Despite the promise of immunotherapy such as the immune checkpoint inhibitors(ICIs)anti-PD-1 and anti-CTLA-4 for advanced melanoma,only 26%–52%of patients respond,and many experience grade III/IV immune-related adverse *** by the need for an effective therapy for patients non-responsive to clinically approved ICIs,we have developed a novel nanoimmunotherapy that combines locally administered Prussian blue nanoparticle-based photothermal therapy(PBNP-PTT)with systemically administered agonistic anti-CD137 monoclonal antibody therapy(aCD137).PBNP-PTT was administered at various thermal doses to melanoma cells in vitro,and was combined with aCD137 in vivo to test treatment effects on melanoma tumor progression,animal survival,immunological protection against tumor rechallenge,and *** administered at a melanoma-specific thermal dose,PBNP-PTT elicits immunogenic cell death(ICD)in melanoma cells and upregulates markers associated with antigen presentation and immune cell co-stimulation in ***,PBNP-PTT eliminates primary melanoma tumors in vivo,yielding long-term tumor-free ***,the antitumor immune effects generated by PBNP-PTT cannot eliminate secondary tumors,despite significantly slowing their *** addition of aCD137 enables significant abscopal efficacy and improvement of survival,functioning through activated dendritic cells and tumor-infiltrating CD8^(+)T cells,and generates CD4^(+)and CD8^(+)T cell memory that manifests in the rejection of tumor rechallenge,with no long-term *** study describes for the first time a novel and effective nanoimmunotherapy combination of PBNP-PTT with aCD137 mAb therapy for melanoma.

Ca^2+ and SO4^2- interactions with dissolved organic matter: Implications of groundwater quality for CKDu incidence in Sri Lanka

It has recently been proposed that recalcitrant dissolved organic carbon(DOC) in groundwater plays a potent etiological role in the peculiar distribution of chronic kidney disease of unknown etiology(CKDu).This study aims to elucidate the interactions of Ca^2+and SO4^2-with a model organic fraction of humic acid(SHA) to determine the possible relationship of CKDu incidence with the DOC in drinking *** and FT-IR methods respectively determined the surface functional groups and chemical composition of protonated dissolved organic carbon(HDOC) in a CKDu high-risk zone(HR) of Sri Lanka and in *** surface C composition(87.9%) and lower O(11.4%) were observed for HDOC from the HR region than for SHA(C: 73.8%, O: 24.7%).Aromatic C with less Ocontaining functional groups was observed in *** IR band at 1170 cm^-1 confirms the formation of organic sulfonate(C–SO3^-) on SHA.A band at 1213 cm^-1 due to organic sulfonate in HDOC from the CKDu HR region was also *** IR band at 866 cm^-1 evidenced the formation of CaCO3 on SHA above pH *** data confirmed the presence of sulfur oxidation states corresponding to SO3^2-and SO4^2-at 168.9 eV and 170.1 eV binding energies,*** binding energies at 347 eV and 351 eV for Ca 2p3/2 and Ca 2p1/2 eV,respectively, confirmed the bidentate complexation of Ca^2+with COO-and sulfonate groups on *** organic sulfonate formed is postulated as a uremic toxicant.

Ureteric Stone Containing Gas: First Case Report with Review of Cases of Urolithiasis with Gas within the Stone and Its Clinical Implications

We report the first case of a ureteric stone containing gas. This rare stone with gas within it was found during the management of a diabetic patient with urosepsis as the initial presentation. Literature review of cases of renal stone containing gas, mechanisms of gas within the stone, and clinical implications of stone containing gas are discussed. Also, a new terminology is proposed to describe this phenomenon.

