Assessment of bone marrow changes in postmenopausal women with varying bone densities： magnetic resonance spectroscopy and diffusion magnetic resonance imaging

Background Recent studies suggest that bone marrow adipose tissue might play a role in the pathogenesis of osteoporosis. There are inconsistent findings on the relationship among marrow fat content, bone mineral density and apparent diffusion coefficient （ADC）. This study aimed to prospectively explore the efficacy of MR spectroscopy （MRS） and diffusion-weighted MR imaging （DWl） in detecting vertebral marrow changes in postmenopausal women with varying bone densities. Methods Both MRS and DWl of the lumber spine were performed in 102 postmenopausal women （mean age, （67.3±6.5） years; range, 55-83 years）, who underwent dual X-ray absorptiometry. Marrow fat content and ADC were compared and correlated among three groups： 24 with normal bone density, 31 with osteopenia and 47 with osteoporosis. Results Vertebral marrow fat content was significantly increased in the osteoporotic group （（65.60±7.68）%, P 〈0.001） and the osteopenic group （（57.68±6.45）%, P 〈0.001）, when compared with the normal bone density group （（51.67±3.27）%）. ADC values were significantly decreased in the osteoporotic group （（0.39±0.03）×10^-3mm^2/s, P 〈0.001） and in the osteopenic group （（0.42±0.02）×10^-3mm^2/s, P 〈0.001）, when compared with the normal bone density group （（0.47±0.03）×10^-3mm^2/s）. The marrow fat content negatively correlated with both bone density （r=-0.731, P 〈0.001） and marrow ADC （r=-0.572, P 〈0.001）. The bone density positively correlated with the ADC values （r=0.802, P 〈0.001）. Conclusions Postmenopausal women experience a corresponding increase in vertebral marrow fat content as the bone density decreases. Marrow fat content and ADC correlate to the bone density. MRS and DWl may indirectly assess the early bone marrow changes in postmenopausal women.

Primary porcine tracheal epithelial cells(PTECs) are an appropriate model for studying the molecular mechanism of various porcine respiratory diseases, including swine-origin mycoplasmas, which are isolated from respiratory tract of pigs and mainly found on the mucosal surface surrounding swine trachea. However, the short proliferation ability of primary PTECs greatly limits their lifespan. In this study, primary PTECs were carefully isolated and cultured, and immortal PTECs were constructed by transfecting primary PTECs with the recombinant constructed plasmid pEGFPhTERT containing human telomerase reverse transcriptase(hTERT). Immortal PTECs(hTERT-PTECs) maintained both the morphological and functional characteristics of primary PTECs, as indicated by the expression of cytokeratin 18, cellcycle analysis, proliferation assay, Western blotting, telomerase activity assay, karyotype analysis and quantitative RTPCR. Compared to primary PTECs, hTERT-PTECs had an extended replicative lifespan, higher telomerase activity, and enhanced proliferative activity. In addition, this cell line resulted in a lack of transformed and grown tumors in nude mice, suggesting that it could be safely applied in further studies. Moreover, hTERT-PTECs were vulnerable to all swineorigin mycoplasmas through quantitative analysis as indicated by 50% color changing unit(CCU_(50)) calculation, and no significant differences of adhesion ability between primary and immortal PTECs were observed. For the representative swine mycoplasma Mycoplasma hyopneumoniae(Mhp), except for DNA copies quantitative real-time PCR assay, indirect immunofluorescence assay and Western blotting analysis also depicted that hTERT-PTECs was able to adhere to different Mhp strains of different virulence. In summary, like primary PTECs, hTERT-PTECs could be widely used as an adhesion cell model for swine-origin mycoplasmas and in infection studies of various porcine respiratory pathogens.

