Reversible protein phosphorylation is a central regulatory mechanism of cell function. Deregulation of the balanced actions of protein kinases and phosphatases has been frequently associated with several pathological conditions, including cancer. Many studies have already addressed the role of protein kinases misregulation in cancer. However, much less is known about protein phosphatases influence. Phosphoprotein Phosphatase 1 (PPP1) is one of the major serine/threonine protein phosphatases who has three catalytic isoforms: PPP1CA, PPP1CB, and PPP1CC. Its function is achieved by binding to regulatory subunits, known as PPP1-interacting proteins (PIPs), which may prefer a catalytic isoform. Also, some inhibitors/enhancers may exhibit isoform specificity. Here we show that, prodigiosin (PG), a molecule with anticancer properties, promotes the formation of PPP1CA-AKT complex and not of PPP1CC-MAPK complex. Both, AKT and MAPK, are well-known PIPs from two pathways that crosstalk and regulate melanoma cells survival. In addition, the analysis performed using surface plasmon resonance (SPR) technology indicates that PPP1 interacts with obatoclax (OBX), a drug that belongs to the same family of PG. Overall, these results suggest that PG might, at least in part, act through PPP1C/PIPs. Also, this study is pioneer in demonstrating PPP1 isoform-specific modulation by small molecules.

Zeta potential measurement on lithium lanthanum titanate nanoceramics

The zeta potential, isoelectric point, and agglomeration of ***3 （LLTO） nanoparticles dispersed in aqueous media at different ionic strengths have been studied. The zeta potential was determined from electrophoretic mobility measurements, according to Smoluchowski＇s equation, for Li0.5La0.5TiO3 suspen- sions in NaCl and KCI electrolytes with ionic strengths of 1, 10, and 100 mmol/dm3. The isoelectric point （IEP）, zeta potential （ζ）, and the agglomeration were shown to strongly depend on the ionic strength of the Li0.5La0.5TiO3 aqueous colloidal suspension in both NaCI and KCI electrolytes, which allows the deter- mination of the effects of environmental conditions for ***3 manipulation in aqueous colloidal systems. The suspensions of Li0.5La0.5TiO3 nanoparticles reach the IEP in the pH range 0f3-5. The （ of Li0.5La0.5TiO3 nanoparticles varied from positive to negative values with a pH increase, which allows for the control of the surface charge depending on the purpose. The pH range of 7-g and an ionic strength ≤1 mmol/dm3 are recommended as the most suitable conditions for both the LLTO colloidal shaping techniques application and the LLTO-based nanocomposite formation.

Let （Ω, ∑） be a measurable space and mo ： E→ Xo and m1 ： E → X1 be positive vector measures with values in the Banach KSthe function spaces Xo and X1. If 0 〈 a 〈 1, we define a X01-ax1a new vector measure [m0, m]a with values in the Calderdn lattice interpolation space and we analyze the space of integrable functions with respect to measure [m0, m1]a in order to prove suitable extensions of the classical Stein Weiss formulas that hold for the complex interpolation of LP-spaces. Since each p-convex order continuous Kothe function space with weak order unit can be represented as a space of p-integrable functions with respect to a vector measure, we provide in this way a technique to obtain representations of the corresponding complex interpolation spaces. As applications, we provide a Riesz-Thorin theorem for spaces of p-integrable functions with respect to vector measures and a formula for representing the interpolation of the injective tensor product of such spaces.

This paper presents an upper limb exoskeleton that allows cognitive(through electromyography signals)and physical user interaction(through load cells sensors)for passive and active exercises that can activate neuroplasticity in the rehabilitation process of people who suffer from a neurological *** the exoskeleton to be easily accepted by patients who suffer from a neurological injury,we used the ISO9241-210:2010 as a methodology design *** the first steps of the design process,design requirements were collected from previous usability tests and ***,as a second step,a technological solution is proposed,and as a third step,the system was evaluated through performance and user *** part of the technological solution and to allow patient participation during the rehabilitation process,we have proposed a hybrid admittance control whose input is load cell or electromyography *** hybrid admittance control is intended for active therapy exercises,is easily implemented,and does not need musculoskeletal modeling to ***,electromyography signals classification models and features were evaluated to identify the best settings for the cognitive human–robot interaction.

Fatty acid oxidation and autophagy promote endoxifen resistance and counter the effect of AKT inhibition in ER-positive breast cancer cells

Tamoxifen(TAM)is the first-line endocrine therapy for estrogen receptor-positive(ER+)breast cancer(BC).However,acquired resistance occurs in∼50%***,although the PI3K/AKT/mTOR pathway is a viable target for treatment of endocrine therapy-refractory patients,complex signaling feedback loops exist,which can counter the effectiveness of inhibitors of this ***,we analyzed signaling pathways and metabolism in ER+MCF7 BC cell line and their TAM-resistant derivatives that are co-resistant to endoxifen using immunoblotting,quantitative polymerase chain reaction,and the Agilent Seahorse XF *** found that activation of AKT and the energy-sensing kinase AMPK was increased in TAM and endoxifen-resistant ***,ERRα/PGC-1βand their target genes MCAD and CPT-1 were increased and regulated by AMPK,which coincided with increased fatty acid oxidation(FAO)and autophagy in TAM-resistant *** of AKT feedback-activates AMPK and ERRα/PGC-1β-MCAD/CPT-1 with a consequent increase in FAO and autophagy that counters the therapeutic effect of endoxifen and AKT ***,our results indicate increased activation of AKT and AMPK with metabolic reprogramming and increased autophagy in TAM-resistant *** inhibition of AKT and FAO/autophagy is necessary to fully sensitize resistant cells to endoxifen.

