Calcarea carbonica treatment rescues lipopolysaccharide-induced inflammatory response in human mononuclear cells via downregulation of inducible cyclooxygenase pathway

Objective: Prolonged use of nonsteroidal anti-inflammatory drugs is associated with severe side effects and toxicity. Therefore, we studied the anti-inflammatory role of Calcarea carbonica which had minimal toxicity at the low ***: THP-1 human mononuclear cells were treated with C. carbonica to evaluate the 50% cytotoxicity concentration(CC_(50)) and 50% effective concentration(EC_(50)). Cell survival was evaluated in lipopolysaccharide-stimulated C. carbonica-treated cells. Nitric oxide(NO) and tumor necrosis factor-a(TNF-a) were measured to evaluate the anti-inflammatory activity of C. carbonica. Cyclooxygenase-2(COX-2) protein expression was determined by Western blotting analysis, and the interaction of C. carbonica with the COX-2 protein was evaluated using molecular docking ***: The CC_(50) and EC_(50) of C. carbonica were found to be 43.26 and 11.99 mg/mL, respectively. The cell survival assay showed a 1.192-fold(P = 0.0129), 1.443-fold(P = 0.0009) and 1.605-fold(P = 0.0004)increase in cell survival at 24, 48 and 72 h after initiating C. carbonica treatment, respectively. C. carbonica-treated cells showed a reduction in NO levels by 2.355 folds(P = 0.0001), 2.181 folds(P = 0.0001) and 2.071 folds(P = 0.0001) at 24, 48 and 72 h, respectively. The treated cells also showed a reduction in TNFa levels by 1.395 folds(P = 0.0013), 1.541 folds(P = 0.0005) and 1.550 folds(P = 0.0005) at 24, 48 and72 h, respectively. In addition, a 1.193-fold reduction(P = 0.0126) in COX-2 protein expression was found in C. carbonica-treated cells. The molecular docking showed interaction of C. carbonica with the phenylalanine 367 residue present in active site of ***: C. carbonica exhibited anti-inflammatory properties in lipopolysaccharide-stimulated cells by significantly reducing NO production and TNF-a level through downregulation of the COX-2 protein. This effect is probably mediated through interaction of C. carbonica with the phenylalanine

Object-Oriented Database Connectivity for Hand Held Devices

Due to the lack of representation of an object-oriented database on hand-held devices, the present work describes an approach to send the information in the form of an object-oriented database on hand-held devices. A smart phone is selected as a hand-held device and connectivity between the object-oriented databases has been established by the use of the object-oriented programming language C#.Net. For modeling purpose, Unified Modeling Language (UML) is used to specify a UML Class Model and Sequence diagram. A concept of star schema is also used to represent the object-oriented database.

Surrogate markers of insulin resistance:A review

Insulin resistance is a hallmark of obesity,diabetes,and cardiovascular diseases,and leads to many of the abnormalities associated with metabolic syndrome. Our understanding of insulin resistance has improved tremendously over the years,but certain aspects of its estimation still remain elusive to researchers and *** quantitative assessment of insulin sensitivity is not routinely used during biochemical investigations for diagnostic purposes,but the emerging importance of insulin resistance has led to its wider application research *** of a number of clinical states where insulin sensitivity is compromised calls for assessment of insulin resistance. Insulin resistance is increasingly being assessed in various disease conditions where it aids in examining their pathogenesis,etiology and consequences. The hyperinsulinemic euglycemic glucose clamp is the gold standard method for the determination of insulin sensitivity,but is impractical as it is labor-and time-intensive.A number of surrogate indices have therefore been employed to simplify and improve the determination of insulin *** object of this review is to highlight various aspects and methodologies for current and upcoming measures ofinsulin sensitivity/***-depth knowledge of these markers will help in better understanding and exploitation of the condition.

Representation of Object-Oriented Database for the Development of Web Based Application Using Db4o

Impedance mismatch of data flow is the major problem in the relational database when one is using the web based application development on the computer system. It effects the development of scalable and reusable web application. Due to this, the research cost of development and maintenance increases. Therefore the present work is an attempt towards an object-oriented database system by using Db4o to overcome the impedance mismatch problem. In the current work, the development of web application for Indian Postal Services by using Db4o is demonstrated on dot NET platform. Before developing the application, the authors used a Unified Modeling Language (UML) model in the form of UML class, sequence and use-case diagrams. The Db4o is used to store the object-oriented database and the performance of Db4o is observed through several object-oriented queries and corresponding results are demonstrated.

