Cytokinesis is required for faithful division of cytoplasmic components and duplicated nuclei into two daughter ***,a protein-dense organelle that forms at the intercellular bridge,is indispensable for success-ful ***,the regulatory mechanism of cytokinesis at the midbody still remains ***,we unveil a critical role for NudC-like protein 2(NudCL2),a co-chaperone of heat shock protein 90(Hsp90),in cytokinesis regulation by stabilizing regulator of chromosome condensation 2(RCC2)at the midbody in mam-malian ***2 localizes at the midbody,and its downregulation results in cytokinesis failure,multinu-cleation,and midbody *** iTRAQ-based quantitative proteomic analysis,we find that RCC2 levels are decreased in NudCL2 knockout(KO)***,Hsp90 forms a complex with NudCL2 to stabilize RCC2,which is essential for ***2 depletion mirrors phenotypes observed in NudCL2-downregulated ***,ectopic expression of RCC2 rescues the cytokinesis defects induced by NudCL2 deletion,but not vice ***,our data reveal the significance of the NudCL2/Hsp90/RCC2 pathway in cytokinesis at the midbody.

白介素15受体α链(IL-15 Rα)是介导IL-15信号转导的高亲和力膜受体复合物的重要亚基。通过RACE技术克隆得到松江鲈(Trachidermus fasciatus Heckel IL-15 Rα)基因cDNA全长序列(命名为TfIL-15Rα),TfIL-15Rα全长为998 bp,包括5′-非编码区(5′-UTR)85 bp,开放阅读框(ORF)663 bp和3′UTR 250 bp。基因ORF编码220个氨基酸(aa),前27 aa为信号肽序列。同源比对发现,松江鲈TfIL-15Rα与其他鱼类的序列一致性为19%~46%,保守度较低。多序列比对结果显示,IL-15Rα保守序列主要分布于sushi结构域(31~95 aa),4个高度保守的半胱氨酸形成的两对二硫键,是结构域发挥配体结合功能的重要位点。qRT-PCR分析表明,TfIL-15Rα广泛表达于松江鲈各组织中。腹腔注射LPS后,在皮肤、血液和肝脏中,刺激后2 h,IL-15Rα表达量迅速上调至最高峰,分别为对照组的14倍、52倍和21倍;而在脾脏中,刺激后12 h,TfIL-15RαmRNA达到最大表达量。由以上实验结果推测,TfIL-15Rα参与到了松江鲈抵抗外界刺激的先天免疫过程中。

Regulated cell death is a form of cell death that is actively controlled by *** studies have shown that regulated cell death plays a key role after spinal cord *** and ferroptosis are newly discovered types of regulated cell deaths that have been shown to exacerbate inflammation and lead to cell death in damaged spinal ***,a complex form of cell death that is interconnected with various regulated cell death mechanisms,has garnered significant attention in the study of spinal cord *** injury triggers not only cell death but also cellular survival *** signaling pathways play pivotal roles in influencing the processes of both deterioration and repair in spinal cord injury by regulating pyroptosis,ferroptosis,and ***,this review aims to comprehensively examine the mechanisms underlying regulated cell deaths,the signaling pathways that modulate these mechanisms,and the potential therapeutic targets for spinal cord *** analysis suggests that targeting the common regulatory signaling pathways of different regulated cell deaths could be a promising strategy to promote cell survival and enhance the repair of spinal cord ***,a holistic approach that incorporates multiple regulated cell deaths and their regulatory pathways presents a promising multi-target therapeutic strategy for the management of spinal cord injury.

Western North Pacific Tropical Cyclone Database Created by the China Meteorological Administration

This paper describes the access to, and the content, characteristics, and potential applications of the tropical cyclone(TC) database that is maintained and actively developed by the China Meteorological Administration, with the aim of facilitating its use in scientific research and operational services. This database records data relating to all TCs that have passed through the western North Pacific(WNP) and South China Sea(SCS) since 1949. TC data collection has expanded over recent decades via continuous TC monitoring using remote sensing and specialized field detection techniques,allowing collation of a multi-source TC database for the WNP and SCS that covers a long period, with wide coverage and many observational elements. This database now comprises a wide variety of information related to TCs, such as historical or real-time locations(i.e., best track and landfall), intensity, dynamic and thermal structures, wind strengths, precipitation amounts, and frequency. This database will support ongoing research into the processes and patterns associated with TC climatic activity and TC forecasting.

Prediction and stratification for the surgical adverse events after minimally invasive esophagectomy:A two-center retrospective study

BACKGROUND Minimally invasive esophagectomy(MIE)is a widely accepted treatment for esophageal cancer,yet it is associated with a significant risk of surgical adverse events(SAEs),which can compromise patient recovery and long-term *** preoperative identification of high-risk patients is critical for improving *** To establish and validate a risk prediction and stratification model for the risk of SAEs in patients with *** This retrospective study included 747 patients who underwent MIE at two centers from January 2019 to February *** were separated into a train set(n=549)and a validation set(n=198).After screening by least absolute shrinkage and selection operator regression,multivariate logistic regression analyzed clinical and intraoperative variables to identify independent risk factors for SAEs.A risk stratification model was constructed and validated to predict the probability of *** SAEs occurred in 10.2%of patients in train set and 13.6%in the validation *** with SAE had significantly higher complication rate and a longer hospital stay after *** key independent risk factors identified included chronic obstructive pulmonary disease,a history of alcohol consumption,low forced expiratory volume in the first second,and low albumin *** stratification model has excellent prediction accuracy,with an area under the curve of 0.889 for the training set and an area under the curve of 0.793 for the validation *** The developed risk stratification model effectively predicts the risk of SAEs in patients undergoing MIE,facilitating targeted preoperative interventions and improving perioperative management.

BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor *** its clinical severity,the genetic alterations,mechanisms,and signaling pathways underlying gallbladder NEC remain *** SUMMARY This case study presents a rare instance of primary gallbladder NEC in a 73-year-old female patient,who underwent a radical cholecystectomy with hepatic hilar lymphadenectomy and resection of liver segments IV-B and *** gene sequencing and bioinformatics analysis tools,including STRING,GeneMANIA,Metascape,TRRUST,Sangerbox,cBioPortal and GSCA,were used to analyze the biological functions and features of mutated genes in gallbladder *** mutations(APC,ARID2,IFNA6,KEAP1,RB1,SMAD4,TP53,BTK,GATA1,GNAS,and PRDM3)were identified,and the tumor mutation burden was determined to be 9.52 muts/Mb via targeted gene sequencing.A protein-protein interaction network showed significant interactions among the twelve mutated *** Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were used to assess mutation functions and *** results revealed 40 tumor-related pathways.A key regulatory factor for gallbladder NEC-related genes was identified,and its biological functions and features were compared with those of gallbladder *** Gallbladder NEC requires standardized *** with other gallbladder carcinomas revealed clinical phenotypes,molecular alterations,functional characteristics,and enriched pathways.
