MIS 5e sea surface temperature estimation;a multi-proxy approach using a marine macrofossil assemblage(Mar Piccolo,Gulf of Taranto,Southern Italy)

It is reported a multi-proxy palaeoclimatic study conducted on a MIS 5e calcarenite from the Mar Piccolo Basin(MP),Gulf of Taranto(GT)(Central Mediterranean).The calcarenite returned a rich malacofauna consisting of 120 extant species,including four of the tropical Senegalese Fauna,today absent in the *** biogeographic-climatic affinity of the assemblage shows,compared to today,a double percentage of warm affinity species,while the cold affinity species are nearly equally represented,indicating a warmer but not strictly tropical *** is confirmed by the most recurring preferred SST ranges of the assemblage,indicating an average of 20℃.The skeletal compositions of five well-preserved molluscan and coral specimens were analyzed for trace elements and stable isotopes for further mean SST *** the comparison of the results of several equations available in literature,it appears that only some SST estimations are realistic,converging into similar values of,on average,20.8±0.9℃.Considering all the used proxies,the MIS 5e SST difference compared to today falls in the range 1.2—2.0℃for the GT(being a more reasonable scenario)and 2.0—2.8℃for the *** is not a firmly tropical-like SST setting as suggested by the sole Senegalese fauna,indicating at least 2.7℃—3.5℃more than today’s GT and MP,*** approximations and assumptions made for obtaining SST values with any single proxy indicate the need of a multi-proxy approach to define the best SST estimation.

Objective: As the literature on conventional criteria for discriminating early-onset (EO) from late-onset (LO) Alzheimer’s disease (AD) is sparse and controversial, the aim of this study was to establish a precise age at onset (AAO) criterion, by using a specific statistical procedure, and to describe the clinical characteristics of the two sub-groups. Methods: Admixture analysis was performed to establish the AAO cut-off in a multi-center study including 2000 AD patients consecutively recruited in eight Italian Memory Clinics. None of the patients were taking acetylcholinesterase inhibitors, antipsychoticor anti-depressant drugs. At the first diagnosticvisit, they were administered the Mini Mental StateExamination, the Basic and Instrumental Activities of Daily Living and the Neuropsychiatric Inventorytoassess clinical phenomenology. Results: Using a specific statistical procedure, we established that AAO that discriminated EO-from LO-AD was 66. Compared with the LO-AD group, the EO-AD group showed longer duration of illness and a higher educational level as well as less severe functional impairment and delusions. Conclusions: Differences in sociodemographic and clinical characteristics, such as duration of illness, education and delusion severity, suggested the involvement of different pathogenic processes. Additional studies are needed to further investigate the mechanisms underlying the disorder in the two sub-groups of AD patients.

The purpose of this work is to study the weathering processes of the granulite rocks of the Serre Massif (southern Calabria,Italy) using a multidisciplinary approach based on field studies,geochemical modeling,and minero?petrographical *** granulite rocks are plagioclase-rich with minor amphibole,clinopyroxene,orthopyroxene,biotite,and garnet and their texture are *** reaction path modeling was performed to simulate the evolution of groundwaters upon interaction with local granulite by means of the software package EQ3/6,version *** were performed in kinetic (time) mode under a closed system at a constant temperature of 11.5℃,(which reproduces the average temperature of local area) and fixing the fugacity of CO2 at 10-2.34 bar (mean value).During the most advanced stage of weathering the main mineralogical changes are: partial destruction and transformation of biotite and plagioclase associated with neoformation of ferruginous products and secondary clay minerals producing a change in the origin rock *** secondary solid phases observed during the geochemical modeling (kaolinite,vermiculite and ferrihydrite) are similar to those found in this natural ***,the soil-like material mainly characterized by mostly sand to gravel grain-size fractions is the final result of the weathering processes.

Mitochondrial dysfunction in Parkinson＇s disease： a possible target for neuroprotection

Mitochondria are dynamic organelles which are required for maintaining cellular homeostasis. Thus, it is not surprising that irregularities in mitochondrial function result in cellular damage and are linked with neurodegenerative diseases, such as Parkinson＇s disease. Evidence that mitochondrial dysfunction is key to the pathogenesis of Parkinson＇s disease is founded in studies in post-mortem tissue from patients with Parkinson＇s disease, and also from genetic studies stemming from patients with familial Parkinson＇s disease. Whether triggered by environmental or genetic factors, mitochondrial dysfunction occurs early in the pathogenic process, and is central to Parkinson＇s disease pathology. As such, targeting the mitochondria to slow or halt disease progression is an attractive strategy for disease-modifying agents in Parkinson＇s disease. Indeed, several therapies which target the mitochondria have been investigated as neuroproteetive treatments for Parkinson＇s disease. This review will discuss the evidence supporting mitochondrial dysfunction in Parkinson＇s disease pathology as well as treatment strategies that target the mitochondria.

