Exploration of Methane Mitigation Efficacy Using Asparagopsis-Derived Bioactives Stabilized in Edible Oil Compared to Freeze-Dried Asparagopsis in Vitro

Asparagopsis oil products are of interest due to the stabilizing effects of the Asparagopsis-derived antimethanogenic bioactive compound bromoform (CHBr3). The objective of this in vitro series is to characterize antimethanogenic efficacy of freeze-dried Asparagopsis (FD-Asp) and Asparagopsis oil (Asp-Oil) and compare relative antimethanogenic response over time at multiple levels of CHBr3 delivery. Relative methane (CH4) emissions (mL/g) are based on in vitro apparent feed digested dry matter (IVDDM) after 24, 48, and 72 h of fermentation. CHBr3 contained in FD-Asp was included at 95, 191, and 286 mg/kg, and CHBr3 contained in Asp-Oil was included at 78, 117, and 175 mg/kg, to produce the Low, Mid, and High inclusions, respectively. Low FD-Asp had no significant impact on CH4 emissions, Mid FD-Asp demonstrated 91%, 44%, and 37% reductions, and the High FD-Asp demonstrated complete inhibition of CH4, after 24, 48, and 72 h of fermentation, respectively. Comparatively, Low Asp-Oil demonstrated a 46%, 28%, and 18% CH4 reduction, Mid Asp-Oil resulted in 99%, 92%, and 73% reductions, and the High Asp-Oil demonstrated complete inhibition of CH4 after 24, 48, and 72 h of fermentation, respectively. IVDDM and total volatile fatty acid (tVFA) production were not changed by the inclusion of FD-Asp and Asp-Oil. The results from this study show that Asparagopsis is not only a compelling CH4 mitigating feed supplement but is also able to be delivered in edible oil forms which will strengthen its applicability to on-farm use. This study is promising for the utility of Asp-Oil, and in vivo trials are essential to demonstrate the extent of efficacy of Asp-Oil in ruminant animals because FD-Asp has consistently demonstrated greater antimethanogenic efficacy in vivo compared to in vitro.

BACKGROUND:Ectopic pancreas is defined as pancreatic tissue found outside its usual anatomical position,with no ductal or vascular communication with the native pancreas. We describe a case of ectopic pancreas of the small bowel and mesentery causing recurrent episodes of pancreatitis, initially suspected on computed tomography(CT)and magnetic resonance cholangiopancreatography(MRCP),and confirmed on histological review of the resection. METHODS:A 67-year-old woman presented with clinical symptoms and biochemical evidence of pancreatitis.She had similar episodes over the past 30 years with unrevealing investigations,and was concluded to have idiopathic pancreatitis. She underwent CT and MRCP,with findings suggestive of ectopic pancreas,a diagnosis confirmed on histology of the resection. RESULTS:MRCP identified a mass in the proximal small bowel mesentery isointense to the native pancreas,with a small duct draining into a proximal jejunal loop.The resected specimen consisted of normal parenchyma with lobulated acinar tissue with scattered islets of Langerhans,an occasional ductular structure,and admixed areas of adipose tissue.The patient remained asymptomatic with normal biochemistry six months post-operatively. CONCLUSION:In an individual with abdominal pain,elevated serum amylase/lipase,but imaging findings of a normal native pancreas,ectopic pancreatitis should be considered,and can be evaluated by CT and MRCP.

Numerical Optimization of a Walk-on-Spheres Solver for the Linear Poisson-Boltzmann Equation

Stochastic walk-on-spheres(WOS)algorithms for solving the linearized Poisson-Boltzmann equation(LPBE)provide several attractive features not available in traditional deterministic solvers:Gaussian error bars can be computed easily,the algorithm is readily parallelized and requires minimal memory and multiple solvent environments can be accounted for by reweighting trajectories.However,previouslyreported computational times of these Monte Carlo methods were not competitive with existing deterministic numerical methods.The present paper demonstrates a series of numerical optimizations that collectively make the computational time of these Monte Carlo LPBE solvers competitive with deterministic methods.The optimization techniques used are to ensure that each atom’s contribution to the variance of the electrostatic solvation free energy is the same,to optimize the bias-generating parameters in the algorithm and to use an epsilon-approximate rather than exact nearest-neighbor search when determining the size of the next step in the Brownian motion when outside the molecule.

