Characterization and genetic manipulation of primed stem cells into a functional na?ve state with ESRRB

AIm To identify differences between primed mouse embryonic stem cells(ESCs) and fully functional naive ESCs; to manipulate primed cells into a naive state. mETHODS We have cultured 3 lines of cells from different mouse strains that have been shown to be naive or primed as determined by generating germline-transmitting *** were put through a battery of tests to measure the different features. RNA from cells was analyzed using microarrays, to determine a priority list of the differentially expressed genes. These were later validated by quantificational real-time polymerase chain reaction. Viral cassettes were created to induce expression of differentially expressed genes in the primed cells through lentiviral transduction. Primed reprogrammed cells were subjected to in-vivo incorporation *** most results show that both primed and naive cells have similar features(morphology, proliferation rates, stem cell genes expressed). However, there were some genes that were differentially expressed in the na?ve cells relative to the primed cells. Key upregulated genes in na?ve cells include ESRRB, ERAS, ATRX, RNF17, KLF-5, and mYC. After over-expressing some of these genes the primed cells were able to incorporate into embryos in-vivo, re-acquiring a feature previously absent in these cells. CONCLUSION Although there are no notable phenotypic differences, there are key differences in gene expression between these na?ve and primed stem cells. These differences can be overcome through overexpression.

A VSC-based model for Power Flow Assessment of multi-terminal VSC-HVDC Transmission Systems

This paper puts forward a new practical voltage source converter(VSC)based AC-DC converter model suitable for conducting power flow assessment of multi-terminal VSCbased high-voltage direct current(VSC-mTDC)*** model uses an advanced method to handle the operational limits and control modes of VSCs into the power flow *** new model is incorporated into a unified framework encompassing AC and DC power grids and is solved by using the Newton-Raphson method to enable quadratically convergent iterative *** use of complementarity constraints,together with the Fischer-Burmeister function,is proposed to enable the seamless incorporation of operational control modes of VSC and automatic enforcement of any converter’s operational limits that become violated during the iterative solution ***,a dedicated process for checking limits is no longer ***,all existing relationships between the VSC control laws and their operational limits are considered directly during the solution of the power flow *** applicability of the new model is demonstrated with numerical examples using various multi-terminal AC-DC transmission networks,one of which is a utility-sized power system.

Historical relationships of areas of endemism of the Brazilian Atlantic rain forest： a cladistic biogeographic analysis of harvestman taxa （Arachnida： Opiliones）

Based on a cladistic biogeographic analysis of 6 species-level phylogenies of harvestman taxa, wesearched for congruence in the historical relationships of 12 areas of endemism of the BrazilianAtlantic Rain Forest. We constructed general area cladograms using Primary Brooks ParsimonyAnaJysis （BPA）, BPA of nodes, and paralogy-free subtree analysis. These analyses resulted in 6general area cladograms, that allow to infer a general pattern of the relationships among areas ofendemism from the Brazilian Atlantic Rain Forest. Northern areas resulted related basally showingmain disjunctions at the Doce River Valley and Todos os Santos Bay/S^o Francisco River *** remaining areas of endemism were included in a southern and a southeastern block, sepa-rated by the Ribeira do Iguape Valley. Incongruence Length Differences tests showed no significantincongruence among the resulting cladograms and other matrix partitions. We concluded that tec-tonism and ancient marine transgressions were the probable processes responsible for the maindisjunctions, whereas Neogene refugia seem to have caused the more recent disjunctions. Thegeneral pattern and redundancy in area relationships suggest a model of main reiterative barriersin diversification at multiple times for the evolution of the Atlantic Rain Forest. The renewal of cla-distic biogeography and the search for common biogeographic patterns are discussed.

Localized micronutrient patches induce lateral root foraging and chemotropism in Nicotiana attenuata

Nutrients are distributed unevenly in the *** plasticity in root growth and proliferation may enable plants to cope with this variation and effectively forage for essential nutrients. However, how micronutrients shape root architecture of plants in their natural environments is poorly understood. We used a combination of field and laboratory-based assays to determine the capacity of Nicotiana attenuata to direct root growth towards localized nutrient patches in its native environment. Plants growing in nature displayed a particular root phenotype consisting of a single primary root and a few long, shallow lateral roots. Analysis of bulk soil surrounding the lateral roots revealed a strong positive correlation between lateral root placement and micronutrient gradients, including copper, iron and zinc. In laboratory assays, the application of localized micronutrient salts close to lateral root tips led to roots bending in the direction of copper and iron. This form of chemotropism was absent in ethylene and jasmonic acid deficient lines,suggesting that it is controlled in part by these two hormones. This work demonstrates that directed root growth underlies foraging behavior, and suggests that chemotropism and micronutrient-guided root placement are important factors that shape root architecture in nature.