One-year survival in recipients older than 50 bridged to heart transplant with Impella 5.5 via axillary approach

BACKGROUND Optimizing patients with advanced heart failure before orthotopic heart transplantation(OHT), especially in patients greater than 50 years old, is imperative to achieving successful post-transplant outcomes. Complications are well-described for patients bridged to transplant(BTT) with durable left ventricular assist device(LVAD) support. Given the lack of data available in older recipients after the recent increase in mechanical support use, we felt it crucial to report our center’s one-year outcomes in older recipients after heart transplantation with percutaneously placed Impella 5.5 as a *** Forty-nine OHT patients were supported with the Impella 5.5 intended as a bridge between December 2019 and October 2022 at Mayo Clinic in Florida. Data were extracted from the electronic health record at baseline and during their transplant episode of care after Institutional Review Boards approval as exempt for retrospective data *** Thirty-eight patients aged 50 or older were supported with Impella 5.5 as BTT. Ten patients underwent heart and kidney transplantation within this cohort. The median age at OHT was 63(58–68) years, with 32 male(84%) and six female patients(16%). Etiology was divided into ischemic(63%) and non-ischemic cardiomyopathy(37%). The baseline median ejection fraction was 19%(15–24). Most patients were in blood group O(60%), and 50% were diabetic. The average duration of support was 27 days(range 6–94). The median duration of follow-up is 488 days(185–693). For patients that have reached the 1-year follow-up timeframe(22 of 38, 58%), the 1-year post-transplant survival is 95%.CONCLUSION Our single-center data provides awareness for using the Impella 5.5 percutaneously placed axillary support device in older heart failure patients in cardiogenic shock as a bridge to transplantation. One-year survival outcomes after heart transplantation are excellent despite the older recipient’s age and prolonged pre-transplant support.

Autoheated thermophilic aerobic sludge digestion and metal bioleaching in a two-stage reactor system

A two-stage process has been developed for stabilization of sludge and removal of heavy metals from the secondary activated sludge with high rate of energy and time conservation. The first stage of the process involves autoheated thermophilic aerobic digestion at 55-60℃ inoculated with less-acidophilic thermophilic sulfur-oxidizing microorganisms （ATAD）. The results show that it is possible to maintain the autoheated conditions （55-60℃） in the ATAD reactor up to 24 hr, leading to reduction of 21% total solids （TS）, 27% volatile solids （VS）, 27% suspended solids （SS） and 33% volatile suspended solids （VSS） from the sludge. The sludge pH also decreased from 7 to 4.6 due to the activity of less-acidophilic thermophilic microorganisms. In the second stage operation, the digested sludge （pH 4.6, TS 31.6 g/L） from stage one was subjected to bioleaching in a continuous stirred tank reactor, operated at mean hydraulic retention times （HRTs） of 12, 24 and 36 hr at 30℃. An HRT of 24 hr was found to be sufficient for removal of 70% Cu, 70% Mn, 75% Ni, and 80% Zn from the sludge. In all, 39% VSS, 76% Cu, 78.2% Mn, 79.5% Ni and 84.2% Zn were removed from the sludge in both the stages.

Acute infections,including those due to Coronaviridae and other viruses,often stimulate a febrile response.A mild fever appears to improve outcome;it appears to diminish viral replication by several mechanisms,including virion entry into host cells and genome transcription,and improving host defence mechanisms against the ***,a fever may also damage host cellular and tissue function and increase metabolic *** temperatures at the lower end of the febrile range,the benefit of the fever appears to outweigh the detrimental ***,at higher temperatures,the outcome worsens,suggesting that the disadvantages of fever on the host predominate.A non-infective fever is associated with a worse outcome at lower temperatures,suggesting that hyperthermia carries less benefit in the absence of *** review discusses the risks and benefits of a fever on the host response,focusing on the effects of a fever on viral replication and host response,and the detrimental effect on the host.

XGBoost model for electrocaloric temperature change prediction in ceramics

An eXtreme Gradient Boosting(XGBoost)machine learning model is built to predict the electrocaloric(EC)temperature change of a ceramic based on its composition(encoded by Magpie elemental properties),dielectric constant,Curie temperature,and characterization conditions.A dataset of 97 EC ceramics is assembled from the experimental *** sampling data from clusters in the feature space,the model can achieve a coefficient of determination of 0.77 and a root mean square error of 0.38 K for the test *** analysis shows that the model captures known physics for effective EC *** Magpie features help the model to distinguish between materials,with the elemental electronegativities and ionic charges identified as key *** model is applied to 66 ferroelectrics whose EC performance has not been ***-free candidates with a predicted EC temperature change above 2 K at room temperature and 100 kV/cm are identified.
