Chronic training status affects muscle excitation of the vastus lateralis during repeated contractions

This study examined electromyographic amplitude(EMGRMS)-force relationships during repeated submaximal knee extensor muscle actions among chronic aerobically-(AT),resistance-trained(rt),and sedentary(SED)*** adults(5/group)attempted 20 isometric trapezoidal muscle actions at 50%of maximal *** electromyography(EMG)was recorded from vastus lateralis(VL)during the muscle *** the first and last successfully completed contractions,linear regression models were fit to the log-transformed EMGRMS-force relationships during the linearly increasing and decreasing segments,and the b terms(slope)and a terms(antilog of y-intercept)were *** was averaged during steady *** the AT completed all 20 muscle *** the first contraction,the b terms for rt(1.3010.197)were greater than AT(0.9100.123;p?0.008)and SED(0.9120.162;p?0.008)during the linearly increasing segment,and in comparison to the linearly decreasing segment(1.0180.139;p?0.014),*** the last contraction,the b terms for rt were greater than AT during the linearly increasing(rt?1.3730.353;AT?0.8830.129;p?0.018)and decreasing(rt?1.5260.328;AT?0.9700.223;p?0.010)*** addition,the b terms for SED increased from the linearly increasing(0.9680.144)to decreasing segment(1.2680.126;p?0.015).There were no training,segment,or contraction differences for the a *** during steady force increased from the first-([64.0851.68]μV)to last-contraction([86.7349.55]μV;p?0.001)collapsed across training *** b terms differentiated the rate of change for EMGRMS with increments in force among training groups,indicating greater muscle excitation to the motoneuron pool was necessary for the rt than AT during the linearly increasing and decreasing segments of a repetitive task.

Increased hepcidin expression in colorectal carcinogenesis

AIM:To investigate whether the iron stores regulator hepcidin is implicated in colon cancer-associated anae- mia and whether it might have a role in colorectal car- cinogenesis. METHODS: Mass spectrometry (MALDI-TOF MS and SELDI-TOF MS) was employed to measure hepcidin in urine collected from 56 patients with colorectal cancer. Quantitative Real Time rt-PCR was utilised to determine hepcidin mRNA expression in colorectal cancer tissue. Hepcidin cellular localisation was determined using im- munohistochemistry. RESULTS: We demonstrate that whilst urinary hepcidin expression was not correlated with anaemia it was posi- tively associated with increasing T-stage of colorectal cancer (P rthermore, we report that hepcidin mRNA is expressed in 34% of colorectal cancer tissue specimens and was correlated with ferroportin repres- sion. This was supported by hepcidin immunoreactivity in colorectal cancer tissue. CONCLUSION: We demonstrate that systemic hepcidin expression is unlikely to be the cause of the systemic anaemia associated with colorectal cancer. However, we demonstrate for the first time that hepcidin is expressed by colorectal cancer tissue and that this may represent a novel oncogenic signalling mechanism.

Bi_(2)Te_(3)-based alloys are known to have outstanding thermoelectric *** structure–property relations have been studied,still,detailed analysis of the atomic and nano-scale structure of Bi_(2)Te_(3)thin film in relation to their thermoelectric properties remains poorly ***,highly-textured(HT)and single-crystal-like(SCL)Bi_(2)Te_(3)films have been grown using pulsed laser deposition(PLD)on Si wafer covered with(native or thermal)SiOx and mica *** films are highly textured with c-axis out-of-plane,but the in-plane orientation is random for the films grown on oxide and single-crystal-like for the ones grown on *** power factor of the film on thermal oxide is about four times higher(56.8μW·cm^(−1)·K^(−2))than that of the film on mica(12.8μW·cm^(−1)·K^(−2)),which is comparable to the one of the polycrystalline ingot at room temperature(rt).Reduced electron scattering in the textured thin films results in high electrical conductivity,where the SCL film shows the highest ***,its Seebeck coefficient shows a low *** measured properties are correlated with the atomic structure details unveiled by scanning transmission electron *** instance,the high concentration of stacking defects observed in the HT film is considered responsible for the increase of Seebeck coefficient compared to the SCL *** study demonstrates the influence of nanoscale structural effects on thermoelectric properties,which sheds light on tailoring thermoelectric thin films towards high performance.

Expression in Arabidopsis of a Strawberry Linalool Synthase Gene Under the Control of the Inducible Potato PI2 Promoter

To investigate the role of inducible linalool in Arabidopsis-insect interactions, the FANES 1 linalool synthase （LIS） cDNA from strawberry with plastid targeting and a synthetic intron （LIS＇） was placed under the control of the wound inducible proteinase inhibitor 2 （PI2） promoter from potato. The construct pBin-PPi2-LIS＇ was transformed to Arabidopsis thaliana ecotype Columbia 0. Kanamycin resistant TO seedlings were confirmed for the presence and transcription of the LIS＇ gene by PCR analysis on genomic DNA and by rt-PCR analysis on RNA. Genomic and rt-PCR products were sequenced to confirm correct splicing of the synthetic intron. The expression of active linalool synthase by the PPI2-LIS＇ gene construct in the transgenic lines was assessed by measuring linalool emission using solid phase micro-extraction （SPME） GC-MS measurements after induction with methyl jasmonate. Among 30 tested independent T2 transgenic lines, 10 exhibited linalool production. Linalool expression could be induced by methyl jasmonate treatment, but not by diamondback moth larvae.

