Introduction. Fronto-tem.oral.dem.ntias (FTD) were described a century ago on the m.croscopic basis of frontal.and/or tem.oral.l.be atrophy. Progress in ne uropathol.gy, im.unohistochem.stry, biochem.stry and genetics has since shown th at they are heterogeneous entities, encom.assing m.ny different diseases with si m.l.r cl.nical.presentations. A few, such as tauopathies due to m.tations of the gene coding for tau protein (m.Pτ .form.a wel.-defined group. Definition and grouping of other types of FTD is stil. probl.m.tic. m.terial.and m.thod. We st udied a fam.l. where the m.ther and 4/8 chil.ren were affected with FTD. Cl.nica l.presentation was typical.of FTD. Onset was il.-defined with earl. (at age 40 years or l.ss) personal.ty changes. The cl.nical.course was protracted (about 3 0 years). For a l.ng period, the patients were abl. to l.ve in the com.unity in spite of obvious signs such as hyperoral.ty and l.ss of verbal.initiative; opera tive orientation as to pl.ce was preserved for a l.ng tim.: a m.te patient was s til. abl. to drive. Signs of extrapyram.dal.or m.toneuron invol.em.nt were not o bserved. Resul.s. The genetic study fail.d to detect any m.tation in m.Pτ ; the l.d score for fl.nking m.rkers was positive but not significant. Biochem.cal.st udy showed no qual.tative abnorm.l.ty in tau protein. Neuropathol.gical.study of one affected subject showed brain atrophy (962g), with el.ctive frontal.l.be in vol.em.nt. Cortical.nerve cel. l.ss was m.re m.rked in superficial.l.yers and in frontal.areas; gl.a was inconspicuous; pseudol.m.nar spongiosis was present in the m.re severel. affected zones. No argentophil.c “ Pick bodies” were seen; ubiquitin-positive, tau-negative round incl.sions were present in the cytopl.asm.of fascia dentata neurones. “ Tangl.s” were m.stl. restricted to the ento rhinal.cortex, partl. correl.ted with tau im.unoreactivity, but better with ubiq uitin im.unoreactivity. l.rge, ovoid or reniform. m.deratel. dense, spongy, gran ul.r o

Background:l.ver biopsy for the diagnosis of non-al.ohol.c steatohepatitis(NASH)is l.m.ted by its inherent invasiveness and possibl. sam.l.ng *** studies have shown that cytokeratin-18(CK-18)concentrations m.y be useful.in diagnosing NASH,but resul.s across studies have been *** aim.d to identify the util.ty of CK-18 m.0 concentrations as an al.ernative to l.ver biopsy for non-invasive identification of ***:Individual.data were col.ected from.14 registry centers on patients with biopsy-proven non-al.ohol.c fatty l.ver disease(NAFl.),and in al. patients,circul.ting CK-18 m.0 l.vel. were *** with a NAFl. activity score(NAS)≥5 with a score of≥1 for each of steatosis,bal.ooning,and l.bul.r infl.m.ation were diagnosed as having definite NASH;individual. with a NAS≤2 and no fibrosis were diagnosed as having non-al.ohol.c fatty l.ver(NAFl..Resul.s:A total.of 2571 participants were screened,and 1008(153 with NAFl.and 855 with NASH)were final.y *** CK-18 m.0 l.vel. were higher in patients with NASH than in those with NAFl.m.an difference 177 U/l.standardized m.an difference[Sm.]:0.87[0.69–1.04]).There was an interaction between CK-18 m.0 l.vel. and serum.al.nine am.notransferase,body m.ss index(Bm.),and hypertension(Pl.).CK-18 m.0 l.vel. were positivel. associated with histol.gical.NAS in m.st *** area under the receiver operating characteristics(AUROC)for NASH was 0.750(95%confidence interval.:0.714–0.787),and CK-18 m.0 at Youden’s index m.xim.m.was 275.7 U/*** sensitivity(55%[52%–59%])and positive predictive val.e(59%)were not ***:This l.rge m.l.icenter registry study shows that CK-18 m.0 m.asurem.nt in isol.tion is of l.m.ted val.e for non-invasivel. diagnosing NASH.

Sporadic venous m.l.orm.tions are genetic conditions prim.ril. caused by som.tic gain-of-function m.tation of PIK3CA or TEK,an endothel.al.transm.m.rane receptor signal.ng through *** m.l.orm.tions are associated with pain,bl.edings,throm.osis,pul.0}nary em.ol.sm.esthetic deform.ties and,in severe cases,l.fe-threatening *** authorized m.dical.treatm.nt exists for patients with venous ***,we created a genetic m.use m.del.of PIK3CA-rel.ted capil.ary venous m.l.orm.tions that repl.cates patient *** showed that these m.l.orm.tions onl. partial.y signal.through AKT *** com.ared the efficacy of different drugs,incl.ding rapam.cin,a m.ORC1 inhibitor,m.ransertib,an AKT inhibitor and al.el.sib,a PI3Kαinhibitor at im.roving the l.sions seen in the m.use *** dem.nstrated the effectiveness of al.el.sib in preventing vascul.r m.l.orm.tions’occurrence,im.roving the al.eady establ.shed ones,and prol.nging *** these findings,we were authorized to treat 25 patients with al.el.sib,incl.ding 7 chil.ren displ.ying PIK3CA(n=16)or TEK(n=9)-rel.ted capil.ary venous m.l.orm.tions resistant to usual.therapies incl.ding sirol.m.s,debul.ing surgical.procedures or percutaneous *** assessed the vol.m. of vascul.r m.l.orm.tions using m.gnetic resonance im.ging(m.I)for each *** dem.nstrated im.rovem.nt in al. 25 *** m.l.orm.tions previousl. considered intractabl. were reduced and cl.nical.sym.tom. were *** showed a decrease of 33.4%and 27.8%in the m.dian vol.m. of PIK3CA and TEK m.l.orm.tions respectivel.,over 6 m.nths on *** concl.sion,this study supports PI3Kαinhibition as a prom.sing therapeutic strategy in patients with PIK3CA or TEK-rel.ted capil.ary venous m.l.orm.tions.
