Inheritance of marsh spot disease resistance in cranberry common bean(Phaseolus vulgaris l.)

Common bean(Phaseolus vulgaris) is an annual legume crop that is grown worldwide for its edible dry seeds and tender pods. Marsh spot(MS) of the seeds is a physio-genic stress disease affecting seed quality in beans. Studies have suggested that this disease involves a nutritional disorder caused by manganese deficiency, but the inheritance of resistance to this disease has not been reported. A biparental genetic population composed of 138 recombinant inbred lines(RIls) was developed from a cross between an MS resistant cultivar ‘Cran09’ and an MS susceptible cultivar ‘Messina’. The 138 RIls and their two parents were evaluated for MS resistance during five consecutive years from 2015 to 2019 in sandy and heavy clay soils in Morden, Manitoba, Canada. The MS incidence(MSI) and the MS resistance index(MSRI) representing disease severity were shown to be both highly correlated heritable traits that had high broad-sense heritability values(H;) of 86.5% and 83.2%, respectively. No significant differences for MSI and MSRI were observed between the two soil types in all five-(MSI) or four-year(MSRI) data collection, but significant correlations among years were observed despite MS resistance was moderately affected by year. The MSIs and MSRIs displayed a right-skewed distribution, indicating a mixed genetic model involving a few major genes and polygenes. Using the joint segregation analysis method, the same four major genes with additive-epistasis effects showed the best fit for both traits, explaining 84.4% and85.3% of the phenotypic variance for MSI and MSRI, respectively. For both traits, the M1, M2, M3 and m4 acted as the favorable(resistant) alleles for the four genes where M and m represent two alleles of each gene. However, due to epistatic effects, only the individuals of the M1 M2 M3 M4 haplotype appeared to be highly resistant, whereas those of the m1 m2 m3 M4 haplotype were the most susceptible. 禾谷孢囊线虫(cereal cyst nematode,CCN)是一类重要的土传小麦病原线虫,危害我国小麦的主要是燕麦孢囊线虫(Heterodera avenae)和菲利普孢囊线虫(***)。我国对这些病原线虫的抗性资源十分缺乏,寻找新抗源是当前抗性育种的重要工作。本研究通过3年的田间病圃和温室接种鉴定,发现加拿大的硬粒小麦品种Waskana和Waskowa对***(河南许昌群体,Hfc-1致病型)和***(河南荥阳群体,Ha43致病型)都表现很强的抗性,单株孢囊数显著少于感病的普通小麦品种矮抗58、石4185和温麦19。显微观察可见,虽然2种线虫的幼虫都能够侵入Waskana和Waskowa的根组织内,但是根内的线虫数量显著少于感病对照普通小麦品种,最终在根系上形成的可见孢囊数量也较少。Waskana和Waskowa对2种病原线虫的抗性为我国抗CCN小麦品种选育提供了有较高利用价值的新抗源。根据南澳大利亚研究所的土传病害检测服务系统对土壤中病原线虫的分子检测结果,抗CCN品种Waskana和Waskowa根际土壤中的线虫虫卵量低于感病小麦品种,因此种植后可能降低土壤中禾谷孢囊线虫危害的风险。

Background:Pine savannas are primarily managed with frequent prescribed fire(≤3 years)to promote diversity of flora and fauna,and to maintain open,park-like conditions needed by species such as the endangered red-cockaded woodpecker(Picoides borealis).However,a knowledge gap exists in our understanding of bobcat(lynx rufus)habitat selection in longleaf pine savannas and research is warranted to direct our future management ***:We examined bobcat habitat selection in a pine savanna managed with frequent fires at two spatial scales(i.e.,study area boundary[hereafter,landscape scale])and annual area of use[95%kernel density;local scale],and assessed effects of prescribed fire on bobcat habitat ***,we monitored 45 bobcats(16 males and 29 females)during 2001–***:We found differential habitat selection by *** the landscape scale,female bobcats were closer to mixed pine-hardwoods,young pine,and secondary roads,but farther from mature pine and hardwoods stands relative to *** found no difference in selection of agriculture,shrub-scrub,and primary roads between *** the annual area of use scale,female bobcats were closer to secondary roads,but farther from agriculture and shrub-scrub relative to *** found no difference in selection of mature pine,mixed pine-hardwoods,hardwoods,young pine,and primary roads between *** primarily selected for stands burned≤1.1 years ***:Our results show that bobcats exploit a broad range of habitat types in pine landscapes managed with frequent fire and commonly use recently burned stands(≤1.1 year post-fire),suggesting prey in many areas of this system are at risk of bobcat ***,we suggest land managers consider scale of selection by bobcats when developing habitat management strategies.

