The presenilin genes(PSEN1 and PSEN2)are mainly responsible for causing early-onset familial Alzheimer’s disease,harboring~300 causative mutations,and representing~90%of all mutations associated with a very aggressive disease *** 1 is the catalytic core of theγ-secretase complex that conducts the intramembranous proteolytic excision of multiple transmembrane proteins like the amyloid precursor protein,Notch-1,N-and E-cadherin,LRP,Syndecan,Delta,Jagged,CD44,ErbB4,and *** 1 plays an essential role in neural progenitor maintenance,neurogenesis,neurite outgrowth,synaptic function,neuronal function,myelination,and ***,an imbalance caused by mutations in presenilin 1/γ-secretase might cause aberrant signaling,synaptic dysfunction,memory impairment,and increased Aβ42/Aβ40 ratio,contributing to neurodegeneration during the initial stages of Alzheimer’s disease *** review focuses on the neuronal differentiation dysregulation mediated by PSEN1 mutations in Alzheimer’s ***,we emphasize the importance of Alzheimer’s disease-induced pluripotent stem cells models in analyzing PSEN1 mutations implication over the early stages of the Alzheimer’s disease pathogenesis throughout neuronal differentiation impairment.

Bioinformatic Analysis of Potential Pathogenicity Effectors of <i>Candidatus</i>Liberibacter asiaticus, Causal Agent of Citrus Huanglongbing

Huanglongbing (HLB) or citrus greening is currently the most important citrus disease, caused by the bacterium Candidatus Liberibacter asiaticus (CLas). The impossibility of isolating it causes understanding its pathogenic mechanisms to be a complicated task. Recent studies identified 16 proteins with the signal peptide needed to be secreted in the plant and cause the disease. The present study aims to perform a bioinformatic analysis of these proteins with the function prediction approach by gene ontology (GO) and the detection of conserved domains. It was observed that of the 16 proteins analyzed not all are found in different infective strains reported in the literature. The GO analysis allowed us to relate different proteins with the biological process of energy and pathogenic activity, especially CLIBASIA_03315 and CLIBASIA_05115, respectively. The domain analysis allowed the observation of a β-CA domain, tentatively related to the damage caused to the chloroplast and a PAAR domain associated with the T6SS secretory system. Our results provide information on the possible function of potential pathogenicity effectors in Cont-family:Verdana;">Las.

Effect of Mushroom Extract Hericium erinaceus on Spatial Memory and Morphology of Neurons in the CA1 and CA3 Regions of the Hippocampus in ovariectomized Rats

Various studies indicate that low levels of estradiol negatively impact cognitive abilities. Extracts from the fungus Hericium erinaceus (HE) contain bioactive components that promote the proper functioning of the nervous system and potential effects on protection against neurodegenerative diseases, including dementia and motor dysfunctions. The objective was to evaluate the effects of the administration of the HE mushroom extract on visuospatial memory and morphology of neurons in the CA1 and CA3 regions of the hippocampus in ovariectomized rats. 40 young Wistar rats weighing 90 ± 10 g BW were used, which were distributed into four groups of 10 animals;Control Group, non-ovariectomized and untreated rats;Group E2, ovariectomized rats treated with estradiol (2 μg/kg/body weight);Group HE, ovariectomized rats treated with the extract of the fungus Hericium erinaceus (0.5 mg/kg body weight) and Group ovx/ST, ovariectomized rats, without treatment. The animals were tested in the Barnes and open Field maze, then they were sacrificed, and their brains were obtained to perform a histological analysis of neuronal morphology in the CA1 and CA3 areas of the hippocampus. The most outstanding results showed that the ovx/ST group recorded the longest time to arrive at the escape box and stay in the Barnes maze. A correlation was observed between neuronal damage and function;in the groups that did not present satisfactory performance in the maze tests, morphological alterations were identified such as the presence of some neuronal somata with degeneration characteristics such as pyknosis, nuclear basophilia and shrinkage of the cells. Its soma, as well as a decrease in the nuclear area of CA1 and CA3 neurons. It is concluded that the fungus Hericium erinaceus exerted a neuroprotective effect on the neuronal bodies of the hippocampus, associated with better performance in the visuospatial recognition memory test.

Deduction and Validation of an Eulerian-Eulerian Model for Turbulent Dilute Two-Phase Flows by Means of the Phase Indicator Function-Disperse Elements Probability Density Function

A statistical formalism overcoming some conceptual and practical difficulties arising in existing two-phase flow (2PHF) mathematical modelling has been applied to propose a model for dilute 2PHF turbulent Hows. Phase interaction terms with a clear physical meaning enter the equations and the formalism provides some guidelines for the avoidance of closure assumptions or the rational approximation of these terms. Continuous phase averaged continuity, momentum, turbulent kinetic energy and turbulence dissipation rate equations have been rigorously and systematically obtained in a single step. These equations display a structure similar to that for single-phase flows. It is also assumed that dispersed phase dynamics is well described by a probability density function (pdf) equation and Eulerian continuity, momentum and fluctuating kinetic energy equations for the dispersed phase are deduced. An extension of the standard k-e turbulence model for the continuous phase is used. A gradient transport model is adopted for the dispersed phase fluctuating fluxes of momentum and kinetic energy at the non-colliding, large inertia limit. This model is then used to predict the behaviour of three axisymmetric turbulent jets of air laden with solid particles varying in size and concentration. Qualitative and quantitative numerical predictions compare reasonably well with the three different sets of experimental results, studying the influence of particle size, loading ratio and flow confinement velocity.

