Conditionally Reprogrammed Human Normal Airway Epithelial Cells at ALI: A Physiological Model for Emerging Viruses

Cancer cell lines have been used widely in cancer biology, and as biological or functional cell systems in many biomedical research fields. These cells are usually defective for many normal activities or functions due to significant genetic and epigenetic changes. Normal primary cell yields and viability from any original tissue specimens are usually relatively low or highly variable. These normal cells cease after a few passages or population doublings due to very limited proliferative capacity. Animal models(ferret, mouse, etc.) are often used to study virus-host interaction. However, viruses usually need to be adapted to the animals by several passages due to tropism restrictions including viral receptors and intracellular restrictions. Here we summarize applications of conditionally reprogrammed cells(CRCs), long-term cultures of normal airway epithelial cells from human nose to lung generated by conditional cell reprogramming(CR) technology, as an ex vivo model in studies of emerging viruses. CR allows to robustly propagate cells from non-invasive or minimally invasive specimens, for example, nasal or endobronchial brushing. This process is rapid(2 days) and conditional. The CRCs maintain their differentiation potential and lineage functions, and have been used for studies of adenovirus, rhinovirus, respiratory syncytial virus, influenza viruses, parvovirus, and SARS-CoV. The CRCs can be easily used for airliquid interface(ALI) polarized 3 D cultures, and these coupled CRC/ALI cultures mimic physiological conditions and are suitable for studies of viral entry including receptor binding and internalization, innate immune responses, viral replications, and drug discovery as an ex vivo model for emerging viruses.

Synthesis of Di-substituted Proazaphosphatrane and Its Application in Carbon-carbon Double-bond Isomerization

Diisopropyltren was synthesized from the reaction of tren with benzaldehyde in toluene, followed by reducing the imine intermediate with NaBH4, and debenzylation catalyzed by 10% Pd/C in methanol. It was then converted to its corresponding azaphosphatrane via ring-closing reaction of HMPT and triflic acid. Deprotonation of azaphosphatrane with potassium t-butoxide afforded the target diisopropyl proazaphosphatrane ld. A comparative study of compound ld for isomerization of allylic system and methylene-interrupted diene system revealed that compound ld is more efficient than its tri-substituted analoque（lb）. In acetonitrile at 40 ℃, allylaromatics were selectively isomerized to 1-arylpropene generally in an isolated yield more than 95% with trans, cis molar ratios in a range of 87/13 to 96/4. Allyl phenylsulfide was converted to 1-phenylthiopropene（molar ratio of Z/E=54/46） in a yield of 93%-95%. Methylene-interrupted dienes were also isomerized in high yield.

Effect of acupuncture at Foot-Yangming Meridian on gastric mucosal blood flow, gastric motiiity and brain-gut peptide

