Promotion of structural plasticity in area V2 of visual cortex prevents against object recognition memory deficits in aging and Alzheimer's disease rodents

Memory deficit,which is often associated with aging and many psychiatric,neurological,and neurodegenerative diseases,has been a challenging issue for treatment.Up till now,all potential drug candidates have failed to produce satisfa ctory effects.Therefore,in the search for a solution,we found that a treatment with the gene corresponding to the RGS14414protein in visual area V2,a brain area connected with brain circuits of the ventral stream and the medial temporal lobe,which is crucial for object recognition memory(ORM),can induce enhancement of ORM.In this study,we demonstrated that the same treatment with RGS14414in visual area V2,which is relatively unaffected in neurodegenerative diseases such as Alzheimer s disease,produced longlasting enhancement of ORM in young animals and prevent ORM deficits in rodent models of aging and Alzheimer’s disease.Furthermore,we found that the prevention of memory deficits was mediated through the upregulation of neuronal arbo rization and spine density,as well as an increase in brain-derived neurotrophic factor(BDNF).A knockdown of BDNF gene in RGS14414-treated aging rats and Alzheimer s disease model mice caused complete loss in the upregulation of neuronal structural plasticity and in the prevention of ORM deficits.These findings suggest that BDNF-mediated neuronal structural plasticity in area V2 is crucial in the prevention of memory deficits in RGS14414-treated rodent models of aging and Alzheimer’s disease.Therefore,our findings of RGS14414gene-mediated activation of neuronal circuits in visual area V2 have therapeutic relevance in the treatment of memory deficits.

Colorecal cancer(CRC)is one of the main causes of cancer-related mortality in the deweloped world despite recent developments in detection and treatment Several epidemiologial studis indicate that metformin,a widdy prescribed antidiabetic drug,exerts a protective effect on different cancers incuding CRC.Furthermore,a recent double blind placebo controlled,randomized trial showed that metformin significantly decreased colorectal adenoma recurrence.Studics exploring the medhanism of action of metformin in cells derived from different types of ancers reported many efects including rapiratory chain complex 1 inhibition,Akt phosphorylation inhibition,ATP depltion,PKA activation and Wnt signaling inhibition.However,many of these results were obtained employing metformin at concentrations several fold higher than those achieved in target tissuas in diabetic patients receiving therapeutic recommended doses of metformin.In contrast,recent studies obtained with metformin at concentrations compatible with those detected in human intestins revealed that metformin dicit responses that targetβ-catenin,PI3KAkt,e-adherin,pl20-catenin and focal adhesion kinase which are key molecules and signaling pathways associated to colorectal cancar development.This brief review revisit several know aspects as well as nowd ones on the effects of metformin on cancer cells.

研究激光粉末床熔融增材制造(AM)(又称直接金属激光烧结)技术制备的Ti6Al4V合金等离子体电解氧化(PeO)处理及其电化学行为。通过短时间(eO处理(也称闪速PeO),在AM合金和传统合金表面制备了3~10μm厚、含Ca和P的涂层。然后在改良的α-MeM溶液中,通过动电位极化曲线和电化学阻抗谱(eIS)评估了合金的电化学行为。与传统合金相比,AM合金中形成了细小的层片状α显微组织和层间小尺寸的β相颗粒,这促使了火花的产生,从而促进了PeO涂层的生长。闪速PeO涂层提高了传统合金和AM合金的耐腐蚀性,最薄的涂层(eO处理也足以成功避免这种情况。

Role of bioengineering structures made of willow cuttings in marly sediment trapping: assessment of three real-size experiments in the Southern French Alps

Improving knowledge on the ability of bioengineering structures made of willow cuttings to enhance efficient and sustainable sediment trapping in marly gullies in the Southern French Alps under a mountainous Mediterranean climate, to decreasesediment trapped per experiment, the mean annual values reached 0.25, 0.14 and 0.08 m3 yr-1 in experiments C, A and B, respectively. Maximum values for one structure reached 1.94 m3 per year in experiment C. The significance of the volumes ofsediment yield at their outlets, is a key issue today for the international scientific community working in geosciences and ecology. This study therefore aims to assess three real-size experiments(A, B and C) carried out between 2003 and 2013 in this environment. A total of 157 bioengineering structures using purple and white willow(Salix purpurea and Salix incana)cuttings – which have been shown to resprout and survive more than 2 years after their installation,corresponding to brush layers with brush mats on wooden sills(BLM), 1.2 m wide and 2 m long,installed on the floors of 33 experimental marly gullies, were monitored. The results showed that sediment trapping occurred upstream of the vegetation barriers from the 1 st year onwards.Considering the depth of sediment trapped per experiment, the mean annual values reached 11.2 cm yr-1 after 3 years in experiment A, 7.7 cm yr-1 after 2–4 years in experiment C and 5.1 cm yr-1 after 5 years in experiment B. Occasionally, BLMs showed that they could trap up to 16 and 15 cm yr-1 in experiments A and C, respectively. Considering the volumes oftrapped sediment and the sustainability of sediment trapping are discussed, and rules for bioengineering strategies are proposed.