Adipokines,cytokines and body fat stores in hepatitis Cvirus liver steatosis

AIM: To identify patients with or without liver steatosis and its severity in treatment-na?ve patients affected by hepatitis C virus(HCV) ***: We included 56 HCV infected patients, and assessed the amount of liver fat by histomorphometry, and its relationships with fat and lean mass at different parts of the body(by densitometry), hormones [insulin, homeostatic model assessment(HoMA)], adipokines(resistin, adiponectin, leptin), and cytokines(tumor necrosis factor α, interleukin-6).RESULTS: Although the intensity of liver steatosis is related to trunk fat mass and HoMA, 33% of patients showed no liver steatosis, and this finding was not related to body mass index or genotype. Besides trunkfat mass, no other factor was related to the presence or not of liver steatosis, or to the intensity of it, by multivariate analysis. Lean mass was not related to liver steatosis. Adiponectin levels were lower among patients. No differences were observed in leptin and ***: Steatosis in HCV infection is common(67.2%), and closely related to trunk fat, and insulin resistance, but not with leg fat mass or adipokines.

Metal alginates for polyphenol delivery systems: Studies on crosslinking ions and easy-to-use patches for release of protective flavonoids in skin

Near Surface Carbon Dioxide and Methane in Urban Areas of Costa Rica

Little information is available for Central America regarding methane and carbon dioxide mixing ratios in urban areas. This work reports a representative spatial and seasonal study of near surface carbon dioxide and methane, carried out between July 2014 and January 2015 (27 weeks) in the Central Valley of Costa Rica, and other urban and rural sites across the country and covering three distinct seasons: Mid-summer drought (July-August), wet season (September-November) and transition period (December-January). The mixing ratios of both gases are clearly influenced by the metropolitan area, and by the prevailing atmospheric conditions during the wet season months. Average carbon dioxide concentration (629 ± 80 ppm) and average methane concentration (2192 ± 110 ppb) were up to 8% and up to 10%, respectively, higher during the wet season than the values recorded outside this period. HYSPLIT back air mass trajectories analysis, and weather data available for the Central Valley, suggest that these differences arise as result of a reduction in the mixing layer of depth (~425 m) and the wind speed (~1.5 m/s) across the valley, favoring the accumulation of polluted air masses in the metropolitan area. other natural and anthropogenic sources, like the volcanic emissions of the Turrialba Volcano and the livestock activities at rural sites, apparently influence the mixing ratios of both gases across Costa Rica. Although the scope of this study is limited to representative seasonal conditions of the Central Valley in 2014 and 2015, it is possible considering the information presented in this work that the “dome” phenomenon can be assumed to exist.

Cytotoxic Effect of Methanol Extracts and Partitions of Two Mexican Desert Plants against the Murine Lymphoma L5178Y-R

Pachycereus marginatus (DC.) Britton & Rose and Ibervillea sonorae (S. Watson) Greene have been used in the Mexican traditional medicine for the treatment of various diseases, including cancer. The present study aims to investigate the cytotoxic activity of these plants against a murine lymphoma. Soxhlet extraction of dried and powdered plant material was performed with methanol. Also, a further partitioning of these methanolic extracts with hexane and ethyl acetate was achieved. The in vitro cytotoxic activity against the murine lymphoma L5178Y-R cell line was assessed via the colorimetric MTT assay. The methanol extract from P. marginatus exhibited high cytotoxic activity (up to 94%) at concentrations ranging from 3.9 to 500 μg/mL;however, hexane and ethyl acetate partitions from this methanolic extract showed lower but significant (p oncentration-dependent cytotoxicity (hexane partition up to 94% at 500 μg/mL;ethyl acetate partition up to 94% at 65.5 μg/mL). The methanolic extract and partitions derived from I. sonorae also showed significant (p oncentration-dependent cytotoxicity against L5178Y-R cells at concentrations ranging from 7.81 to 500 μg/mL (methanolic extract up to 63% at 500 μg/mL;hexane partition up to 76% at 250 μg/mL;ethyl acetate partition up to 73% at 500 μg/mL). These results demonstrate that the methanol extracts and partitions from P. marginatus and I. sonorae possess significant cytotoxic activity against the murine lymphoma L5178Y-R and validate the ethnobotanical use of these plants for the treatment of diseases consistent with cancer symptomatology. Previous scientific reports describe the isolation of isoquinoline alkaloids of P. marginatus as well as cucurbitacins from I. sonorae, phytochemicals that could be responsible for their observed cytotoxic activity in this research.
