A simple and fast method to calculate the coil currents and the external poloidal flux and magnetic field consistent with a given internal magnetohydrodynamic(MHD) equilibrium and coil geometry is presented. The constrained optimization technique employed allows to fix the exact position of the X-points. The external poloidal flux and field are calculated using Green's functions expressed in terms of elliptic integrals that are calculated using a fast and accurate *** calculation can be limited to the region between the plasma and the device wall. An example for a double null configuration is presented.

A statistical formalism overcoming some conceptual and practical difficulties arising in existing two-phase flow (2PHF) mathematical modelling has been applied to propose a model for dilute 2PHF turbulent Hows. Phase interaction terms with a clear physical meaning enter the equations and the formalism provides some guidelines for the avoidance of closure assumptions or the rational approximation of these terms. Continuous phase averaged continuity, momentum, turbulent kinetic energy and turbulence dissipation rate equations have been rigorously and systematically obtained in a single step. These equations display a structure similar to that for single-phase flows. It is also assumed that dispersed phase dynamics is well described by a probability density function (pdf) equation and eulerian continuity, momentum and fluctuating kinetic energy equations for the dispersed phase are deduced. An extension of the standard k-e turbulence model for the continuous phase is used. A gradient transport model is adopted for the dispersed phase fluctuating fluxes of momentum and kinetic energy at the non-colliding, large inertia limit. This model is then used to predict the behaviour of three axisymmetric turbulent jets of air laden with solid particles varying in size and concentration. Qualitative and quantitative numerical predictions compare reasonably well with the three different sets of experimental results, studying the influence of particle size, loading ratio and flow confinement velocity.

The porphyries are a group of metabolic disorders characterized by deficiencies in the activity of enzymes involved in the biosynthesis of *** erythropoietic protoporphyria(ePP),in the majority of cases an autosomal dominant disease,there is a mutation of the gene that encodes ferrochelatase(FeCH).FeCH deficiency is associated with increased concentrations of protoporphyrin in erythrocytes,plasma,skin and *** prevalence of this inherited disorder oscillates between 1:75 000 and 1:200 *** manifestations of ePP appear in early infancy upon first exposure to the ***,approximately 5%-20% of patients with ePP develop liver *** of protoporphyrin in the liver is associated with cholestatic phenomena and oxidative stress that predisposes to hepatobiliary disease of varying degrees of severity,such as cholelithiasis,mild parenchymal liver disease,progressive hepatocellular disease with end-stage liver disease and acute liver *** damage is the major risk in ePP patients,so surveillance and frequent clinical and biochemical liver follow-up is *** diagnostic approach consists in detecting increased levels of protoporphyrin,decreased activity of FeCH and genetic analysis of the FeCH gene.A variety of nonsurgical therapeutic approaches have been adopted for the management of ePP associated with liver disease,but none of these has been shown to be unequivocally ***,some may have a place in preparing patients for liver *** transplantation does not correct the constitutional deficiency of ***,there is a risk of recurrence of liver disease after liver transplantation as a result of continuing overproduction of *** authors recommend that bone marrow transplantation should be considered in liver allograft recipients to prevent recurrence of hepatic disease.

Biology of the aphid Therioaphis maculata was studied on alfalfa （Medicago sativa L.）, including four resistant （Mesa-Sirsa, CUF101, Baker and Lahontan） and two susceptible （ARC and Caliverde） alfalfa cultivars, and one of the most cropped Brazilian cultivars, Crioula. Under controlled conditions, antibiosis （i.e., reduced longevity, fecundity and increased mortality of the aphid） was observed mainly on the resistant alfalfa cultivars, except on Lahontan. Crioula seemed to be tolerant to aphids. Present data support geographic limitation usage of cultivars, and we suggest Baker and Mesa-Sirsa as sources of antibiosis, and provide biological information of a tropical T. maculata biotype on alfalfa.

Tamoxifen(TAM)is the first-line endocrine therapy for estrogen receptor-positive(eR+)breast cancer(BC).However,acquired resistance occurs in∼50%***,although the PI3K/AKT/mTOR pathway is a viable target for treatment of endocrine therapy-refractory patients,complex signaling feedback loops exist,which can counter the effectiveness of inhibitors of this ***,we analyzed signaling pathways and metabolism in eR+MCF7 BC cell line and their TAM-resistant derivatives that are co-resistant to endoxifen using immunoblotting,quantitative polymerase chain reaction,and the Agilent Seahorse XF *** found that activation of AKT and the energy-sensing kinase AMPK was increased in TAM and endoxifen-resistant ***,eRRα/PGC-1βand their target genes MCAD and CPT-1 were increased and regulated by AMPK,which coincided with increased fatty acid oxidation(FAO)and autophagy in TAM-resistant *** of AKT feedback-activates AMPK and eRRα/PGC-1β-MCAD/CPT-1 with a consequent increase in FAO and autophagy that counters the therapeutic effect of endoxifen and AKT ***,our results indicate increased activation of AKT and AMPK with metabolic reprogramming and increased autophagy in TAM-resistant *** inhibition of AKT and FAO/autophagy is necessary to fully sensitize resistant cells to endoxifen.

Nutlin-3a is a MDM2 antagonist and preclinical drug that activates p53. Cells with MDM2 gene amplification are especially prone to Nutlin-3a-induced apoptosis, though the basis for this is unclear. Glucose metabolism can inhibit apoptosis in response to Nutlin-3a through mechanisms that are incompletely understood. Glucose metabolism through the pentose phosphate pathway （PPP） produces NADPH that can protect cells from potentially lethal reactive oxygen species （ROS）. We compared apoptosis and glucose metabolism in cancer cells with and without MDM2 gene amplification treated with Nutlin-3a. Apoptosis in MDM2-amplified cells was associated with a reduction in glycolysis and the PPP, reduced NADPH, increased ROS, and depletion of the transcription factor SP1, which normally promotes PPP gene expression. In contrast, glycolysis and the PPP were maintained or increased in MDM2 non-amplified cells treated with Nutlin-3a. This was dependent on p53-mediated AKT activation and was associated with maintenance of SP1 and continued expression of PPP genes. Knockdown or inhibition of AKT, SP1, or the PPP sensitized MDM2-non-amplified cells to apoptosis. The data indicate that p53 promotes AKT and SP1-dependent activation of the PPP that protects cells from Nutlin-3a-induced apoptosis. These findings provide insight into how glucose metabolism reduces Nutlin-3a-induced apoptosis, and also provide a mechanism for the heightened sensitivity of MDM2-amplified cells to apoptosis in response to Nutlin-3a.
