B-1 cells modulate the murine macrophage response to Leishmania major infection

AIM To investigate the modulatory effect of B-1 cells on murine peritoneal macrophages infected with Leishmania major(L. major) in *** Peritoneal macrophages obtained from BALB/c andBALB/c XID mice were infected with L. major and cultured in the presence or absence of B-1 cells obtained from wild-type BALB/c mice. Intracellular amastigotes were counted, and interleukin-10(IL-10) production was quantified in the cellular supernatants using an enzymelinked immunosorbent assay. The levels of the lipid mediator prostaglandin E2(PGE2) were determined using a PGE2 enzyme immunoassay kit(Cayman Chemical, Ann Arbor, MI), and the number of lipid bodies was quantified in the cytoplasm of infected macrophages in the presence and absence of B-1 cells. Culturing the cells with selective PGE2-neutralizing drugs inhibited PGE2 production and confirmed the role of this lipid mediator in IL-10 production. In contrast, we demonstrated that B-1 cells derived from IL-10 KO mice did not favor the intracellular growth of L. *** We report that B-1 cells promote the growth of L. major amastigotes inside peritoneal murine macrophages. We demonstrated that the modulatory effect was independent of physical contact between the cells, suggesting that soluble factor(s) were released into the cultures. We demonstrated in our co-culture system that B-1 cells trigger IL-10 production by L. major-infected macrophages. Furthermore, the increased secretion of IL-10 was attributed to the presence of the lipid mediator PGE2 in supernatants of L. major-infected macrophages. The presence of B-1 cells also favors the production of lipid bodies by infected macrophages. In contrast, we failed to obtain the same effect on parasite replication inside L. major-infected macrophages when the B-1 cells were isolated from IL-10 knockout mice. CONCLUSION Our results show that elevated levels of PGE2 and IL-10 produced by B-1 cells increase L. major growth, as indicated by the number of parasites

Antimicrobial Susceptibility,Biofilm Production and Adhesion to HEp-2 Cells of Pseudomonas aeruginosa Strains Isolated from Clinical Samples

A hundred Pseudomonas aeruginosa strains from several clinical specimens from five hospitals in Sao Luís-MA were evaluated for biofilm production, prevalence of the gene algD, adhesion to HEp-2 cells and antimicrobial susceptibility. The most affected clinical specimens and hospital sectors were also evaluated. Most isolates were obtained from the tracheal aspirate (21.0%) and the most affected hospital sector was the ICU (43.0%). The antibiotics with the highest sensitivity rate were amikacin, piperacillin/tazobactam, fluoroquinolones, gentamicin and meropenem and the ones with the highest resistance rate were aztreonam, ceftazidime and cefepime. All samples were sensitive to polymyxin B. In relation to the expression of the gene for ESBL, 50.0% (17/34) of the multiresistant strains showed the enzyme TEM. Most strains showed high hydrophobicity and 96% of the isolates produced biofilm on a polystyrene microplate, 52% were capsule producers, 19% showed mannose-sensitive fimbriae and 39% expressed the gene algD. We observed adhesion to HEp-2 cells and to the coverslip. These factors may be reported in the pathogenesis of this bacterium, what represents a potential risk for colonization of medical devices which favor the establishment of chronic nosocomial infections.

Methanolic extracts from the leaves of Manihot esculenta (Two cultivars) and Manihot glaziovii, consumed as traditional vegetables in DR. Congo was chemically characterized by Thin layer Chromatography and High Performance Liquid Chromatography. In vitro biochemical activities of extracts against Radical Oxidative Species (ROS) production were assessed in cellular models, on enzymes, Myeloperoxidase (MPO) and Horseradish Peroxidase (HRP) involved in inflammation. The microscopic analysis of the powder of leaves showed that each species displays specific and discriminating botanical microscopic features. Varieties of M. esculenta had a chemical fingerprint different from M. glaziovii. The majority of compounds were polyphenols, represented mainly by rutin, kaempferol-3-O-rutinoside, amentoflavone, phenolic acids such as gallic acid. All extracts exhibited high cellular antioxidant activity in the range of 0.1 to 10 μg·mL-1 using lucigenin with neutrophils, but a moderate cellular antioxidant activity ranging between 10 and 100 μg·mL-1 with DCFDA on HL60 monocytes. Extracts from Manihot leaves showed a pronounced inhibitory effect on the production of extracellular ROS, on HRP and myeloperoxidase activity. Cellular antioxidant activities, the inhibitory effect on HRP of extracts from M. glaziovii, M. esculenta cultivar Mwambu were significantly higher, but their inhibitory effect on the activity of MPO was lower than those of M. esculenta cultivar TEM 419. The biological activities of Manihot esculenta and Manihot glaziovii were well correlated to their phytochemicals that could justify their traditional use as vegetables, potential functional foods or nutraceutical resources and medicines.

