许多城市的政府部门应用数字技术来提高城市软硬件设施系统,以培育创业型社会,开创新型服务,提高市民生活质量,优化城市体验,提振地方经济。建设有传统基础设施的城市系统往往天然具有垄断性,而基于数据的数字平台可以促进多元利益相关方的参与。本文提出了混合型数字化集成城市系统的概念以及设计方法论,旨在将数字技术和实体设施融入到城市环境中。而后介绍了由麻省理工学院可感知城市实验室学生设计、笔者指导的设计项目,以加拿大拉瓦尔市卡雷拉瓦尔公园建设与改造项目为例,整合了数字技术并提出设计方案—6名学生在课程中每人提出一项设计方案,分别为"屏幕激活""无限记忆""论坛""智能桌""我的a.世界"和"饮食足迹",这些设计能够促进公园改造项目建设进程中的公众参与。以人为本、因地制宜地进行方案设计可以促进数字技术与城市系统更好地融合,并增加城市对本地新兴利益相关方的吸引力,为他们营造新的城市体验。

背景脂肪基质血管细胞群（str.ma. va.cula. fr.0}tion cells，SVFCs）是再生医学中脂肪来源间充质干细胞（a.ipose-der.ved mesenchyma. stem cells，a.MSCs）的重要来源。从脂肪组织中获取SVFCs的方法有很多，包括机械法和酶消化法。获得的SVFCs的细胞组成可对其能力产生影响。因此，我们比较了采用不同方法从抽吸脂肪中获得的SVFCs的细胞组成和数量，包括离心、涡旋和胶原酶消化。方法采用真空负压抽吸法，从9例成年人皮下脂肪组织获取脂肪，年龄21～55岁，平均BMI为28。分别采用高速离心，离心后涡旋分离3min和酶消化法分离SVFCs。红细胞裂解后，对SVFCs进行细胞计数、增殖能力检测和流式细胞术检测。计量a.MSCs（CIM5-CD73＋CD90＋）、内皮细胞（CD45-CD31＋）和单核细胞／巨噬细胞的相对百分比。结果酶消化法获得的SVFCs数量最高（2．3×10^5／ml抽吸脂肪），高速离心法获得的SVFCs数量为其1／10，涡旋分离法获得的SVFCs数量为其1／20。3种方法获得的细胞活性相似（80％～90％）。3种方法获得的SVFCs成分有很大差异。酶消化法获得的SVFCs中，造血源性细胞的比例（32％）少于机械分离法获得的SVFCs中造血源性细胞的比例（70％～85％）；酶消化法获得的a.MSCs数量（60％）多于机械分离法获得的a.MSCs数量（6％～13％）。另外，与机械分离法相比，酶消化法获得的SVFCs中，炎性单核细胞／巨噬细胞的数量较少，而内皮细胞数量较多。结论虽然3种分离方法均能获得再生细胞，但机械分离法获得的造血细胞和炎性细胞的数量较高，而a.MSCs和内皮细胞的数量较低，这可能会降低其在再生医学中应用的潜能。
