Discovery.of New Stilbene Antioxidants of the Bio-Elicited Peanut Sprout Powder (BPSP) and Longevity.Extension of Mice Fed with BPSP-Supplemented Diets

Biosy.thesis of peanut stilbenes, including resveratrol as the secondary.metabolites, could be enhanced by.subjecting the kernels to germination and wound-stress. Investigations of the bio-elicited peanut sprout powder (BPSP) addressed on characterization of the comprising stilbenes and effectiveness in longevity.extension deserves intensive research. In this study. peanut kernels were subjected to germination and wound-stress in preparation of BPSP. The methanol extracts of BPSP were medium pressure liquid chromatographic (MPLC) fractionated and semi-preparative HPLC recovered and followed by.instrumental identification and biological activity.determinations of the isolated stilbenes. In longevity.experiments, 16 female 11-mon-old BALB/c mice and both genders of 12-mon-old ICR mice were daily.fed with BPSP supplemented diets at doses of 0, 0.1 and 0.5 g BPSP/kg bw for 750 and 762 day., respectively. Based on chemical characterization, enriched quantity.of stilbenes in the BPSP up to ca. 1% (w/w) was detected. Two new stilbene compounds, namely. 4, 5’-dihy.roxy.6’’-hy.roxy.ethy., 6’’-methy.py.ano [2’’, 3’’: 3’, 4’] stilbene and 3, 4, 5’-trihy.roxy.6’’, 6’’-dimethy.py.ano [2’’, 3’’: 3’, 4’]stilbene along with 5 known stilbenes were isolated. The 7 stilbenes exhibited potent antioxidative and antigly.ative activities and varied with structure-activity.nature. Based on the resultant survival curves and average lifespans of both mouse models, basal diets supplemented with BPSP are effective to extend mouse longevity.by.a dose dependent manner. It is of merit to demonstrate that peanut kernels as a potent producer could be bio-elicited to biosy.thesize a broad spectrum of bioactive stilbenes to prepare BPSP which is effective to extend mouse longevity.as science-evidenced by.the two long-term animal experiments.

落花生之黄曲毒素防治与其保健植物化合物之探讨

落花生是全球熟悉与普遍消费的作物,其特殊之香气与酥脆质地吸引绝大多数消费者的味口。然而,由于落花生可能被黄曲毒素产生菌污染产毒,导致许多消费者常将落花生与黄曲毒素(系一种天然致癌物质)联想在一起,因而犹豫甚至拒绝进食含落花生之产品。落花生虽然是黄曲毒素产生菌生长之良好基质,但必须遭受其侵入,且得到充分生长才会产毒。田间栽种时有适当灌溉、采收后花生荚果迅速干燥,以及良好仓储条件,是落花生采收前后有效降低黄曲毒素污染的最重要方法。加工时,先将花生仁轻度焙烤,再脱膜并以色差电子自动筛检,则能更进一步将被霉菌污染之花生仁筛除,筛选后之花生仁可加工制作安全无虞之高品质食品。另外,有鉴于黄曲毒素产生菌普遍存在于土壤中,但采收后之健康花生荚或花生仁几乎均无黄曲毒素之污染,经探讨得知,落花生具有生合成二苯乙烯类化合物(包括熟知的白藜芦醇在内之植物防御素)以抵抗微生物入侵之本能。因此,落花生可经由发芽制备花生芽作为机能蔬菜,并促进生合成二苯乙烯类化合物,包括白藜芦醇,Arachidin-1,Arachidin-3及Isopentadieny.resveratrol(IPD)等,这些二苯乙烯类化合物目前已知,具有相当值得重视的抗氧化与抗发炎活性,具有进一步开发作为饮食补充或保健产品之潜力。

We study.galvanic currents on a heterogeneous surface. In electrochemistry. the oxidation-reduction reaction producing the current is commonly.modeled by.a nonlinear elliptic boundary.value problem. The boundary.condition is of exponential ty.e with periodically.vary.ng parameters. We construct an approximation by.first homogenizing the problem, and then linearizing about the homogenized solution. This approximation is far more accurate than both previous approximations or direct linearization. We establish convergence estimates for both the two and three-dimensional case and provide two-dimensional numerical experiments.