Antineoplastic phospholipids are a new class of antitumor *** molecules interact with the plasma membrane,changing numerous pathways that induce cell death,with high selectivity for cancer cells.A representative of this class of antineoplastic agents is 2-aminoethyl dihydrogen phosphate(2-AEH2P).It is present in high intracellular concentrations in various tissues and organelles with antitumor,antiproliferative and pro-apoptotic ***,4T1 triple-negative tumor cells were treated in different concentrations in order to assess the cytotoxic potential and its effects on the modulation of cell death pathways in association with the chemotherapeutic drug Paclitaxel.2-AEH2P promoted cytotoxicity in tumor cells and significant morphological changes,however,it did not cause these effects in normal *** was positive regulation of proteins involved in the intrinsic pathway of cell death by apoptosis and regulation of the phases of cell cycle ***,structural and distribution changes in mitochondria,as well as decreased cell density and regression of the cytoskeleton were *** 2-AEH2P demonstrated a modulatory potential of apoptotic pathways inducing cell death,being a new compound with antitumor properties.

Association between Tooth Loss and Hypertension: A Systematic Review

Background: An independent relationship between tooth loss and hypertension appears to exist. Objectives: The aim of this study was to systematically review the relationship between tooth loss/number of teeth and hypertension. Materials and Methods: This review was conducted based on Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) Guidelines. We searched all the relevant studies in international databases of PubMed and Google Scholar search engines from January 2000 to December 2023. Results: Fourteen studies (11 cross sectionals, 2 descriptives and 1 Cohort) met the inclusion criteria for this review. Most cross-sectional studies showed that participants with more tooth loss exhibited a greater proportion of hypertension and higher systolic blood pressure than those with less tooth loss. Cohort study showed that the loss of 8 or more teeth was associated with a higher risk of developing hypertension. Descriptive studies reported that missing teeth were marginally significantly associated with hypertension. Conclusion: This study has revealed a significantly higher SBP in patients with fewer remaining teeth or more tooth loss. In addition, those with more tooth loss showed a higher risk of hypertension than those with less tooth loss.

Intraperitoneally Administered Lidocaine Attenuates Thermal Allodynia in a Murine “Two-Hit” Chronic Constriction Injury Model

Background: Mechanical ventilation (hit one) during surgery (hit two) is often needed and both induce an inflammatory response. Dysregulation of the inflammatory response can cause chronic postoperative pain. Methods: Healthy C57BL6 mice (n = 56) were mechanically ventilated (MV) and allocated to receive sham (MV-sham) or mechanically ventilation with chronic constriction injury (MV-CCI) surgery in the left hind paw. Plasma interleukin (IL)-1β, IL-6, IL-10, keratinocyte derived chemokine (KC) and tumor necrosis factor (TNF)-α were determined on day 0 and 16. Sensory testing was performed on day 0, 3, 7 and 16 by cold plate test (number of lifts (NOL) and cumulative reaction time (CRT)) and von Frey test. The effect of lidocaine on cytokines and sensory testing was analyzed. Results: MV-Sham showed an increase in IL-1β and TNF-α, and MV- CCI-lido increased levels of KC compared with MV on day 0. No difference in cytokine levels was observed on day 16. NOL of the left paw versus the right was increased in MV-CCI on day 7, and in MV-CCI-lido on day 7 and 16. The NOL of the left paw was decreased in MV-sham and MV-CCI-lido compared with MV-CCI on day 16. The CRT of the left paw was increased for MV-CCI on day 3 and 7, and for MV-CCI-lido on day 7. On day 16, MV-sham and MV-CCI-lido showed a decreased CRT of the left paw compared with MV-CCI. Conclusion: Nerve injury and not systemic inflammatory response seems mandatory for development of neuropathic pain in this “two-hit” model. Lidocaine attenuates cold allodynia in mice.

