Amyloid-b,tau pathology,and biomarkers of neurodegeneration make up the core diagnostic biomarkers of Alzheimer disease(AD).However,these proteins represent only a fraction of the complex biological processes underlying AD,and individuals with other brain diseases in which AD pathology is a comorbidity also test positive for these diagnostic *** ADspecific early diagnostic and disease staging biomarkers are *** this study,we performed tandem mass tag proteomic analysis of paired cerebrospinal fluid(CSF)and serum samples in a discovery cohort comprising 98 *** biomarkers were validated by parallel reaction monitoring–based targeted proteomic assays in an independent multicenter cohort comprising 288 *** quantified 3,238 CSF and 1,702 serum proteins in the discovery cohort,identifying 171 and 860 CSF proteins and 37 and 323 serum proteins as potential early diagnostic and staging biomarkers,*** the validation cohort,58 and 21 CSF proteins,as well as 12 and 18 serum proteins,were verified as early diagnostic and staging biomarkers,*** 19-protein CSF and an 8-protein serum biomarker panels were built by machine learning to accurately classify mild cognitive impairment(MCI)due to AD from normal cognition with areas under the curve of 0.984 and 0.881,*** 19-protein CSF biomarker panel also effectively discriminated patients with MCI due to AD from patients with other neurodegenerative ***,we identified 21 CSF and 18 serum stage-associated proteins re-flecting AD *** findings provide a foundation for developing bloodbased tests for AD screening and staging in clinical practice.

Association between body mass index and in vitro fertilization/intra-cytoplasmic sperm injection outcomes:An analysis of 15,124 normal ovarian responders in China

Effect of Tanshinone IIA on High Glucose-induced Apoptosis and Oxidative Stress Damage in Vascular Endothelial Cells

Objective: The primary cause of microvascular disease in diabetic complications is long-term hyperglycemia, wherein the damage and apoptosis of vascular endothelial cells play a significant role. Sodium tanshinone IIA sulfonate(STS) has been found to have beneficial effects on cardiovascular health. This study aimed to investigate the impact of STS on high glucose-induced apoptosis and oxidative stress damage in vascular endothelial cells, as well as its potential protective mechanisms. Methods:Human umbilical vein endothelial cells(HUVECs) were divided into five groups:low-glucose group, high-glucose group, and three STS groups(STS-a, STS-b, and STS-c). The low-glucose group was incubated with DMEM low-sugar medium containing 5.5 mmol·L-1glucose, while the high-glucose group was treated with 33.3mmol·L-1glucose. The STS groups were exposed to 10, 30, and 50 μg·m L-1of STS,respectively. Each group was cultured for 72 h, and the MTT method was utilized to assess cell proliferation. Additionally, flow cytometry was employed to monitor changes in cell apoptosis and cellular oxidative stress indicators at 24, 48, and 72 h of cell culture in each group. Results: As time went on, the cell proliferation ability and apoptosis rate of each group gradually increased. The high-glucose group exhibited lower proliferation ability compared to the other groups. The STS-c group demonstrated the highest OD value for proliferation ability(24 h: 1.19±0.12; 48 h: 1.20±0.13; 72 h:1.25 ±0.12), but it was still lower than that of the low-sugar group. Notably, the high-glucose group had the highest cell apoptosis rate, while the low-glucose group had the lowest. The apoptosis rate of the STS-c group(24 h: 8.02±0.13; 48 h: 10.10±0.12;72 h: 13.18 ±0.11)% was between that of the low-glucose group and the high-glucose group, and lower than the STS-a and STS-b groups. Salinity effect-induced ENSO amplitude modulation in association with the interdecadal Pacific Oscillation

