Therapeutic Effect of Dimethyl Dimethoxy Biphenyl Dicarboxylate on Collagen-Induced Arthritis in Rats

Objective： To study the effect of oral administration of dimethyl dimethoxy biphenyl dicarboxylate （DDB） on adjusting angiogeneic/inflammatory mediators and ameliorating the pathology of bones in rats with collagen-induced arthritis （CIA）. methods： Wistar rat model of CIA was set up using bovine collagen type H. Fifty rats were divided into five groups randomly： normal, CIA model, DDB treatment, methotrexate （mTX） treatment, and combined DDB＋mTX treatment. Ankle joints of rats were imaged with digital X-ray machine to show the destruction of joints. Fore and hind paw and knee joints were removed above the ankle joint then processed for haematoxylin and eosin staining. Plasma levels of vascular endothelial growth factor （VEGF）, platelet derived growth factor, interleukin-8 （IL-8）, IL-4, tumor necrosis factor α （TNF-α）, and cyclooxygenase-2 （COX-2） were quantified by enzyme-linked immunosorbent assay. Nitric oxide levels were detected by Griess reagent. Results： Compared with the CIA model group, a remarkable reduction in various angiogenic （VEGF and IL-8） and inflammatory mediators （TNF-α, IL-4 and COX-2） after treatment with DDB either alone or combined with mTX （P〈0.05 or P〈0.01）. Histopathological and X-ray findings were confirmatory to the observed DDB anti-arthritic effect. The DDB-treated group showed amelioration in signs of arthritis which appeared essentially similar to normal. Conclusion： Our data shed light on the therapeutic efficacy of DDB in experimental rheumatoid arthritis （RA） compared with a choice drug （mTX） and it may be offered as a second-line drug in the treatment of RA.

Tumor necrosis factor-alpha -308G/A polymorphism and risk of hepatocellular carcinoma in hepatitis C virus-infected patients

Tumor necrosis factor-alpha (TNF-α) is an important cytokine in generating an immune response against infection with hepatitis C virus (HCV). The functions of TNF-α may be altered by single-nucleotide polymorphisms (SNPs) in its gene structure. We hypothesized that SNPs in TNF-α may be important in determining the outcome of an HCV infection. To test this hypothesis, we investigated the role of the polymorphism -308G/A, which is located in the promoter region of the TNF-α gene, in the progression of HCV infection in Egyptian patients using a quantitative real-time polymerase chain reaction (qRT-PCR). The distribution of this polymorphism and its impact on the serum level of TNF-α was compared between 90 HCV-infected patients [45 with HCV-induced cirrhosis and 45 with HCV-related hepatocellular carcinoma (HCC)] and 45 healthy Egyptian volunteers without any history of liver disease. Our results showed that at the TNF-α -308 position, the G/G allele was most common (78.5% ) in the study population, with the G/A and A/A alleles occurring less frequently (13.3% and 8.1% , respectively). Frequencies of G/G, G/A, and A/A genotypes were 87%, 7%, and 6% in patients with liver cirrhosis and were 94% , 4% , and 2% in patients with HCC, respectively. Serum levels of TNF-α were significantly higher in HCV-infected patients than in healthy controls, indicating that the TNF-α -308 polymorphism does not influence the production of TNF-α. The serum level of TNF-α was positively correlated with HCV infection. Taken together, these findings suggest that the TNF-α -308 polymorphism may not be a host genetic factor associated with the severity of HCV infection, but may be an independent risk factor for HCC.

Rapid immunodiagnostic assays for mycobacterium Tuberculosis infection

Purpose: There is a need for a continued effort to develop rapid immunodiagnostic assays for tuberculosis (TB) infection with greater sensitivity and specificity that can be used in the field and in the laboratory and that can be formatted for use with multiple species. This would help to obtain definitive early diagnosis of TB. The present study was developed to determine the role of using early secreted antigenic target-6 (ESAT-6) in immunodiagnosis of mycobacterium tuberculosis. methods: Serum samples were obtained from TB infected patients and normal healthy controls. Two rapid immunodiagnostic assays (Enzyme-linked immunosorbent assay (ELISA) and Immunoblotting) were performed. Results: The sensitivity of immunoblotting assay was 100%;however, ESAT-6 antigen was not able to discriminate between patients and normal controls. Application of direct ELISA using ESAT-6 antigen yielded 97.6% sensitivity and 75% specificity for the diagnosis of TB infection. Conclusion: In conclusion, the detection of antibodies against ESAT-6 antigen in the sera of TB patients by direct ELISA could be used as a preliminary assay for diagnosis of human m. tuberculosis infection. A combination of the ELISA with either radiological or microscopic examination is required to overcome the low specificity of the assay for negative results.