2-aminoethyl Dihydrogen Phosphate as a Modulator of Proliferative and Apoptotic Effects in Breast Cancer Cell Lines

Background: Breast cancer is a type of cancer that affects more women throughout the world, in developing anddeveloped countries. 2-AEH2P is a phospholipid analog of cellular membrane, which makes it different from existing molecules fortheir absorption, stability and display anti-inflammatory, anti-proliferative and pro-apoptotic properties. Methods: MCF-7 humanbreast adenocarcinoma cells were treated with 2-AEH2P. The viability and adhesion cells were evaluated by MTT assay. Cell cyclephases, apoptosis, markers and mitochondrial potential were assessed by flow cytometry. Morphological ultrastructural analyzeswere performed by laser confocal microscopy. Results: MCF-7 Tumor cells acquired round shapes, lost cytoplasmic expansions,formed clusters in suspension and decreased significantly viability. There were changes in the morphology, membrane fragmentationand loss of cytoplasmic projection. The obtained concentrations for IC50% were 37.2;25.8;1.8 mM for periods of 24, 48 and 72 h,respectively. Changes in the distribution of cell population phases of the cell cycle showed an increase in fragmented DNA and anincrease in the G2/M phase. The expression β-gal showed proliferative reduction induced by 2-AEH2P. Laser confocal microscopyshowed changes in the mitochondrial membrane and alteration in distribution. Proliferative index of MCF-7 tumor cells treated with2-AEH2P decreased significantly when compared to fibroblast normal cells. The compound 2-AEH2P is a phospholipid withantiproliferative potential and apoptosis modulator.

Background:To monitor the prevalenee of schistosomiasis in school-aged children(SAC),the National Bilharzia Control Program(PNLB)was set up by the Senegalese authorities;however,geographically isolated Bedik ethnic groups that did not benefit from this program were found to be heavily infected with Schistosoma *** observation led us to implement a new schistosomiasis control program in 2008 under the aegis of the non-governmental organi-zation wLe KaTcedratwand in partnership with the PNLB/WHO to monitor the prevalence of schistosomiasis in this *** the village of Assoni,where 100%of SAC were infected,analysis of the stools of pre-school-aged children(PSAC)showed that they were massively infected,so we decided to focus our program on them.

Twin Anemia Polycythemia Sequence: Knowledge and Insights After 15 Years of Research

Twin anemia polycythemia sequence (TAPS) is a chronic form of unbalanced feto-fetal transfusion through minuscule placental anastomoses in monochorionic twin pregnancies, leading to anemia in the donor twin and polycythemia in the recipient twin. TAPS can occur spontaneously in up to 5% of monochorionic twins or can arise in 2%-16% of cases after incomplete laser surgery for twin-twin transfusion syndrome. TAPS can develop across the entire second and third trimester. Antenatal diagnosis for TAPS is reached via Doppler measurement of the fetal middle cerebral artery peak systolic velocity, showing an increased velocity in the donor, combined with a decreased velocity in the recipient. Treatment options for TAPS include expectant management, preterm delivery, intrauterine blood transfusion with or without a partial exchange transfusion, fetoscopic laser surgery and selective feticide. The best treatment option is unclear and is currently being investigated in an international multicenter randomized trial (the TAPS trial). Spontaneous fetal demise occurs in 5%-11% of TAPS twins, more often in donors (8%-18%) than in recipients (2%-5%). Severe long-term neurodevelopmental impairment is seen in 9% of TAPS twins, with donors having an increased risk for cognitive impairment and hearing problems (15%).