The first seedling or all-stage resistance （R） R gene against stripe rust isolated from Moro wheat （Triticum aes- tivum l.） using a map-based cloning approach was identified as Yr10. Clone 4B of this gene encodes a highly evolutionary- conserved and unique CC-NBS-lRR sequence. Clone 4E, a homolog of Yr10, but lacking transcription start site （TSS） and putative TATA-box and CAAT-box, is likely a non-expressed pseudogene. Clones 4B and 4E are 84% identical and divergent in the intron and the lRR domain. Gene silencing and transgenesis were used in conjunction with inoculation with differen- tially avirulent and virulent stripe rust strains to demonstrate Yr10 functionality. The Yr10 CC-NBS-lRR sequence is unique among known CC-NBS-lRR R genes in wheat but highly conserved homologs （E = 0.0） were identified in Aegilops tauschii and other monocots including Hordeum vulgare and Brachypodium distachyon. Related sequences were also identified in genomic databases of maize, rice, and in sorghum. This is the first report of a CC-NBS-lRR resistance gene in plants with limited homologies in its native host, but with numerous homologous R genes in related monocots that are either host or non-hosts for stripe rust. These results represent a unique example of gene evolution and dispersion across species.

Morphological characterization of fungi associated with the ascochyta blight complex and pathogenic variability of Mycosphaerella pinodes on field pea crops in central Alberta

Field pea crops in central Alberta were surveyed for ascochyta blight from 2011 to 2012 and fungal isolates were recovered from foliar lesions on selected plants. Cultural and microscopic characterization of the 275 isolates obtained revealed that 272 were of Mycosphaerella pinodes and three were of Phoma medicaginis var. pinodella. Ascochyta pisi or Phoma koolunga were not identified. Isolates of M. pinodes were divided into two groups,GI and GII, based on visual assessment of culture characteristics. GI isolates(light to dark, mostly gray colony color; pycnidial distribution radial and concentric; conidia10.5–14.5 × 4.2–6.2 μm most with one septum, occasionally two, constricted at the septum;spore mass light buff to flesh color) were predominant(83%), while GII isolates(dark to gray colony color; pycnidia abundant; conidia 8–16 × 3.5–6.2 μm most with 1 septum, constricted at the septum; spore mass light buff to flesh color) were less common(17%). The cultures of GII isolates were similar to recent descriptions of A. pisi, but they differed in spore color. In a host differential study, 13 pathotypes of M. pinodes were identified from 110 single-spore isolates. Pathotype I was predominant(88 isolates) and virulent on all nine differential genotypes. The other pathotypes(pathotypes II–XIII) were rare(1–6 isolates of each).Comparison of the present results with earlier studies suggests that pathotype I has been prevalent for many years, and that its aggressiveness on the host differentials has increased over time. Emphasis should be placed on breeding for resistance to M. pinodes in field pea cultivars intended for deployment in central Alberta.

Evaluation of host resistance and fungicide application as tools for the management of root rot of field pea caused by Aphanomyces euteiches

Aphanomyces root rot(ARR) of field pea(Pisum sativum), caused by Aphanomyces euteiches, can cause severe root damage, wilting, and large yield losses under wet soil conditions. To identify ways to manage this disease, the effect of A. euteiches inoculum density on field pea was studied under greenhouse and field conditions in 2015 and 2016. Increases in inoculum density reduced seedling emergence, root nodulation, and plant vigor, and resulted in increased root rot severity in both field and greenhouse tests. Seed treatments with the fungicides Apron Advance(thiabendazole + fludioxonil + metalaxyl) + Vibrance(difenoconazole + metalaxylM + sedaxane), INTEGO Solo(ethaboxam), BAS 516F(boscalid + pyraclostrobin), BAS 720F(metalaxyl + pyraclostrobin + fluxapyroxad), and BAS 516F + BAS 720F(3:1) were evaluated for their efficacy against ARR. All seed treatments except Apron Advance + Vibrance reduced root rot severity under controlled conditions. BAS 516F, BAS 720F and INTEGO Solo improved plant vigor and all treatments reduced seedling blight to varying degrees under greenhouse conditions, but not in the field. A collection of 22 pea genotypes was evaluated for resistance to root rot in field plot experiments. line 00–2067 showed the least severe root rot symptoms,whereas ‘Spring D' showed the lowest reduction in yield. The results suggest that there may be an opportunity to combine partial host resistance and fungicidal seed treatments to adequately manage ARR of field pea.