p53 promotes AKT and SP1-dependent metabolism through the pentose phosphate pathway that inhibits apoptosis in response to Nutlin-3a

Nutlin-3a is a MDM2 antagonist and preclinical drug that activates p53. Cells with MDM2 gene amplification are especially prone to Nutlin-3a-induced apoptosis, though the basis for this is unclear. Glucose metabolism can inhibit apoptosis in response to Nutlin-3a through mechanisms that are incompletely understood. Glucose metabolism through the pentose phosphate pathway （PPP） produces NADPH that can protect cells from potentially lethal reactive oxygen species （ROS）. We compared apoptosis and glucose metabolism in cancer cells with and without MDM2 gene amplification treated with Nutlin-3a. Apoptosis in MDM2-amplified cells was associated with a reduction in glycolysis and the PPP, reduced NADPH, increased ROS, and depletion of the transcription factor SP1, which normally promotes PPP gene expression. In contrast, glycolysis and the PPP were maintained or increased in MDM2 non-amplified cells treated with Nutlin-3a. This was dependent on p53-mediated AKT activation and was associated with maintenance of SP1 and continued expression of PPP genes. Knockdown or inhibition of AKT, SP1, or the PPP sensitized MDM2-non-amplified cells to apoptosis. The data indicate that p53 promotes AKT and SP1-dependent activation of the PPP that protects cells from Nutlin-3a-induced apoptosis. These findings provide insight into how glucose metabolism reduces Nutlin-3a-induced apoptosis, and also provide a mechanism for the heightened sensitivity of MDM2-amplified cells to apoptosis in response to Nutlin-3a.

Astrocytes and retrograde degeneration of nigrostriatal dopaminergic neurons in Parkinson’s disease:removing axonal debris

Objective:The dopaminergic nigrostriatal neurons(DA cells)in healthy people present a slow degeneration with aging,which produces cellular debris throughout *** 2%-5%of people present rapid cell degeneration of more than 50%of DA cells,which produces Parkinson's disease(PD).Neuroinflammation accelerates the cell degeneration and may be critical for the transition between the slow physiological and the rapid pathological degeneration of DA cells,particularly when it activates microglial cells of the medial forebrain bundle near dopaminergic *** synaptic debris produced by DA cell degeneration may trigger the parkinsonian neuroinflammation,this study investigated the removal of axonal debris produced by retrograde degeneration of DA cells,paying particular attention to the relative roles of astrocytes and ***:Rats and mice were injected in the lateral ventricles with 6-hydroxydopamine,inducing a degeneration of dopaminergic synapses in the striatum which was not accompanied by non-selective tissue damage,microgliosis or *** possible retrograde degeneration of dopaminergic axons,and the production and metabolization of DA-cell debris were studied with immunohistochemical methods and analyzed in confocal and electron microscopy ***:The selective degeneration of dopaminergic synapses in the striatum was followed by a retrograde degeneration of dopaminergic axons whose debris was found within spheroids of the medial forebrain *** spheroids retained mitochondria and most(e.g.,tyrosine hydroxylase,the dopamine transporter protein,and amyloid precursor protein)but not all(e.g.,α-synuclein)proteins of the degenerating dopaminergic *** showed initial(autophagosomes)but not late(lysosomes)components of autophagy(incomplete autophagy).These spheroids were penetrated by astrocytic processes of the medial forebrain bundle,which provided the lysosomes needed to continue the degradation of dopaminergic debri

Earth observation satellites produce large amounts of images/data that not only must be processed and preserved in reliable geospatial platforms but also efficiently disseminated among partners/researchers for creating derivative products through collaborative *** can face up this challenge in a cost-effective manner by using cloud ***,outages and violations of integrity/confidentiality associated to this technology could *** article presents FedIDS,a suite of cloud-based components for building dependable geospatial *** Fed component enables organizations to build shared geospatial data infrastructure through federation of independent cloud resources to withstand outages,whereas IDS avoids violations of integrity/confidentiality of images/data in sharing information and collaboration workflows.A FedIDS prototype,deployed in Spain and Mexico,was evaluated through a study case based on a satellite imagery captured by a Mexican antenna and another based on a satellite imagery of a European observation *** acquisition,storage and sharing of images among users of the federation,the exchange of images between Mexican and Spanish sites and outage scenarios were *** evaluation revealed the feasibility,reliability and efficiency of FedIDS,in comparison with available solutions,in terms of performance,storage consume and integrity/confidentiality when sharing images/data in collaborative scenarios.

A Dynamic Model of Degradation of Air Systems in Conflict

In the evaluation of the necessary measurement of aeronautical means to face to a certain military operative problem, the development of an appropriate tool is of importance. Conventional techniques are considered inadequate in the analysis of military operative problems in presence of a self-attrition behavior from one of the involved parts. In this paper a traditional algorithm for measuring military power is implemented and analyzed and then we improve this traditional algorithm on the basis of the kind of degradation that systematically takes place in self-destructive systems. In terms of traditional analysis, the evolution of a war is essentially an unreal sequence of repetitive cycles. So a time-dependent function was introduced in improved the algorithm. The development of this tool of prediction has the aim to argue the management of taking decisions in this type of crisis and complements itself with historical arguments or references of rigor. A computer software show results that allows to estimate the costs of the support and to visualize the graphs associated with the degradation of the system and his temporary evolution. The improved algorithm is more suitable than the traditional one in software simulations.