Restoration of Mitochondrial Structure and Function within Helicobacter py.ori VacA Intoxicated Cells

The Helicobacter py.ori vacuolating cy.otoxin (VacA) is an intracellular, mitochondrial-targeting exotoxin that rapidly.causes mitochondrial dy.function and fragmentation. Although VacA targeting of mitochondria has been reported to alter overall cellular metabolism, there is little known about the consequences of extended exposure to the toxin. Here, we describe studies to address this gap in knowledge, which have revealed that mitochondrial dy.function and fragmentation are followed by.a time-dependent recovery.of mitochondrial structure, mitochondrial transmembrane potential, and cellular ATP levels. Cells exposed to VacA also initially.demonstrated a reduction in oxidative phosphory.ation, as well as increase in compensatory.aerobic gly.oly.is. These metabolic alterations were reversed in cells with limited toxin exposure, congruent with the recovery.of mitochondrial transmembrane potential and the absence of cy.ochrome c release from the mitochondria. Taken together, these results are consistent with a model that mitochondrial structure and function are restored in VacA-intoxicated cells.

Identification of the Tumor Factor of Abnormal Cancer Methy.ation Enzy.es as the Cataly.ic Subunit of Telomerase

OBJECTIVE The objective was to study.the relationship between the tumor factor of cancer MATLT and the cataly.ic subunit of telomerase. The function of telomerase in the blockade of cell differentiation and in the protection of DNA MT resembles closely.the function of the tumor factor of cancer MATKw. Because of this close similarity.we made an attempt to examine the possibility.that the tumor factor of MATKT might be the cataly.ic subunit of telomerase. METHODS We used purified MAT isozy.es, telomerase antibody. immunoprecipitation, and a selective inhibitor of the tumor factor of MATLT from urine to study.the relationship between the tumor factor of MATLT and telomerase. RESULTS We were able to show that the tumor MATcw, but not the liver MATL, was selectively.inhibited by.the telomerase antibody. and the tumor MATLT, but not the liver MATL, was preferentially.immunoprecipitated with the telomerase antibody. The cataly.ic subunit of telomerase was detectable in the tumor MATLT preparation by.immunoblotting, but was undetectable in the liver MATc preparation and the tumor MATc preparation stripped off of the tumor factor. In addition, PP-0.39, which is an effective differentiation inducer purified from urine previously.found to selectively.antagonize the tumor factor of MATLT, was found in this study.to be a potent inhibitor of telomerase. The inhibition of telomerase by.PP-0.39 was far more sensitive than the elimination of the tumor factor from MATLT. CONCLUSION All results are consistent with the hy.othesis that the tumor factor of MATLT is the cataly.ic subunit of telomerase. Thus, the blockade of cell differentiation by.telomerase is mediated through its interaction with MAT to affect methy.ation enzy.es, so that hy.omethy.ation of nucleic acids necessary.for the cell to undergo differentiation cannot take place.

Naringenin在碘酸钠诱导的视网膜色素上皮细胞变性和激光诱导的脉络膜新生血管鼠模型中的作用(英文)

目的:研究naringenin滴眼对碘酸钠诱导的大鼠视网膜色素上皮(retinal pigment epithelium,RPE)变性以及对激光诱导的脉络膜新生血管(choroidal neovascularization,CNV)的作用。方法:10g/Lnaringenin滴眼液预先处理1wk(3次/d),1wk后予35mg/kg碘酸钠舌下静脉注射诱导大鼠RPE变性,在2wk和4wk末,视网膜电图(electroretinogram,ERG)测量C波。另预处理1wk(3次/d),在2wk和4wk末用荧光素血管造影(fluorescein angiography.FA)和荧光显微镜测量CNV面积。结果:碘酸钠注射后2wk,碘酸钠组ERG的C波下降至对照组的53%(P<0.01)。而naringenin+碘酸钠组则无明显变化。4wk后,碘酸钠组下降至对照组的37%(P<0.01),而naringenin+碘酸钠组下降至对照组的57%(P<0.01)。与碘酸钠组比较,naringenin+碘酸钠组控制了66%的C波下降(P<0.05)。35mg/kgnaringenin组FA测量的CNV面积2,4wk末分别是对照组的53%和49%(P<0.01)。4wk后naringenin组荧光显微镜测量的CNV面积是对照组的47%(P<0.01)。结论:10g/Lnaringenin可以显著保护碘酸钠诱导的RPE变性,也能减小CNV的形成。
