Alzheimer’s disease (AD) is a degenerative disease of the central nervous system. It results in cognitive dysfunction due to the loss of functional neurons and a deficit of new neurons， which can lead to death in severe cases. Repairing damaged neurons by promoting hippocampal neurogenesis has become therapeutic modality to combat neurodegenerative diseases. The novel isoflavone alkaloid LY01， found in the edible fruits of Sophora alopecuroides L.， has various neuroprotective effects. However， the molecular mechanism by which it promotes the proliferation of endogenous neural stem cells (NSCs) is not yet precisely known. The effects of LY01 were investigated in vivo and in vitro. In vivo experiments showed that LY01 could counteract the toxic damage of Aβ， enhance the learning ability and memory capacity of 5×FAD mice， and improve the morphology of neurons in the hippocampal region. In vitro experiments showed that LY01 was effective against antioxidant damage， improved the cell morphology of C17.2 mouse NSCs after hydrogen peroxide injury， and increased cell viability. Both in vitro and in vivo experiments promoted NSC proliferation by upregulating the mRNA and protein levels of the critical genes of the Wnt/β-catenin pathway， which increased levels of doublecortin to facilitate new neuronal generation. This indicates that the ability of LY01 to counteract Aβ toxicity and alleviate oxidative damage in early AD is associated with activation of the Wnt/β-catenin signaling pathway to promote NSC proliferation and thus repair damaged neurons.

A Cytotoxic Saponin with Two Monoterpenoids from Albizia julibrissin

Objective to explore the effect of lactational exposure alone to TCDD on growih development and induction of lungs cytochrome P450 1A1(CYP1A1)in mice *** Post-parturition mice were treated immediately by TCDD intraperitonal injection(i.p.).TCDD treatment was divided into 2 groups with different doses of 40μg and 20μg TCDD/kg body ***;there were two vehicle controls with treatment of a mixed-liquor from ethybenzene and peanutoil(ratio=1:4),and one animal control without any *** TCDD maternal administration was completed by three times on post-parturition days1,3,and 5. Mice offspring were weaned on postnatal days(PND)21 and sacrificed on *** Body weight of mice offspring in both TCDD treatments were significantly lower than those in two vehicle controls and animal control from *** vagina-opening days of female offspring were shortened to 17.5 and 18.4 days respectively in two 40μg and 20μg TCDD/kg body weight treatments,compared to 29.1 and 31.2 days in two vehicle *** testes-dascending days of male offspring were delayed to 27.3 days in 40μg TCDD/kg body weight,compared to 24.0 days in 40μg vehicle *** days of eyes-opening and maxillary teeth-eruption in mice offspring were also shortened in 40μg TCDD/kg body *** immunohistochemical sections in two TCDD treatments showed that alwolar wall thickened,alveolar cavity expanded,and the expression of CYP1A1 was increased *** These results indicate that lactational TCDD exposure alone affected the development of mice offspring and damage lung tissues of mice offspring.

AIM:To determine whether mitochondrial dysfunction resulting from high-fat diet is related to impairment of the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt,also known as PKB) pathway. METHODS:Rat models of nonalcoholic fatty liver were established by high-fat diet feeding. The expression of total and phosphorylated P13K and Akt proteins in hepatocytes was determined by Western blotting. Degree of fat accumulation in liver was measured by hepatic triglyceride. Mitochondrial number and size were determined using quantitative morphometric analysis under transmission electron microscope. The permeability of the outer mitochondrial membrane was assessed by determining the potential gradient across this ***:After Wistar rats were fed with high-fat diet for 16 wk,their hepatocytes displayed an accumulation of fat (103.1 ± 12.6 vs 421.5 ± 19.7,P < 0.01),deformed mitochondria (9.0% ± 4.3% vs 83.0% ± 10.9%,P < 0.05),and a reduction in the mitochondrial membrane potential (389.385% ± 18.612% vs 249.121% ± 13.526%,P < 0.05). In addition,the expression of the phosphorylated P13K and Akt proteins in hepatocytes was reduced,as was the expression of the anti-apoptotic protein Bcl-2,while expression of the pro-apoptotic protein caspase-3 was increased. When animals were treated with pharmacological inhibitors of P13K or Akt,instead of high-fat diet,a similar pattern of hepatocellular fat accumulation,mitochondrial impairment,and change in the levels of PI3K,Akt,Bcl-2 was observed. CONCLUSION:High-fat diet appears to inhibit the PI3K/Akt signaling pathway,which may lead to hepa-tocellular injury through activation of the mitochondrial membrane pathway of apoptosis.

Introduction:Newer skin tests,including the ESAT6-CFP10(EC)skin test,were recommended for diagnosing Mycobacterium tuberculosis(***)***,no data exist assessing the diagnostic performance of the EC skin test among foreign students with different skin ***:A cohort study at Nanjing medical university screened incoming foreign *** EC skin test was used to assess for *** infection,and results were read at 24,48,72,and 96-hours ***:Among 96 participants,*** infection rates at 24,48,72,and 96-hours post-injection were 3.13%,7.29%,13.54%,and 9.38%,*** infection rates were lower among individuals with darker skin tones,the difference was not statistically significant(P=0.186),and variations were consistent across different measurement *** analysis revealed 5.3%in the continuousincreasing group,86.5%in the low-stable group,and 5.2%in the elevated-decreasing ***,participants in the elevated-decreasing group had lighter skin tones,with trajectory patterns consistent across different skin ***:The EC skin test is safe,and redness diameter is a more reliable indicator than *** should be collected within 48 to 72 hours,with verification at 72 hours crucial if initial results are ***,skin color does not affect EC skin test outcomes.