Structural and Tectonic Evolution of El-Faiyum Depression,North Western Desert,Egypt Based on Analysis of Landsat ETm+,and SRTm Data

False color Landsat ETm＋（bands 7, 4, 2 in RGB） and SRTm images of the EI-Faiyum depression, Egypt, highlights major NE-SW faults and other lineaments trending NW-SE and N-S. Airborne magnetic data reveal some E-W subsurface faults which are not recognized on Landsat ETm＋ and SRTm images. Ratio images （5/7, 3/1, 4/3） and （5/7, 5/1, 4） were used for lithological discrimination of different rock types. The EI-Faiyum area is occupied by sedimentary rocks of middle Eocene, Upper Eocene, Oligocene, Lower miocene, Pliocene, Pleistocene and Holocene. Understanding the structural regime of EI-Faiyum depression in the Western Desert of Egypt is significant in relation to the neotectonics of the Nile Delta and to the development of new communities west of the delta. The area is covered by Late Cretaceous, middle to Late Eocene deposits, Oligocene deposits （comprising fluviatile sediments and basalt sheets） and Pliocene and Quaternary alluvium and sand deposits. New geological and structural maps were prepared at a scale of 1 ： 100 000 using Landsat ETm＋ and SRTm images coupled with extensive field work. The major structural trends are E-W, ENE, NS and NW--the same trends observed elsewhere in North Africa from NW Sudan to NE Egypt including north and central Sinai. EI-Faiyum depression is a structurally-controlled tectonic basin, marked by northeast-striking faults along its northern margin. These faults are parallel to similar faults of the Syrian arc fold belt, which includes a group of related faults, folds, push-up structures, and basins. Qarun Lake is located in the northern part of EI-Faiyum depression, at a right-step between two strands of the NE striking fault system. We have suggested elsewhere that EI-Faiyum depression is a pull-apart basin related to extension between these mismatched strands of strike-slip faults. Dissipation of chlorpyrifos, pendimethalin, and thiophanate-methyl pesticides in compost based-versus peat-moss based-biomixture of biobeds

Biobed is a smart bioremediation system used to treat point-source pesticide *** is the main component of biobeds,and pesticide dissipation is affected by its *** study aimed to compare the effectiveness of compost-based(C)versus peat-moss-based(P)biomixtures of biobeds on tested pesticide *** concentrations(25,50,and 75 mg/kg)of chlorpyrifos,pendimethalin,and thiophanate methyl were added separately and as a mix to both biomixtures C and *** data showed the effect of biomixture type on the dissipation of the tested *** chlorpyrifos,its dissipation rate in biomixture P was more rapid than that in biomixture *** result was confirmed by the mineralization kinetic experiment,since 25%of the initial ^(14)C-chlorpyrifos concentration accumulated in the form ^(14)CO_(2) in biomixture P compared to only 14%in biomixture *** addition,the chlorpyrifos dissipation rate was influenced by the initial concentration when applied individually in biomixture *** contrast,biomixture C was more effective at pendimethalin dissipation than biomixture P,since>76%of pendimethalin was dissipated in biomixture C versus 67%in biomixture P at the same incubation *** thiophanate-methyl,the abilities of both biomixtures C and P were similar and less efficient than those of the other tested *** addition of the three tested pesticides to biomixture C only had a positive effect on both chlorpyrifos and thiophanate-methyl dissipation,while pendimethalin dissipation was similar when applied separately or as a *** activity was stimulated by the addition of separately or mixed pesticides to biomixtures C and P as measured by dehydrogenase activity.

In computer vision,emotion recognition using facial expression images is considered an important research *** learning advances in recent years have aided in attaining improved results in this *** to recent studies,multiple facial expressions may be included in facial photographs representing a particular type of *** is feasible and useful to convert face photos into collections of visual words and carry out global expression *** main contribution of this paper is to propose a facial expression recognitionmodel(FERm)depending on an optimized Support Vector machine(SVm).To test the performance of the proposed model(FERm),AffectNet is *** uses 1250 emotion-related keywords in six different languages to search three major search engines and get over 1,000,000 facial photos *** FERm is composed of three main phases:(i)the Data preparation phase,(ii)Applying grid search for optimization,and(iii)the categorization *** discriminant analysis(LDA)is used to categorize the data into eight labels(neutral,happy,sad,surprised,fear,disgust,angry,and contempt).Due to using LDA,the performance of categorization via SVm has been obviously *** search is used to find the optimal values for hyperparameters of SVm(C and gamma).The proposed optimized SVm algorithm has achieved an accuracy of 99%and a 98%F1 score.

Impact of cirrhosis on surgical outcome after pancreaticoduodenectomy

AIm:To elucidate surgical outcomes of pancreaticoduodenectomy(PD)in patients with liver ***:We studied retrospectively all patients who underwent PD in our centre between January 2002and December *** A comprised patients with cirrhotic livers,and Group B comprised patients with non-cirrhotic *** cirrhotic patients had ChildPugh classes A and B(patient’s score less than 8).Preoperative demographic data,intra-operative data and postoperative details were *** primary outcome measure was hospital mortality *** outcomes analysed included duration of the operation,postoperative hospital stay,postoperative morbidity and survival ***:Only 67/442 patients(15.2%)had cirrhotic *** blood loss and blood transfusion were significantly higher in group A(P=0.0001).The mean surgical time in group A was significantly longer than that in group B(P=0.0001).Wound complications(P=0.02),internal haemorrhage(P=0.05),pancreatic fistula(P=0.02)and hospital mortality(P=0.0001)were significantly higher in the cirrhotic *** stay was significantly longer in group A(P=0.03).The median survival was 19 mo in group A and 24 mo in group *** hypertension(PHT)was present in 16/67 cases of cirrhosis(23.9%).The intraoperative blood loss and blood transfusion were significantly higher in patients with PHT(P=0.001).Postoperative morbidity(0.07)and hospital mortality(P=0.007)were higher in cirrhotic patients with ***:Patients with periampullary tumours and well-compensated chronic liver disease should be routinely considered for PD at high volume centres with available expertise to manage liver *** is associated with an increased risk of postoperative morbidity in patients with liver cirrhosis;therefore,it is only recommended in patients with Child A cirrhosis without portal hypertension.
