Therapy with bone marrow cells reduces liver alterations in mice chronically infected by Schistosoma mansoni

AIM： To investigate the potential of bone marrow mononuclear cells （BM-MCs） in the regeneration of hepatic lesions induced by Schistosoma mansoni （***） chronic infection. METHODS： Female mice chronically infected with *** were treated with BM-MCs obtained from male green fluorescent protein （GFP） transgenic mice by intravenous or intralobular injections. Control mice received injections of saline in similar conditions, Enzyme-linked immunosorbent assay （ELISA） assay for transforming growth factor-beta （TGF-β）, polymerase chain reaction （PCR） for GFP DNA, immunofluorescence and morphometric studies were performed. RESULTS： Transplanted GFP^＋ cells migrated to granuloma areas and reduced the percentage of liver fibrosis. The presence of donor-derived cells was confirmed by Fluorescence in situ hybridization （FISH） analysis for detection of cells bearing Y chromosome and by PCR analysis for detection of GFP DNA. The levels of TGF-β, a cytokine associated with fibrosis deposition, in liver fragments of mice submitted to therapy were reduced. The number of oval cells in liver sections of ***-infected mice increased 3-4 fold after transplantation. A partial recovery in albumin expression, which is decreased upon infection with ***, was found in livers of infected mice after cellular therapy. CONCLUSION： In conclusion, transplanted BMCs migrate to and reduce the damage of chronic fibrotic liver lesions caused by ***.

Feasibility and safety of autologous bone marrow mononuclear cell transplantation in patients with advanced chronic liver disease

AIM： To evaluate the safety and feasibility of bone marrow cell （BMC） transplantation in patients with chronic liver disease on the waiting list for liver transplantation. METHODS： Ten patients （eight males） with chronic liver disease were enrolled to receive infusion of autologous bone marrow-derived cells. Seven patients were classified as Child-Pugh B and three as Child-Pugh C. Baseline assessment included complete clinical and laboratory evaluation and abdominal MRI. Approximately 50 mL of bone marrow aspirate was prepared by centrifugation in a ficoll-hypaque gradient. At least of 100 millions of mononuclear-enriched BMCs were infused into the hepatic artery using the routine technique for arterial chemoembolization for liver tumors. Patients were followed up for adverse events up to 4 mo. RESULTS： The median age of the patients was 52 years （range 24-70 years）. All patients were discharged 48 h after BMC infusion. Two patients complained ofmild pain at the bone marrow needle puncture site. No other complications or specific side effects related to the procedure were observed. Bilirubin levels were lower at 1 （2.19 ± 0.9） and 4 mo （2.10 ± 1.0） after cell transplantation that baseline levels （238 ± 1.2）. Albumin levels 4 mo after BMC infusion （3.73 ± 0.5） were higher than baseline levels （3.47 ± 0.5）. International normalized ratio （INR） decreased from 1.48 （SD = 0.23） to 1.43 （SD = 0.23） one month after cell transplantation. CONCLUSION： BMC infusion into hepatic artery of patients with advanced chronic liver disease is safe and feasible. In addition, a decrease in mean serum bilirubin and INR levels and an increase in albumin levels are observed. Our data warrant further studies in order to evaluate the effect of BMC transplantation in patients with advanced chronic liver disease.

Variations in Atmospheric Electromagnetism and Their Impacts on Mortality Rates in the Southern and Northeastern Regions of Brazil