The interaction between the classes or within the classes shows the complexity of the design. For one smaller problem, there may be more than one software design but who will be the best;depends on the complexity level of software design. Therefore, coupling and cohesion which shows the interlinking of classes and strength of classes;control the complexity of the design. The best software object oriented design is based upon the low coupling and high cohesion level. In the present work, a real case study of Life Insurance policy for handicapped person is demonstrated through the UML Class Diagram;coupling and cohesion levels are measured and results are demonstrated in the form of tables.

Association of Protein Energy Wasting with Income in CKD Stage 3 Patients

Protein energy wasting (PEW) is a major challenge in CKD. Objective: To assess PEW in predialysis patients on their first visit to a nephrologist. Methods: Three day dietary intake of 484 CKD stage 3 patients was taken. Appetite was assessed with ADAT. Patients were divided into groups based on appetite and BMI. Results: Male and female parameters are serum albumin 3.7 ± 0.84/3.68.8 ± 0.81 g/dL, total protein 7.02 ± 1.27/6.94 ± 1.26 g/dL, creatinine 4.68 ± 4.19/3.74 ± 3.36 mg% creatinine clearance 33.22 ± 30.48/37.55 ± 33.87 ml/minute, BMI 22.60 ± 4.29/23.43 ± 4.77kg/m2 energy/kg 16.97 ± 0.65/16.8 ± 0.64, protein g/kg 0.65 ± 0.28/0.64 ± 0.30, carbohydrate g/kg 2.98 ± 1.54/2.98 ± 0.1.36, fat g/kg 2.98 ± 0.23/2.79 ± 0.22, respectively. As appetite decreased, dietary protein and energy intake decreased significantly. Appetite in males and females: Average 14.46%, 4.13%, poor 9.7%, 18.18%, anorexic 13.2%, 7.4%. Income had strong correlation with BMI (p 0.000), dietary protein (p 0.000), energy (p 0.000) and carbohydrate (p 0.000). Appetite correlated with creatinine (p 0.019), dietary energy, protein, carbohydrate and fat (p 0.000) intake. BMI correlated (p 0.000) with fat, carbohydrate, energy and creatinine clearance. ANOVA showed significant difference within and between appetite groups in energy, protein, fat, carbohydrate, creatinine clearance (p 0.000) and serum albumin (p 0.025). There was significant difference in protein (p 0.026), energy intake (p 0.000) and creatinine clearance (p 0.038) within and between BMI groups. Based on income, there was significant difference among groups in BMI (p 0.000), energy (p 0.019), protein (p 0.031) and albumin (0.001).

General Study of Mobile Agent Based Intrusion Detection System (IDS)

The extensive access of network interaction has made present networks more responsive to earlier intrusions. In distributed network intrusions, there are many computing nodes that are assisted by intruders. The evidence of intrusions is to be associated from all the held up nodes. From the last few years, mobile agent based technique in intrusion detection system (IDS) has been widely used to detect intrusion over distributed network. This paper presented survey of several existing mobile agent based intrusion detection system and comparative analysis report between them. Furthermore we have focused on each attribute of analysis, for example technique (NIDS, HIDS or Hybrid), behavior layer, detection techniques for analysis, uses of mobile agent and technology used by existing IDS, strength and issues. Their strengths and issues are situational wherever appropriate. We have observed that some of the existing techniques are used in IDS which causes low detection rate, behavior layers like TCP connection for packet capturing which is most important activity in NIDS and response time (technology execution time) with memory consumption by mobile agent as major issues.

Resources Allocation and Failures in Step Topology under Distributed Computing System

In the past years, distributed computing is gaining the popularity due to reduction in execution time and low cost involvement. On the basis of this, Mobile Adhoc Network (MANET) is also increasing worldwide with major advantage that it has no involvement of wire and transfer of data can be done by the virtual paths if the existing path is congested. In the present work, MANET is considered in the form of step topology which consists of heterogeneous collection of the devices. The work demonstrates the resources allocation for execution of tasks and it consists of selection of right path if the link failures and by pass link failures. It also consists of the resource management over the new proposed step topology. Entire work is modeled with the help of well known modeling language known as Unified Modeling Language (UML) and model demonstrates the resources allocation for execution of the tasks.