A 110-year ensemble simulation of an ocean general circulation model(OGCM)was analyzed to identify the modulation of salinity interdecadal variability on El Niño-Southern Oscillation(ENSO)amplitude in the tropical Pacific during *** simulating results show that sea surface salinity(SSS)variation in the region exhibits notable and coherent interdecadal variability signal,which is closely associated with the Interdecadal Pacific Oscillation(IPO).As salinity increases or reduces,the SSS modulations on ENSO amplitude during its warm/cold events vary asymmetrically with positive/negative IPO ***,salinity interdecadal variability can enhance or reduce ENSO-related conditions in upper-ocean stratification,contributing noticeably to ENSO *** anomalies associated with the mixed layer depth and barrier layer thickness can modulate ENSO amplitude during positive and negative IPO phases,resulting in the asymmetry of sea surface temperature(SST)anomaly in the tropical *** positive IPO phases,SSS interdecadal variability contributes positively to El Niño amplitude but negatively to La Niña amplitude by enhancing or reducing SSS interannual variability,and vice versa during negative IPO ***,the results indicate that the modulation of the ENSO amplitude by the SSS interdecadal variability is 15%-28%during negative IPO phases and 30%-20%during positive IPO phases,***,the SSS interdecadal variability associated with IPO and its modulation on ENSO amplitude in the tropical Pacific are among factors essentially contributing ENSO diversity.

The evolution of solar magnetic fields is significant for understanding and predicting solar *** our knowledge of solar magnetic fields largely depends on the photospheric magnetic *** this paper,based on the spherical harmonic expansion of the photospheric magnetic field observed by Wilcox Solar Observatory,we analyze the time series of spherical harmonic coefficients and predict Sunspot Number as well as synoptic maps for Solar Cycle *** find that solar maximum years have complex short-period disturbances,and the time series of coefficient g_(7)~0 is nearly in-phase with Sunspot Number,which may be related to solar meridional *** Long Short-Term Memory networks(LSTM),our prediction suggests that the maximum of Solar Cycle 25 is likely to occur in June 2024 with an error of 8 months,the peak sunspot number may be 166.9±22.6,and the next solar minimum may occur around January *** incorporating Empirical Mode Decomposition,we enhance our forecast of synoptic maps truncated to Order 5,validating their relative *** prediction not only addresses a gap in forecasting the global distribution of the solar magnetic field but also holds potential reference value for forthcoming solar observation plans.

We utilize molecular dynamics simulations to investigate the microstructures of ions and polyelectrolytes in aqueous solutions under external electric *** focusing on the multi-body interactions between ionic components and H_(2)O molecules,as well as their responses to the external electric fields,we clarify several nontrivial molecular features of the ionic and polyelectrolyte solutions,such as the solvations of cations and anions,clustering of the ions,and dispersions/aggregations of polyelectrolyte chains,as well as the corresponding responses of H_(2)O molecules in these *** simulations illustrate the variations in structures of ionic solutions caused by reversing the charge sign of the ions,and elucidate the disparity in structures between anionic and cationic polyelectrolyte solutions in the presence of the external electric *** work clarifies the mechanism for the alternations in complex multi-body interactions in aqueous solutions caused by the application electric field,which can contribute to the fundamental understanding of the physical and chemical natures of ion-containing and charged polymeric systems.

Recent advances in electrocatalytic generation of indole-derived radical cations and their applications in organic synthesis

Indole-derived radical cations, open-shell reactive species, display distinctive dual reactivity due to the carbon-centered radical and more electrophilic carbocation, which frequently appear in a variety of single electron oxidation reactions for synthesizing structurally diverse functionalized indoles and indolines. Electrocatalysis is considered as a synthetically attractive and environmentally friendly alternative for driving the single electron oxidation of indoles. Remarkable achievements in electrocatalytic indolederived radical cation-mediated indole functionalization have been realized so far. This review comprehensively summarizes the recent progresses in the applications of electrocatalytic indole radical cations,including C(sp~2)–H functionalization, dearomative 2,3-difunctionalization, and ring-opening reaction, emphasizing the vital single electron oxidation steps of indoles, the substrates scope and limitations, and the reaction mechanisms.