This study examines the relationship between geomagnetic indices and mortality rates from specific diseases in the Northeast (NE) and Southern (S) regions of Brazil from 1996 to 2020. Solar activity data, including the Ap and Sudden Ionospheric Disturbance (SID) indices, were sourced from the World Data Center, while mortality data were obtained from the Mortality Information System (SIM-DATASUS). Acute Myocardial Infarction (AMI) emerged as the leading cause of mortality, with average death rates of 60.4, 56.8, and 58.3 deaths per 100,000 inhabitants in Pernambuco (PE), Rio Grande do Sul (RS), and Paraná (PR), respectively. Temporal analysis revealed a consistent upward trend in AMI mortality across most states, except for Santa Catarina (SC). Seasonal patterns identified through Principal Component Analysis (PCA) demonstrated that geomagnetic and climatic indices influenced mortality differently across regions and seasons. The Ap geomagnetic index was strongly correlated with higher AMI mortality rates during summer, while the SID index showed greater relevance during winter. A dipole phenomenon was observed, with AMI deaths increasing alongside geomagnetic activity in the S region but showing an inverse relationship in the NE region. These findings highlight the significant influence of geomagnetic variations on public health, particularly cardiovascular mortality. The study underscores the need for further research into the biological mechanisms underlying these associations and recommends the development of early warning systems and targeted preventive measures to mitigate the potential health impacts of geomagnetic disturbances, especially for vulnerable populations.

Thermal Behavior of Sr（ll） and Ba（ll）-Diclofenac Complexes in Solid State： Study of the Dehydration and Thermal Decomposition

Solid state compounds of general formula Sr（Diclof）2 ·5.7H2O and Ba（Diclof）2·4.8H2O were obtained. Thermo- gravimetry （TG）, differential scanning calorimetry （DSC）, X-ray diffraction powder patterns and microscopy analysis were used to characterize these compounds. Details concerning the dehydration and thermal decomposition as well as data of kinetic parameters have been described here. The kinetic behaviors of these stages were evaluated from several heating rates with samples masses of 2 and 5 mg in open crucibles under a nitrogen atmosphere. The results of the present study improve the knowledge on these compounds including their dehydration and thermal stability. The obtained data depend on the mass of the sample which results in two kinetic behavior patterns.

Glyphosate is a herbicide that has been widely used worldwide and is used in agricultural areas to control weeds and unwanted *** sensors developed from different nanomaterials have high efficiency,excellent cost-benefit,and fast analysis time for detecting traces of environmental *** study aimed to produce an electrochemical sensor with disposable screen-printed electrodes based on carbon black modified with niobium nanoparticles to determine glyphosate in aqueous *** morphology,structure and electrochemical performance of the sensor were characterized by scanning electron microscopy,energy-dispersive X-ray spectroscopy,X-ray diffraction and cyclic *** pulse voltammetry in BR buffer solution at pH 5.0 allowed the generation of a method to quantify glyphosate concentration in a linear range of 5.90-172.30μmol/L(1.00-29.13μg/mL),with a limit of detection calculated at 3.07μmol/L(0.52μg/mL).The method efficiently quantified glyphosate in real water samples and showed no interference from K^(+),Na^(+),Ca^(2+),Mg^(2+)ions or thiamethoxam,imidacloprid and carbendazim pesticides.

Analysis of 12 variants in the development of gastric and colorectal cancers

AIM To evaluate the relation between 12 polymorphisms and the development of gastric cancer(GC) and colorectal cancer(CRC).METHODS In this study,we included 125 individuals with GC diagnosis,66 individuals with CRC diagnosis and 475 cancer-free individuals. All participants resided in the North region of Brazil and authorized the use of their samples. The 12 polymorphisms(in CASP8,CYP2 E1,CYP19 A1,IL1 A,IL4,MDM2,NFKB1,PAR1,TP53,TYMS,UGT1 A1 and XRCC1 genes) were genotyped in a single PCR for each individual,followed by fragment analysis. To avoid misinterpretation due to population substructure,we applied a previously developed set of 61 ancestryinformative markers that can also be genotyped by multiplex PCR. The statistical analyses were performed in Structure v.2.3.4,R environment and SPSS *** After statistical analyses with the control of confounding factors,such as genetic ancestry,three markers(rs79071878 in IL4,rs3730485 in MDM2 and rs28362491 in NFKB1) were positively associated with the development of GC. One of these markers(rs28362491) and the marker in the UGT1 A1 gene(rs8175347) were positively associated with the development of CRC. Therefore,we investigated whether the joint presence of the deleterious alleles of each marker could affect the development of cancer and we obtained positive results in all analyses. Carriers of the combination of alleles RP1 + DEL(rs79071878 and rs28361491,respectively) are at 10-times greater risk of developing GC than carriers of other combinations. Similarly,carriers of the combination of DEL + RARE(rs283628 and rs8175347) are at about 12-times greater risk of developing CRC than carriers of other *** These findings are important for the comprehension of gastric and CRC development,particularly in highly admixed populations,such as the Brazilian population.

