and the MTT method was utilized to assess cell proliferation. Additionally, flow cytometry was employed to monitor changes in cell apoptosis and cellular oxidative stress indicators at 24, 48, and 72 h of cell culture in each group. Results: As time went on, the cell proliferation ability and apoptosis rate of each group gradually increased. The high-glucose group exhibited lower proliferation ability compared to the other groups. The STS-c group demonstrated the highest OD value for proliferation ability(24 h: 1.19±0.12; 48 h: 1.20±0.13; 72 h:1.25 ±0.12), but it was still lower than that of the low-sugar group. Notably, the high-glucose group had the highest cell apoptosis rate, while the low-glucose group had the lowest. The apoptosis rate of the STS-c group(24 h: 8.02±0.13; 48 h: 10.10±0.12;72 h: 13.18 ±0.11)% was between that of the low-glucose group and the high-glucose group, and lower than the STS-a and STS-b groups. Weld seam detection is an important part of automated *** present,few studies have been conducted on annular weld seams,and a lot of defects exist in the point cloud model of the tube sheet obtained by RGB-D cameras and photography *** at the above problems,this paper proposed an annular weld seam detection network named WeldNet where a voxel feature encoding layer was adaptively improved for annular weld seams,the sparse convolutional network and region proposal network(RPN)were used to detect annular weld seam position,and an annular weld seam detection loss function was ***,an annular weld seam dataset was established to train the *** with the random sampling consistency(RANSAC)method,WeldNet has a higher detection accuracy,as well as a higher detection success rate which has increased by 23%.Compared with U-Net,WeldNet has been proven to achieve a better detection result,and the intersection over the union of the weld seam detection is improved by 17.8%.