How do greenhouse gas emissions vary with biofertilizer type and soil temperature and moisture in a tropical grassland?

Greenhouse gases are known to play an important role in global *** this study,we determined the effects of selected soil and climate variables on nitrous oxide(N2O),methane(CH4),and carbon dioxide(CO2)emissions from a tropical grassland fertilized with chicken slurry,swine slurry,cattle slurry,and cattle *** N2O emissions did not differ between treatments and varied from 29.26 to 32.85 mg N m^-***,cumulative CH4 emissions were not significantly different among the treatments and ranged from 6.34 to 57.73 mg CH4 m^-*** and compost application induced CO2 emissions that were significantly different from those in the control *** CH4 conversion factors measured were 0.21%,1.39%,4.39%,and 5.07%for cattle compost,chicken slurry,swine slurry,and cattle slurry,respectively,differing from the recommendations of the Intergovernmental Panel on Climate Change(IPCC).The fraction of added Nemitted as N2O was 0.39%,which was lower than the IPCC default value of 2%.Our findings suggest that N2O emissions could be mitigated by replacing synthetic fertilizer sources with either biofertilizer or *** results indicate the following:N2O emission was mainly controlled by soil temperature,followed by soil moisture and then soil NH^"content;CH4 fluxes were mainly controlled by soil moisture and chamber headspace temperature;and CO2 fluxes were mainly controlled by chamber headspace temperature and soil moisture.

In this study,we identiﬁed the moisture sources for the precipitation associated with tropical cyclones(TCs)during the rapid intensiﬁcation(RI)process from 1980 to 2018 by applying a Lagrangian moisture source diagnostic *** detected sixteen regions on a global scale for RI events distributed as follows:four in the North Atlantic(NATL),two in the Central and East Paciﬁc Ocean(NEPAC),the North Indian Ocean(NIO)and South Indian Ocean(SIO),and three in the South Paciﬁc Ocean(SPO)and the Western North Paciﬁc Ocean(WNP).The moisture uptake(mU)mostly was from the regions where TCs underwent *** Western NATL,tropical NATL,Caribbean Sea,the Gulf of mexico and the Central America and mexico landmass supported~85.4%of the precipitating moisture in the NATL,while the latter source and the eastern North Paciﬁc Ocean provided the higher amount of moisture in NEPAC(~84.3%).The Arabian Sea,the Bay of Bengal and the Indian Peninsula were the major moisture sources in NIO,contributing approximately 81.3%.The eastern and western parts of the Indian Ocean supplied most of the atmospheric humidity in SIO(~83.8%).The combined contributions(~87.9%)from the western and central SPO and the Coral Sea were notably higher in ***,TCs in the WNP basin mostly received moisture from the western North Paciﬁc Ocean,the Philippine Sea and the China Sea,accounting for 80.1%.The remaining moisture support in each basin came from the summed contributions of the remote ***,RI TCs gained more moisture up to 2500 km from the cyclone centre than those slow intensiﬁcation(SI)and the total mU was approximately three times higher during RI than ***,the patterns of the mU differences respond to the typical pathways of moisture transport in each basin.

Kinetic analysis via mathematical modeling for ferrous iron oxidation in a class of SBR-type system

This paper analyses the oxidation of ferrous iron via chemical and biological means in a class of Sequential Batch Reactors(SBR-type).For this,a kinetic model for the study of iron oxidation system is proposed,followed by a parametric sensitivity analysis and a bifurcation analysis,which allow selecting the most influential kinetic parameters in order to ensure a suitable prediction capacity of the mathematical *** system consists of two SBR bioreactors,the first being used to produce hydrogen peroxide(H2O2)that is fed to a second reactor where the iron oxidation is carried out by chemical–biological *** predictions were compared with experimental data for the production of H2O2 and for ferrous iron oxidation,finding suitable correlation coefficients(r2 N 0.98)for each state *** bifurcation analysis showed the trajectories of the main variables,such as,biomass,H2O2 and ferrous iron,under the change of the most influential kinetic *** analysis demonstrates the usefulness of the constructed model to predict the kinetic behaviour of the SBR-type process.

If 0 〈 a 〈 1, we define a X01-ax1a new vector measure [m0, m]a with values in the Calderdn lattice interpolation space and we analyze the space of integrable functions with respect to measure [m0, m1]a in order to prove suitable extensions of the classical Stein Weiss formulas that hold for the complex interpolation of LP-spaces. Since each p-convex order continuous Kothe function space with weak order unit can be represented as a space of p-integrable functions with respect to a vector measure, we provide in this way a technique to obtain representations of the corresponding complex interpolation spaces. As applications, we provide a Riesz-Thorin theorem for spaces of p-integrable functions with respect to vector measures and a formula for representing the interpolation of the injective tensor product of such spaces.