Germanium （Ge） has gained much interest due to the potential of becoming a direct band gap material and an efficient light source for the future complementary metal-oxide-semiconductor （CMOS） compatible photonic integrated circuits. In this paper, highly biaxial tensile strained Ge quantum wells （QWs） and quantum dots （QDs） grown by molecular beam epitaxy are presented. Through relaxed step-graded InGaAs buffer layers with a larger lattice constant, up to 2.3% tensile-strained Ge QWs as well as up to 2.46% tensile-strained Ge QDs are obtained. Characterizations show the good material quality as well as low threading dislocation density. A strong increase of photoluminescence （PL） with highly tensile strained Ge layers at low temperature suggests the existence of a direct band gap semiconductor.

This paper describes the physical model testing of a wave energy converter (WEC) undertaken in the Heriot-Watt wave basin during October 2010 as part of the SUPERGEN2 project funded by the British government,and provides a preliminary analysis of the extreme mooring loads.Tests were completed at 1/20 scale on a single oscillating water column device deployed with a 3-line taut mooring configuration.The model was fully instrumented with mooring line load cells and an optical motion tracker.The tests were preceded by calibration of instrumentation and the wave test environment,and carried out in long crested waves regimes with 12 combinations of peak period T p and significant wave height H s.The main objective for these experiments was to examine the effect of shape and size of the tethered buoy on the leading mooring line on the maximum mooring loads and the excursion of the device.Comparison of the loads at different configurations of the tethered buoy suggests that the results are consistent with the hypothesis that the mooring forces should depend on the change in stiffness of the mooring system.In particular,the results indicate that with the spectral peak period close to the natural period of the moored device of 8 s,peak loads in a configuration with a smaller buoy may be considerably higher than those with a larger buoy.However,when T p was dissimilar,a harder mooring with a smaller spherical buoy appears to result in lower peak loads.The exact configuration should,therefore,be chosen according to the prevalent conditions of any particular location,and will also depend on the design and expected maintenance schedule,as well as matters related to the risk to navigation,environmental effects and the conservation status of the area.

Examples of practical approaches to molecular epidemiology of human cancer are described. Biomarkers of carcinogen exposure or inherited host factors for cancer susceptibility are discussed. Major advances have been made in the detection of carcinogen-macromolecular adducts through the use of high performance liquid chromatography, immunoaffinity chromatography, the 32P-postlabeling assay, enzyme immunoassays, gas chromatography/mass spectroscopy and synchronous spectrophotofluorimetry. The polycyclic aromatic hydrocarbon- DNA adducts are the most extensively studied in this field and together with antibodies to these adducts found in human serum, they have become useful indicators of exposure to carcinogens. Assays for various kinds of alky 1-DNA adducts have also been developed and the presence of these adducts have been documented in human tissues. Carcinogen-protein adducts have proven to be useful molecular dosimeters of carcinogen exposure. For example, 4-aminobiphenyl hemoglobin adducts are highly correlated with exposure to tobacco smoke. The study of the molecular aspects of interindividual differences in the metabolism and activation of xenobiotics and other genetic markers [DNA-restriction fragment length polymorphisms (RFLPs), mutations, and functional loss of specific genes in carcinogenesis] is an emerging new field that is discussed in the context of genetic susceptibility to cancer. The cytochrome P450 phenotypes and acetylation phenotype are examples of genetic markers that indicate an individual's potential for metabolism of exogenous substances. Further, inherited genetic polymorphic markers, e.g., DNA-RFLPs at protooncogene loci (HRAS-1 and L-myc) have been examined in a case-control study of lung cancer. Data concerning mutations of protooncogenes (H-, K-, and N-RAS) and tumor suppressor genes (retinoblastoma and p53 genes) in various common cancers are providing evidence of multiple genetic lesions that occur during the multistage process of carcinogenesis.