AIM： To observe the effect of acupuncture at Foot- Yangming Meridian on gastric mucosal blood flow （GMBF）, gastric motility and brain-gut peptide. METHODS： Sixty SD rats were randomly divided into 6 groups： normal control group, model group （group with gastric mucosal damage, GMD）, Sibai group （with acupuncture at Sibai point ＋ GMD）, Tianshu group （with acupuncture at Tianshu point ＋ GMD）, Zusanli group （with acupuncture at Zusanli point ＋ GMD） and non-acupoint group （with acupuncture at non-acupoint ＋ GMD）. The GMD model group was induced by infusing pure alcohol into gastric cavity. H2 Gas Clearance Test （HGCT） was used to measure GMBF, the frequency and amplitude of gastric motility were measured by the method of aerocyst, the content of brain-gut peptide in sinus ventriculi and bulbus medullae were detected by radioimmunoassay. RESULTS： Inhibitory effect of the frequency and amplitude of gastric motility were shown in model group, and the rates of frequency and amplitude changes were remarkably different from the normal control group （-19.41 ± 17.21 vs-4.71 ± 10.32, P 〈 0.05; -51.61 ± 29.02 vs 1.81 ± 14.12, P 〈 0.01）. In comparison with control group, the GMBF was 0.52 ± 0.161 mL vs 1.03 ± 0.255 mL per 100g tissue/min, P 〈 0.01, the content of motilin in sinus ventriculi and bulbus medullae was 63.04 ± 7.77 pg/mL vs 72.91 ± 8.42 pg/mL, P 〈 0.05 and 50.96 ± 8.77 pg/mL vs 60.76 ± 8.05 pg/mL, P 〈 0.05, but the content of somatostatin in sinus ventriculi and bulbus medullae was 179.85 ± 43.13 ng/g vs 90.54 ± 40.42 ng/g, P 〈 0.01 and 532.86 ± 122.58 ng/g vs 370.91 ± 76.29 ng/g, P 〈 0.05,respectively. In comparison with model group, the amplitude of gastric motility was 1.52 ± 20.13, -6.52 ± 23.31, 6.92 ± 25.21 vs -51.61 ± 29.02, P 〈 0.01 and GMBF was 0.694 ± 0.160 mL vs 0.893 ± 0.210 mL, 1.038 ± 0.301 mL vs 0.52 ± 0.161 mL per 100g tissue/rain, P 〈 0.01, respectively in Tianshu, Sibai and Zusanli groups, the content of motilin in sinus ventriculi and bulbus medul

Corneal blindness caused by limbal stem cell deficiency(LSCD) is one of the most common debilitating eye disorders. Thus far, the most effective treatment for LSCD is corneal transplantation, which is often hindered by the shortage of donors. Pluripotent stem cell technology including embryonic stem cells(ESCs) and induced pluripotent stem cells(iPSCs) have opened new avenues for treating this disease. iPSCs-derived corneal epithelial cells provide an autologous and unlimited source of cells for the treatment of LSCD. On the other hand, iPSCs of LSCD patients can be used for iPSCs-corneal disease model and new drug discovery. However, prior to clinical trial, the efficacy and safety of these cells in patients with LSCD should be proved. Here we focused on the current status of iPSCs-derived corneal epithelial cells used for cell therapy as well as for corneal disease modeling. The challenges and potential of iPSCs-derived corneal epithelial cells as a choice for clinical treatment in corneal disease were also discussed.

BACKGROUND In recent years,many studies have shown that proteasome 26S subunit non-ATPase 6(PSMD6)plays an important role in the occurrence and development of malignant ***,there are no reports on the evaluation of the potential role of PSMD6 in hepatocellular carcinoma(HCC).AIM To comprehensively evaluate the overexpression pattern and clinical significance of PSMD6 in HCC *** This study integrated PSMD6 mRNA expression profiles from 4672 HCC and 3667 non-HCC tissues,along with immunohistochemical scores from 383 HCC and adjacent tissues,to assess PSMD6 overexpression in *** regularly interspaced short palindromic repeats knockout technology evaluated PSMD6’s essential role in HCC cell *** enrichment analysis explored the molecular mechanism of PSMD6 abnormalities in *** sensitivity analysis and molecular docking analysed the effect of abnormal expression of PSMD6 on the drug sensitivity of HCC *** The results of 41 external and two internal datasets showed that PSMD6 mRNA(SMD=0.26,95%CI:0.09-0.42,P<0.05)and protein(SMD=2.85,95%CI:1.19-4.50,P<0.05)were significantly overexpressed in HCC *** integrated analysis results showed that PSMD6 had a significant overexpression pattern in HCC tissues(SMD=0.40,95%CI:0.15-0.66,P<0.05).PSMD6 knockout inhibited HCC cell growth(chronos scores<-1).Functional enrichment implicated ribosome biogenesis and RNA *** enrichment of signalling pathways such as RNA degradation,ribosomes,and chemical carcinogenesis—reactive oxygen *** sensitivity analysis and a molecular docking model showed that high expression of PSMD6 was associated with the tolerance of HCC cells to drugs such as ML323,sepantronium bromide,and *** PSMD6 effectively distinguished HCC tissues(AUC=0.75,95%CI:0.71-0.79).CONCLUSION This study was the first to discover that PSMD6 was overexpressed in HCC ***6 is essential for the growth of HCC cell

