Utilization of RNA sequencing to investigate olfactory dysfunction in chronic rhinosinusitis without nasal polyps:A pilot study

Objectives:Prior research on olfactory dysfunction in chronic rhinosinusitis(CRs)has focused on patients with polyps and suggests that direct inflammation of the olfactory cleft mucosa plays a contributory role.The purpose of this study was to evaluate gene expression in superior turbinate mucosal specimens,comparing normosmic and dysosmic CRs patients without polyps(CRssNP).Methods:Tissue samples were obtained from the superior turbinates of patients with CRssNP at the time of endoscopic sinus surgery.samples subsequently underwent RNA sequencing and functional analysis to investigate biological pathways associated with differentially expressed genes between dysosmic(n=7)and normosmic(n=4)patients.Results:Differential gene expression analysis comparing dysosmic and normosmic CRssNP patients showed upregulation of 563 genes and downregulation of 327 genes.Using stringent criteria for multiple comparisons,one upregulated gene(Immediate Early Response 3[IER3])had an false discovery rate(FDR)correction adjustedP value considered statistically significant(Psis revealed eight biological pathways significantly different between dysosmic and normosmic patients(Psignaling,IL-10 signaling,and rhodopsin-like receptors.Conclusions:RNA sequencing of the superior turbinates in patients with CRssNP can provide valuable information regarding biological pathways and genes involved in olfactory dysfunction.This study supports literature suggesting that Type 2 inflammation may play a role in olfactory dysfunction in at least some patients with CRssNP.This study also prompts questions regarding the role of IL-10,rhodopsin-like receptors,andIER3 in the pathogenesis of olfactory dysfunction.

The Rural Minimum Living standard Guarantee(Rural Dibao)is an important unconditional cash transfer program to alleviate poverty in rural China.Despite the importance of children’s nutrition in breaking poverty cycles,little is known about the impact of Rural Dibao on child nutrition outcomes.Using China Family Panel studies(CFPs),this paper examines the effects of Rural Dibao on child nutrition outcomes and investigates potential pathways and heterogeneous effects.We exploit propensity score matching and difference-in-differences techniques to evaluate the effects of the Rural Dibao program on child nutrition outcomes.Our results suggest that Rural Dibao significantly impacts the nutrition outcomes of children up to 15 years of age.specifically,our results suggest that Rural Dibao improves child height-to-age z-scores by 1.05 standard deviations and lowers the probability of stunting by 11.9 percentage points.Additional analyses suggest that increased protein intake is the main pathway through which Rural Dibao participation contributes to better nutrition outcomes.We also find that the effect of the program is more pronounced among girls,children who are non-left-behind or live with highly educated mothers,and those from low-income families and poor areas.Our findings suggest that Rural Dibao participation helps improve child nutrition outcomes through improving diet quality.

Oral cancer (OC) is the most common form of head and neck cancer. Despite the high incidence and unfavourable patient outcomes, currently, there are no biomarkers for the early detection of OC. This study aims to discover, develop, and validate a novel saliva-based microRNA signature for early diagnosis and prediction of OC risk in oral potentially malignant disorders (OPMD).The Cancer Genome Atlas (TCGA) miRNA sequencing data and small RNA sequencing data of saliva samples were used to discover differentially expressed miRNAs. Identified miRNAs were validated in saliva samples of OC (n=50), OPMD (n=52), and controls(n=60) using quantitative real-time PCR. Eight differentially expressed miRNAs (miR-7-5p, miR-10b-5p, miR-182-5p, miR-215-5p,miR-431-5p, miR-486-3p, miR-3614-5p, and miR-4707-3p) were identified in the discovery phase and were validated. The efficiency of our eight-miRNA signature to discriminate OC and controls was:area under curve (AUC):0.954, sensitivity:86%, specificity:90%,positive predictive value (PPV):87.8%and negative predictive value (NPV):88.5%whereas between OC and OPMD was:AUC:0.911,sensitivity:90%, specificity:82.7%, PPV:74.2%and NPV:89.6%. We have developed a risk probability score to predict the presence or risk of OC in OPMD patients. We established a salivary miRNA signature that can aid in diagnosing and predicting OC,revolutionising the management of patients with OPMD. Together, our results shed new light on the management of OC by salivary miRNAs to the clinical utility of using miRNAs derived from saliva samples.

Pneumocystis spp. infect the lungs of multiple mammalian species and cause disease in immunosuppressed individuals. The Pneumocystis isolates that have been studied to date fall into two major clades, those from primates and those from rodents. Within each of these clades, different species have been described on the basis of host specificity and differences in sequence and morphology. Here, we demonstrate that dexamethasone immunosuppression consistently results in histologically apparent lung infection in gerbils (28/35 animals). sequence analysis of the 18s, 5.8s and internal transcribed spacer regions of the rDNA and a portion of the mitochondrial large subunit rDNA demonstrated that this gerbil Pneumocystis is grouped with other rodent Pneumocystis spp., but is distinct from them. Our results suggest that gerbil Pneumocystis differs sufficiently from Pneumocystis species found in other rodents to be considered a separate species.