Methylation of TIMP3 in esophageal squamous cell carcinoma

AIM: To measure the frequency of DNA methylation of the tissue inhibitor of metalloproteinase 3 (TIMP3) promoter and relate this to any change of gene expression in esophageal squamous cell carcinoma in patients from a region of high incidence in ***: Cancer cell lines were treated with or without the demethylating reagent 5-aza-2’-deoxycytidine. Methylation of the TIMP3 promoter was assessed in three regions by melt curve analysis and its expression was assessed by real-time rt-PCR. Tumors and proximal resection margins were obtained from 64 patients with esophageal squamous cell carcinoma from a region of high incidence in China. Methylation was assessed by melt curve analysis and expression by ***: Methylation in one of the three promoter regions assessed correlated with gene silencing in esophageal cell lines. A degree of methylation of TIMP3 was found in only four esophageal squamous cell carcinomas, and partial loss of TIMP3 protein expression in just ***: Methylation and loss of expression of TIMP3 occurs infrequently in esophageal squamous cell carcinoma in a region of high incidence in China.

Using cylindrical implosions to investigate hydrodynamic instabilities in convergent geometry

AIM: To establish the potential of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx) as a material for tendon repair. METHODS: The biocompatibility of PHBHHx with both rat tenocytes (rt) and human mesenchymal stem cells (hMSC) was explored by monitoring adhesive characteristics on films of varying weight/volume ratios coupled to a culture atmosphere of either 21% O2 (air) or 2% O2 (physiological normoxia). The diameter and stiffness of PHBHHx films was established using optical coherence tomography and mechanical testing, respectively. RESULTS: Film thickness correlated directly with weight/volume PHBHHx (r2 = 0.9473) ranging from 0.1 mm (0.8% weight/volume) to 0.19 mm (2.4% weight/volume). Film stiffness on the other hand displayed a biphasic response which increased rapidly at values > 1.6% weight/volume. Optimal cell attachment of rt required films of ≥ 1.6% and ≥ 2.0% weight/volume PHBHHx in 2% O2 and 21% O2 respectively. A qualitative adhesion increase was noted for hMSC in films ≥ 1.2% weight/volume, becoming significant at 2% weight/volume in 2% O2. An increase in cell adhesion was also noted with ≥ 2% weight/volume PHBHHx in 21% O2. Cell migration into films was not observed. CONCLUSION: This evaluation demonstrates that PHBHHx is a suitable polymer for future cell/polymer replacement strategies in tendon repair.

EXPRESSION OF BCRP GENE IN THE NORMAL LUNG TISSUE AND LUNG CANCER

Objective: To investigate the expression of novel multidrug resistance transporter (BCRP gene) from human MCF-7/AdrVp breast cancer cells in normal lung tissue and non-small lung cancer tissue. Methods: RNA was extracted immediately from fresh normal lung tissue and viable tumor tissue harvested from surgically resected specimens of non-small cell lung cancer patients. cDNA of BCRP gene was prepared by rt-PCR and was then amplified by PCR. cDNA products from those specimens were transferred to blotting membrane through electrophoresis and transferring technique and southern blot hybridization was eventually performed to detect the expression of BCRP gene. Results: RNA were extracted from 8 tumor tissue alone and 12 pairs of tumor tissue and normal lung tissue harvested from the same lung. Four patients’ RNA samples with poor quality due to degrading were discarded. cDNA products of BCRP gene were obtained by rt-PCR and were then amplified by PCR in the remain 16 patients’ RNA samples. Through southern blot hybridization, BCRP gene was found to be slightly expressed in various amounts in all normal lung tissue (10/10) and only in a half of tumor tissue samples (8/16). Conclusion: BCRP gene is slightly expressed in different amount in all normal lung tissue and only in a half of tumor tissue of non-small cell lung cancer patients. It is possible to induce it’s overexpression and to develop multidrug resistance during chemotherapy if using anthracycline anticancer drugs.

KINETIC STUDY OF SPINEL LiMn_2O_4 SYNTHESIZED BY SOLID REACTION WITH SOFT-CHEMICAL METHOD

The differential thermal analysis （DTA） curves were measured at different heating rates in flowing air for studying the synthesis of the spinel LiMn2O3 with Li2CO3 and MnO2, The reaction began at about 503K, and finished at about 873K. The apparent activation energy of Kissinger method was about ***^-1, the reaction orderwas 1.67, the frequency factor was 7.81×10^9, and therefore the dinetic epuation was dδ/dt=A·exp（- E/rt）·（1-δ）^n=7.81×10^9, exp（-122770/rt）·（l-δ）^1.67 . Coats-Redfem integral method was used to analyze the DTA curves of the samples at different heating rates, and the calculated apparent activation energy and frequency factor were 112. 13kj· mol^-1 and 1.18 × 10^9, respectively, rather close to that of Kissinger method. X-ray diffraction （XRD） and scanning electron microscope （SEM） results shown that the synthesized LiMn2O3 possesses pure phase, regular shape and normal particle distribution.