Establishing a protein expression profile database for the normal human pituitary gland using two-dimensional high-performance liquid chromatography combined with LTQ-Orbitrap mass spectrometry

In this study, we selected adult normal pituitary gland tissues from six patients during operations for pituitary microadenomas via the transsphenoidal approach for extended normal pituitary tissue resection around the tumor, and analyzed the protein expression of human normal pituitary using two-dimensional high-performance liquid chromatography combined with LTQ-Orbitrap mass spectrometry proteomics technology. The ten most highly expressed proteins in normal human pituitary were： alpha 3 type VI collagen isoform 5 precursor （abundance among tall pituitary proteins 1.30%）, fibrinogen beta chain preproprotein （0.99%）, vimentin （0.73%）, prolactin （0.69%）, ATP synthase, H~ transporting and mitochondrial F1 complex beta subunit precursor （0.52%）, keratin I （0.49%）, growth hormone （0.45%）, carbonic anhydrase I （0.40%）, heat shock protein 90 kDa I （0.31%）, and annexin V （0.30%）. Based on the biological function classifications of these proteins, the top three categories by content were neuroendocrine proteins （abundance among all pituitary proteins, 40.1%）, catalytic and metabolic proteins （28.3%）, and cell signal transduction proteins （9.8%）. Based on cell positioning classification, the top three categories were cell organelle （24.5%） membrane （20.8%）, and cytoplasm （13.0%）. Based on biological process classification, the top three categories of proteins are involved in physiological processes （42.9%）, cellular processes （40.4%）, and regulation of biological processes （9.1%）. Our experimental findings indicate that a protein expression profile database of normal human pituitary can be precisely and efficiently established by proteomics technology.

Background Vascular access （VA） dysfunction is a major clinical complication in the hemodialysis population and has a direct effect on dialysis outcome. This study was conducted to explore the role of microinflammation in the VA dysfunction in maintenance hemodialysis patients. Methods Forty-seven patients （male 35 and female 12） receiving maintenance hemodialysis were included for this study. They were divided into three groups： group 1 （n=15）, patients with initial hemodialysis and new arteriovenous fistula （AVF）; group 2 （n=18）, patients treated with hemodialysis for long term with well-functional VA; group 3 （n=14）, maintenance hemodialysis patients with VA dysfunction. Biochemical parameters and serum tumor necrosis factor-α （TNF-α）, interleukin 6 （IL-6） and monocyte chemoattractant protein-1 （MCP-1） were determined. High-sensitivity C-reactive protein （hs-CRP） was determined by latex-enhanced immuno-nephelometric method. Tissues of radial artery were taken from group 1 and group 3 for the histological study. Expression of CD68 and MCP-1 in the radial artery was determined by immunohistochemistry. Results Serum hs-CRP in group 3 was significantly higher than those in group 1 and group 2 （（7.40±2.42） mg/L vs （4.21±1.62） mg/L and （5.04±3.65） mg/L, P 〈0.01 and P 〈0.05, respectively）. Serum TNF-α in group 3 was significantly higher than those in group 1 and group 2 （（64.03±9.29） pg/ml vs （54.69±12.39） pg/ml and （54.05±7.68） pg/ml, P 〈0.05 and P 〈0.01, respectively）. Serum IL-6 in group 3 was also significantly higher than those in group 1 and group 2 （（70.09±14.53） pg/ml vs （56.43±10.11） pg/ml and （60.77±9.70） pg/ml, P 〈0.01 and P 〈0.05, respectively）. Patients in group 3 had a thicker internal layer of vessels than in group 1 （（0.356±0.056） mm vs （0.111±0.021） mm, P 〈0.01）. Automated Counting of Rice Planthoppers in Paddy Fields Based on Image Processing

A quantitative survey of rice planthoppers in paddy fields is important to assess the population density and make forecasting decisions. Manual rice planthopper survey methods in paddy fields are time-consuming, fatiguing and tedious. This paper describes a handheld device for easily capturing planthopper images on rice stems and an automatic method for counting rice planthoppers based on image processing. The handheld device consists of a digital camera with WiFi, a smartphone and an extrendable pole. The surveyor can use the smartphone to control the camera, which is fixed on the front of the pole by WiFi, and to photograph planthoppers on rice stems. For the counting of planthoppers on rice stems, we adopt three layers of detection that involve the following：（a） the first layer of detection is an AdaBoost classifier based on Haar features;（b） the second layer of detection is a support vector machine（SVM） classifier based on histogram of oriented gradient（HOG） features;（c） the third layer of detection is the threshold judgment of the three features. We use this method to detect and count whiteback planthoppers（Sogatella furcifera） on rice plant images and achieve an 85.2% detection rate and a 9.6% false detection rate. The method is easy, rapid and accurate for the assessment of the population density of rice planthoppers in paddy fields.
