Genomic analysis uncovers functional variation in the c-terminus of anthocyanin-activating MYB transcription factors

MYB transcription factors regulate diverse aspects of plant development and secondary metabolism,often by partnering in transcriptional regulatory ***,we harness genomic resources to identify novel MYBs,thereby producing an updated eudicot MYB phylogeny with revised relationships among subgroups as well as new information on sequence variation in the disordered c-terminus of anthocyanin-activating ***®and hidden Markov model scans of gene annotations identified a total of 714 MYB transcription factors across the genomes of four crops that span the eudicots:apple,grape,kiwifruit and *** model-based phylogenetic inference identified novel members of previously defined subgroups,and the function of specific anthocyanin-activating subgroup 6 members was assayed transiently in tobacco *** conservation within subgroup 6 highlighted one previously described and two novel short linear motifs in the disordered c-terminal *** novel motifs have a mix of hydrophobic and acidic residues and are predicted to be relatively ordered compared with flanking protein *** of motifs with the Eukaryotic Linear Motif database suggests roles in protein–protein *** of motifs and their flanking regions from strong anthocyanin activators into weak activators,and vice versa,affected *** conclude that,although the MYB c-terminal sequence diverges greatly even within MYB clades,variation within the c-terminus at and near relatively ordered regions offers opportunities for exploring MYB function and developing superior alleles for plant breeding.

The Kentucky Geological Survey(KGS)1 Hanson Aggregates stratigraphic research well,carter county,Kentucky,USA,was drilled to a total depth of 1474 m as a field-scale test of potential cO2 storage reservoir properties in the central Appalachian *** properties of the Rose Run sandstone(upper Ordovician Knox group)were tested for its suitability as a storage reservoir.A 9.8-m thick section of the Rose Run was penetrated at 1000 m drilled depth and a whole-diameter core and rotary sidewall cores were *** porosity and permeability measured in core plugs were 9.1%and 44.6 mD,*** vertical stress gradient calculated in the wellbore was 26 MPa/*** fractures in dolomites underlying and overlying the Rose Run follow the contemporary N53E Appalachian Basin stress *** Rose Run elastic geomechanical properties were calibrated to values measured in core plugs to evaluate its fracturing risk as a cO2 storage *** Young’s modulus and Poisson’s ratio values of the Rose Run were 45 GPa and 0.23,respectively,whereas Young’s modulus and Poisson’s ratio values were 77.1 GPa and 0.28,respectively,in the overlying Beekmantown dolomite,suggesting the Rose Run may fracture if overpressured during cO2 injection but be confined by the *** compressive strength measured in core plugs found the Rose Run and Beekmantown fractured at mean axial stresses of 156.5 MPa and 282.2 MPa,respectively,confirming the Beekmantown as suitable for confining cO2 injected into the Rose Run.A step-rate test was conducted in a mechanically-isolated 18.6-m interval bracketing the Rose *** Rose Run reservoir pressure was 9.3 MPa,and fracture gradient under injection was 13.6 MPa/km,suggesting step-rate testing before cO2 injection,and subsequent pressure monitoring to ensure *** the region around the KGS 1 Hanson Aggregates well is underpressured and adjacent to faulted Precambrian basement,further research is neede

Scalable Deposition Methods for Large-Area Production of Perovskite Thin Films

The recent steady improvements in the performance of the nascent hybrid perovskite photovoltaic(PV)devices have led to power conversion efficiencies that rival the best-performing established PV ***,to scale these laboratory demonstrations to PV module-scale production will require development of scalable deposition methods for perovskite thin *** record result for perovskite PVs so far was achieved via spin coating,a technique that is popular in research laboratories for thin-film coating over relatively small device areas,but not considered to be a method that could be used to scale up the manufacturing of perovskite *** larger thin-film areas are needed for future commercial PV ***,some researchers have focused their efforts on perovskite deposition techniques that can be considered as scalable for mass production and have achieved notable results even on large ***,we present an overview of the solution-based and vapor-based deposition processes;we explain their influence on the molecular crystal growth behavior of perovskite thin films and discuss the morphology as well as other material quality *** presenting a comprehensive comparison of the deposition techniques and the corresponding performance parameters for different device sizes,we intent to guide the growing research community through the methods that might enable mass production of perovskite solar products.

Functionalization of Single- and Multi-Walled carbon Nanotubes with cationic Amphiphiles for Plasmid DNA complexation and Transfection

The possibility of delivering DNA effi ciently to cells represents a crucial issue for the treatment of both genetic and acquired ***,even although the effi ciency of non-viral transfection systems has improved in the last decade,none have yet proven to be suffi ciently effective in *** report herein our results on the functionalization of single-walled carbon nanotubes(SWNT)and multi-walled carbon nanotubes(MWNT)by two cationic amphiphiles(lipid RPR120535 and pyrenyl polyamine),their use for the complexation of plasmid DNA,and their efficiency in transfecting cells in *** experiments have shown that the efficiency of transfection is higher when using SWNT instead of MWNT,and that transfection effi ciency is similar or slightly higher when using nanoplexes(SWNT/lipid RPR120535/DNA)instead of lipoplexes(lipid RPR120535/DNA)and several orders of magnitude higher than that of naked *** study therefore shows both that the transfection is better when using SWNTs and that it is dependent on the nature of the amphiphilic molecules adsorbed on the nanotubes.

Infrared chemical imaging through non-degenerate two-photon absorption in silicon-based cameras

chemical imaging based on mid-infrared(MIR)spectroscopic contrast is an important technique with a myriad of applications,including biomedical imaging and environmental *** MIR cameras,however,lack performance and are much less affordable than mature Si-based devices,which operate in the visible and nearinfrared ***,we demonstrate fast MIR chemical imaging through non-degenerate two-photon absorption(NTA)in a standard Si-based charge-coupled device(ccD).We show that wide-field MIR images can be obtained at 100 ms exposure times using picosecond pulse energies of only a few femtojoules per pixel through NTA directly on the ccD *** this on-chip approach does not rely on phase matching,it is alignment-free and does not necessitate complex postprocessing of the *** emphasize the utility of this technique through chemically selective MIR imaging of polymers and biological samples,including MIR videos of moving targets,physical processes and live nematodes.

Temporal and spatial study of differently charged ions emitted by ns-laser-produced tungsten plasmas using time-of-flight mass spectroscopy

Tungsten(W)is an important material in tokamak walls and *** W ion charge state distribution and the dynamic behavior of ions play important roles in the investigation of plasma–wall interactions using laser-ablation-based diagnostics such as laser-induced breakdown spectroscopy and laser-induced ablation *** this work,we investigate the temporal and spatial evolutions of differently charged ions in a nanosecond-laser-produced W plasma in vacuum using time-of-flight mass *** with different charge states from 1 to 7(W+to W7+)are all *** temporal evolutions of the differently charged ions show that ions with higher charge states have higher velocities,indicating that space separation occurs between the differently charged ion ***-resolved mass spectroscopy measurements further demonstrate the separation *** temporal profile can be accurately fitted by a shifted Maxwell–Boltzmann distribution,and the velocities of the differently charged ions are also obtained from the *** is found that the ion velocities increase continuously from the measured position of 0.75 cm to 2.25 cm away from the target surface,which indicates that the acceleration process lasts through the period of plasma *** acceleration and space separation of the differently charged ions confirm that there is a dynamic plasma sheath in the laser-produced plasma,which provides essential information for the theoretical laser-ablation model with plasma formation and expansion.