Graphene is considered a promising material for industrial application based on the intensive laboratoryscale research in the fields of physics, chemistry, materials science and engineering, and biology over the last decade. Many companies have thus started to pursue graphene materials on a scale of tons(for the flake material) or hundreds of thousands of square meters(for the film material) for industrial applications. Though the graphene industry is still in its early stages, very significant progress in mass production and certain industrial applications has become obvious. In this report, we aim to give a brief review of the mass production of graphene materials for some industrial applications and summarize some features or challenges for graphene in the marketplace.

Facile dry decoration of graphene oxide sheets with aerosol Ag nanocrystals synthesized from an arc plasma source has been demonstrated using an electrostatic force directed assembly technique at room temperature.The Ag nanocrystal-graphene oxide hybrid structure was characterized by transmission electron microscopy(TEM)and selected area diffraction.The ripening of Ag nanocrystals on a graphene oxide sheet was studied by consecutive TEM imaging of the same region on a sample after heating in Ar at elevated temperatures of 100°C,200°C,and 300°C.The average size of Ag nanocrystals increased and the number density decreased after the annealing process.In particular,migration and coalescence of Ag nanocrystals were observed at a temperature as low as 100°C,suggesting a van der Waals interaction between the Ag nanocrystal and the graphene oxide sheet.The availability of affordable graphene-nanocrystal structures and their fundamental properties will open up new opportunities for nanoscience and nanotechnology and accelerate their applications.

AIM:To understand how interactions between hepatitis C virus(HCV) and the host's immune system might lead to viral persistence or effective elimination of HCV.METHODs:Nucleotides 3519-3935 of the non-structural 3(Ns3) region were amplified by using reverse transcription polymerase chain reaction(PCR).PCR products of the HCV Ns3 regions were integrated into a PCR T7TOPO TA vector and then sequenced in both directions using an automated DNA sequencer.Relative major histocompatibility complex binding levels of wild-type and variant peptides were performed by fluorescence polarization-based peptide competition assays.Peptides with wild type and variant sequences of Ns3 were synthesized locally using F-moc chemistry and purified by high-performance liquid chromatography.specific cytotoxic T lymphocytes(CTLs) clones toward HCV Ns3 wild-type peptides were generated through limiting dilution cloning.The CTL clones specifically recognizing HCV Ns3 wild-type peptides were tested by tetramer staining and flow cytometry.Cytolytic activity of CTL clones was measured using target cells labeled with the fluorescence enhancing ligand,DELFIA EuTDA.REsULTs:The pattern of natural variants within three human leukocyte antigen(HLA)-A2-restricted Ns3 epitopes has been examined in one patient with chronic HCV infection at 12,28 and 63 mo post-infection.Results obtained may provide convincing evidence of immune selection pressure for all epitopes investigated.statistical analysis of the extensive sequence variation found within these Ns3 epitopes favors a Darwinian selection model of variant viruses.Mutations within the epitopes coincided with the decline of CTL responses,and peptide-binding studies suggested a signif icant impact of the mutation on T cell recognition rather than peptide presentation by HLA molecules.While most variants were either not recognized or elicited low responses,such could antagonize CTL responses to target cells pulsed with wild-type peptides.CONCLUsION:Cross-recognition of CTL epitopes from wild-type and n

Underwater sound and video observations were made at noon, sunset, and midnight in sand, gravel, and boulderhabitat in the stellwagen Bank National Marine sanctuary, Gulf of Maine, UsA in October 2001 using a remotely operated vehicle(ROV). seventeen species of fish and squid were observed with clear habitat and time differences. Observations of feedingbehavior, disturbance behavior, and both interspecific and intraspecific interactions provided numerous opportunities for potentialsound production; however, sounds were recorded only during a single dive. Although high noise levels generated by the ROVand support ship may have masked some sounds, we conclude that fish sound production in the Gulf of Maine during the fall isuncommon. The recorded fish sounds are tentatively attributed to the cusk Brosme brosme. Cusk sounds consisted variously ofisolated thumps, widely spaced thump trains, drumrolls, and their combinations. Frequency peaks were observed at 188, 539, and1195 Hz. Use of a remotely operated vehicle (ROV) as a passive acoustic observation platform was problematic due to high ROVself-noise and the ROV's inability to maintain a fixed position on the bottom without thruster power. some fishes were clearlyalso disturbed by ROV noise, indicating a potential ROV sampling bias.