Comparison of a monoclonal antigen stool test (Hp StAR) with the ^(13)C-urea breath test in monitoring Helicobacter pylori eradication therapy

AIM： To evaluate the agreement between a mAb-based stool test （HP STAR） and the urea breath test （UBT） in monitoring （H pylon） infection after eradication therapy. METHODS： Patients with discordant results on UBT and Hp STAR underwent endoscopy with biopsies for rapid urease test, culture, and histology to confirm H pylori status. RESULTS： Among 250 patients （50±14 years）, 240 （96.0%） had concordant UBT and Hp STAR tests with a significant correlation between DOB and A values （R = 0.87; P〈0.0001）. The remaining 10 （4.0%） patients had discordant tests （positive Hp STAR and negative UBT） with the Hp STAR inaccurate in five cases （false positive） and UBT inaccurate in the other five cases （false negative）. The “maximal expected” sensitivity, specificity, ＋PV, -PV, ＋LR, and -LR were 91%, 100%, 100%, 97.4%, ∞, and 8.2 respectively, for the UBT, and 100%, 97.4%, 91%, 100%, 38.8, and 0, respectively, for the Hp STAR. Overall accuracy for both tests was 98%. CONCLUSION： Both the UBT and the Hp StAR are equally accurate in monitoring H pylori infection. Nowadays, the choice of the “best” non-invasive H pylori test in the post-treatment setting should be done not only in terms of diagnostic accuracy but also in view of cost and local facilities.

Oleuropein-Specific-β-Glucosidase Activity Marks the Early Response of Olive Fruits (Olea europaea) to Mimed Insect Attack

Olive fruits are seriously deteriorated by pre and postharvest damage due to the attack of insects, such as Bactrocera olaea, which strongly alters the quality of olives. Defence response in olive fruits injured both by pathogens and by mechanical damages has been associated with the enzyme β-glucosidase, which specifically hydrolyses oleuropein, producing highly reactive aldehyde molecules. In situ detection of ^-glucosidase activity in olive fruit tissues following injury, which simulates Bactrocera oleae punctures, is reported. The assay was performed in two cultivars showing different degrees of susceptibilities to fly infestation. In both cultivars, the histochemical assay for β-glucosidase showed that within 20 min after the injury, a strong ^-glucosidase activity could be observed in the damaged tissues. Thereafter a progressive enzyme inactivation occurred starting from tissues around the boundary of the injury with decrease of the enzyme activity and stopped after 3 h. Whereas the mass of active cells reached a distance of （300±50） μm from the edge of the injury. Biochemical analyses showed that in extracts of the injured fruit, β-glucosidase activity rapidly increased within 20 min from injury, thereafter decreasing and reaching values comparable with those in intact fruits. Following puncture, the oleuropein contents did not change significantly in the high susceptibility cultivar, whereas it rapidly decreased in the cultivar showing low susceptibility. The results strongly suggest that olive fruits susceptible towards fly infestation could be related to the ability of the oleuropein-degrading-β-glucosidase to produce the highly reactive molecules in the damaged tissues. As a consequence of puncture, high level of peroxidase activity was detected. This feature also suggested that this enzyme could play a key role in the defence response against insect injuries.

Head geometric morphometrics of two Chagas disease vectors from Venezuela

Triatominae species are considered the main vectors of Chagas disease or American Trypanosomiasis. In Venezuela, the principal vectors are Rhodnius prolixus （Stal, 1959） and Triatoma maculata （Erichson, 1848）, which are belonged to the tribe Rhodniini and Triatomini, respectively. The head conformation and size development of these species can reflect ontogenetic changes which contribute with the vectors biology studies, as well to support of instars determination. The goal of the paper is to the application of geometric morphometric techniques for describing head conformation and size of instars of these species. We photographed 140 heads in R. prolixus： First instar （I： 16）, second instar （II： 17）, third instar （III： 18）, fourth instar （IV： 21）, fifth instar （V： 21）, adult female （F： 26） and adult males （M： 21）; in T. maculata heads of 136 specimens were photographed,I： 20, II： 17, III： 26, IV： 15, V： 19, F： 20 and M： 19. Landmark coordinate （x, y） configurations were registered and aligned by Generalized Procrustes Analysis. Covariance Analyses were implemented with proportions of re-classified groups and MANOVA. Statistical analyses of variance found not significant differences in head isometric size （Kruskal-Wallis） among IV and V instars in both species. The a posteriori re-classification was almost perfect in R. prolixus （82%） and T. maculata （86%）; the main head differences occurs in antenniferous tubercles, postocular and preocular. Our study using quantitative tools for describing the shape differences contributes to explain the morphology variability and development of Chagas disease vectors.

Endoscopic ultrasound and magnetic resonance imaging for re-staging rectal cancer after radiotherapy

AIM： To compare the sensitivity and specificity of two imaging techniques, endoscopic ultrasound （EUS） and magnetic resonance imaging （MRI）, in patients with rectal cancer after neoadjuvant chemoradiation therapy. And we compared EUS and MRI data with histological findings from surgical specimens. METHODS： Thirty-nine consecutive patients （51.3% Male; mean age： 68.2 ＋ 8.9 years） with histologically confirmed distal rectal cancer were examined for staging. All patients underwent EUS and MRI imaging before and after neoadjuvant chemoradiation therapy. RESULTS： After neoadjuvant chemoradiation, EUS and MRI correctly classified 46% （18/39） and 44% （17/39） of patients, respectively, in line with their histological T stage （P 〉 0.05）. These proportions were higher for both techniques when nodal involvement was considered： 69% （27/39） and 62% （24/39）. When patients were sorted into T and N subgroups, the diagnostic accuracy of EUS was better than MRI for patients with T0-T2 （44% vs 33%, P 〉 0.05） and NO disease （87% vs 52%, P = 0.013）. However, MRI was more accurate than EUS in T and N staging for patients with more advanced disease after radiotherapy, though these differences did not reach statistical significance. CONCLUSION： EUS and MRI are accurate imaging techniques for staging rectal cancer, However, after neoadjuvant RT-CT, the role of both methods in the assessment of residual rectal tumors remains uncertain.