POLYMER NETWORKS BY MOLECULAR DYNAMICS SIMULATION: FORMATION, THERMAL, STRUCTURAL AND MECHANICAL PROPERTIES

A molecular dynamics simulation method is presented and used in the study of the formation of polymer networks. We study the formation of networks representing the methylene repeating units as united atoms. The network formation is accomplished by cross-linking polymer chains with dedicated functional end groups. The simulations reveal that during the cross-linking process, initially branched molecules are formed before the gel point; approaching the gel point, larger branched entities are formed through integration of smaller branched molecules, and at the gel point a network spanning the simulation box is obtained; beyond the gel point the network continues to grow through the addition of the remaining molecules of the sol phase onto the gel （the network）; the final completion of the reaction occurs by intra-network connection of dangling ends onto unsaturated cross-linkers. The conformational properties of the strands in the undeformed network are found to be very similar with the conformational properties of the chains before cross-linking. The uniaxial deformation of the formed networks is investigated and the modulus determined from the stress-strain curves shows reciprocal scaling with the precursor chain length for networks formed from sufficiently large precursor chains （N≥ 20）.

Improved positioning algorithm based on two step least square in NLOS environments

Non-line-of-sight （NLOS） propagation is a major source of error for accurate time-of-arrival （TOA） location estimation. The two-step least square （LS） method, which does not need any information about the distribution of the NLOS error, has been studied in related researches to provide efficient location estimation of the mobile terminal （MT）. By observing the estimation of the error covariance matrix in the two-step LS method, this paper finds that the measured distances are more suitable than the initial estimated distances by maximum likelihood （ML） algorithm in NLOS environments. Moreover, by making use of the geometry relations among fixed terminals （FTs）, some measured distances which are corrupted by big NLOS errors may be reduced. Further, an iteration method can be adopted to further improve the performance of the algorithm. Accordingly, an improved algorithm based on the two-step LS method is proposed. Simulation results demonstrate that the improved algorithm has better performance than the two-step LS method and converges more quickly. Meanwhile, it is robust in different NLOS environments.

Insights into the Organotemplate-Free Synthesis of Zeolite Catalysts

As the most important nanoporous material, zeolites, which have intricate micropores, are essential heterogeneous catalysts in industrial processes. Zeolites are generally synthesized with organic templates under hydrothermal conditions; however, this method is environmentally unfriendly and costly due to the formation of harmful gases and polluted water. This article briefly summarizes the role of organic templates and describes designed routes for the organotemplate-free synthesis of zeolites, aided by zeolite seeds and zeolite seeds solution. Furthermore, this review explicates that the micmpore volume decreases with an increase of the Si/Al ratios in the organotemplate-free synthesis of zeolite products, where Na^＋ exists as an alkali cation. This feature is very important in directing the synthesis of zeolite catalysts with controllable Si/AI ratios under organotemplate-free conditions, and is thus important for the efficient design of zeolites.

Evolution of nuclear charge radii in copper and indium isotopes

Systematic trends in nuclear charge radii are of great interest due to universal shell effects and odd-even staggering(OES).The modified root mean square(rms)charge radius formula,which phenomenologically accounts for the formation of neutron-proton(np)correlations,is here applied for the first time to the study of odd-Z copper and indium *** results obtained by the relativistic mean field(RMF)model with NL3,PK1 and NL3^(*)parameter sets are compared with experimental *** results show that both OES and the abrupt changes across N=50 and 82 shell closures are clearly reproduced in nuclear charge *** inverted parabolic-like behaviors of rms charge radii can also be described remarkably well between two neutron magic numbers,namely N=28 to 50 for copper isotopes and N=50 to 82 for indium *** implies that the np-correlations play an indispensable role in quantitatively determining the fine structures of nuclear charge radii along odd-Z isotopic ***,our conclusions have almost no dependence on the effective forces.