The gain of transcription factor binding sites(TFBS) is believed to represent one of the major causes of biological *** we used strategies based on comparative genomics to identify 21,822 TFBS specific to the human lineage(TFBS-HS),when compared to chimpanzee and gorilla genomes. More than 40%(9,206) of these TFBS-HS are in the vicinity of 1,283 genes.A comparison of the expression pattern of these genes and the corresponding orthologs in chimpanzee and gorilla identified genes differentially expressed in human tissues. These genes show a more divergent expression pattern in the human testis and brain, suggesting a role for positive selection in the fixation of TFBS gains. Genes associated with TFBS-HS were enriched in gene ontology categories related to transcriptional regulation, signaling, differentiation/development and nervous ***, genes associated with TFBS-HS present a higher expression breadth when compared to genes in general. This biased distribution is due to a preferential gain of TFBS in genes with higher expression breadth rather than a shift in the expression pattern after the gain of TFBS.

Spatiotemporal Variability of Methane over the Amazon from Satellite Observations

The spatiotemporal variability of the greenhouse gas methane （CH4） in the atmosphere over the Amazon is studied using data from the space-borne measurements of the Atmospheric Infrared Sounder on board NASA＇s AQUA satellite for the period 2003-12. The results show a pronounced variability of this gas over the Amazon Basin lowlands region, where wetland areas occur. CH4 has a well-defined seasonal behavior, with a progressive increase of its concentration during the dry season, followed by a decrease during the wet season. Concerning this variability, the present study indicates the important role of ENSO in modulating the variability of CH4 emissions over the northern Amazon, where this association seems to be mostly linked to changes in flooded areas in response to ENSO-related precipitation changes. In this region, a CH4 decrease （increase） is due to the E1 Nifio-related （La Nifia-related） dryness （wetness）. On the other hand, an increase （decrease） in the biomass burning over the southeastern Amazon during very dry （wet） years explains the increase （decrease） in CH4 emissions in this region. The present analysis identifies the two main areas of the Amazon, its northern and southeastern sectors, with remarkable interannual variations of CH4. This result might be useful for future monitoring of the variations in the concentration of CH4, the second-most important greenhouse gas, in this area.

Capacitorless Solid-state Power Filter for Single-phase DC-AC Converters

Converters rely on passive filtering as a crucial element due to the high-frequency operational characteristics of power *** filtering methods involve a dual inductor-capacitor(LC)cell or an inductor-capacitor-inductor(LCL)***,capacitors are susceptible to wear-out mechanisms and failure ***,the necessity for monitoring and regular replacement adds to an elevated cost of ownership for such *** utilization of an active output power filter can be used to diminish the dimensions of the LC filter and the electrolytic dc-link capacitor,even though the inclusion of capacitors remains an indispensable part of the *** paper introduces capacitorless solid-state power filter(SSPF)for single-phase dc-ac *** proposed configuration is capable of generating a sinusoidal ac voltage without relying on *** proposed filter,composed of a planar transformer and an H-bridge converter operating at high frequency,injects voltage harmonics to attain a sinusoidal output *** design parameters of the planar transformer are incorporated,and the impact of magnetizing and leakage inductances on the operation of the SSPF is *** analysis,supported by simulation and experimental results,are provided for a design example for a single-phase *** total harmonic distortion observed in the output voltage is well below the IEEE 519 *** system operation is experimentally tested under both steady-state and dynamic conditions.A comparison with existing technology is presented,demonstrating that the proposed topology reduces the passive components used for filtering.