Objectives: To evaluate the frequency of medical consultation for gastro-oesophageal reflux symptoms (GORS) and to ascertain reasons for consultation and associated factors. Methods: Telephone survey of 2,500 persons using a validated questionnaire. Data were analyzed using multiple logistic regression, in which the dependent variable was consultation for GORS and the independent variables were sociodemographic factors and the clinical characteristics of GORS. Results: Of those reporting GORS, 28%had consulted their physician about them in the last year. Main reasons for seeking medical advice were GORS-induced concern in 48%, and intensity of GORS in 42.6%. Factors independently associated with medical consultation were: housewife (odds ratio (OR) 1.9; 95%CI 1.1-3.4) or retired status (OR 2.3; 95%CI 1.3-4.1), frequent GORS (OR 2.4; 95%CI 1.6-3.5), severe GORS (OR 2.4; 95%CI 1.4-4.2), nocturnal GORS (OR 1.6; 95%CI 1.1-2.3), and association of epigastric pain (OR 1.7; 95%CI 1.0-2.7) or nausea (OR 1.7; 95%CI 1.1-2.8). Conclusion: The high frequency of medical consultation for GORS in Spain makes it an important healthcare problem. The fact that half the patients seek medical advice due to concern about the nature of GORS illustrates the benefits of explaining the nature, treatment and prognosis of GORS. The clinical relevance of GORS does not reside in its frequency alone, since severe or nocturnal symptoms, albeit occasional, are also associated with medical consultation.

-592 and -1082 interleukin-10 polymorphisms in pulmonary tuberculosis with type 2 diabetes

Objective:To determine the polymorphisms of Interleukin-10(IL-10)(-592,-1082) in pulmonary tuberculosis(PTB)with and without type 2 diabetes(T2D).Methods:We studied a Mexican mestizo population of 37 patients with TB in remission(TBr) and 40 with active pulmonary TB (PTB),21 patients with TB + T2D.47 blood donors accepted,and 13 healthy health-care workers with tuberculin skin lest positive.Determination of IL-10 polymorphisms was performed by real-lime Polymerase chain reaction.Results:IL-10-592C/A presented in a greater proportion in healthy individuals than in patients with type 2 diabetes and TB in a not quite significant statistically manner.IL-10-1082A/A presented more frequently in the group of patients with both diseases,not being statistically significant in comparison with the group of healthy subjects. Conclusions:This study describes two important new findings.First,it reveals that the IL-10 (-592 A/A and -592 C/C) polymorphisms were found in a greater proportion in a group of patients with T2D and TB than in healthy subjects.Second,the study provides evidence that the(-1082 G/G) polymorphism presented with greater frequency in healthy individuals than in patients with both ol these diseases.

Stability analysis plays a central role in nonlinear system theory and engineering application. Over the past few years, the stability analysis of fuzzy systems has been proposed and there are many successful applications in practical engineering. Therefore, in this paper firstly proposed is the stability analysis on oceanic structure by fuzzy models. In the present study, Takagi-Sugeno （T-S） fuzzy model is proposed for a time delay tension leg platform （TLP） system subjected to an external wave force. In terms of stability analysis, linear matrix inequality （LMI） conditions are derived via Lyapunov theory to guarantee the stability of the TLP system.

Plant performance and morphofunctional differentiation in response to edaphic variation in Iberian columbines: cues for range distribution?

Aims experimental studies of the response to environmental variation of closely related taxa are needed to understand the mechanisms underly-ing phenotypic divergence,habitat segregation and range span within a radiating genus.We explored the magnitude of phenotypic differenti-ation and adaptive plasticity in relation to edaphic variation and its sig-nificance for habitat segregation and range span in Iberian columbines.Methods We performed a common garden experiment varying soil type(cal-careous vs.siliceous)and depth(rocky-shallow vs.deep)with two pairs of widespread and narrowly distributed subspecies of Iberian columbines:Aquilegia vulgaris(subspp.vulgaris and nevadensis)and Aquilegia pyrenaica(subspp.pyrenaica and cazorlensis).We com-pared tolerance to edaphic variation,trait differentiation and adaptive plasticity of 10 morpho-functional traits between species and distri-bution ranges.additionally,we obtained estimates of the competitive environment faced by each taxon in two to four populations per taxa.Important Findingsresults partially support hypotheses of higher competitive ability and tolerance to edaphic variation in widespread than in narrow endemic taxa.at the species level,the widely distributed taxon was the most tolerant to edaphic variation.Within species,no consistent pattern emerged since the population from the most widespread subspecies was the most tolerant in A.vulgaris but not in A.pyr-enaica.Columbines were differentiated in many traits at species and range level.However,the pattern of differentiation does not fully support the hypothesis of higher specialization and stress toler-ance in narrow endemics.although plasticity was generally low,the results support the hypothesis of adaptive plasticity in widespread but not in restricted taxa at least at species level.ecological differences(adaptive plasticity and competitive ability among others)may have contributed to phenotypic divergence and edaphic niche segregation,as well as to differences in range span among columbines.

Objective:To assess the role of polymerase chain reaction(PCR)in serum sauples,in the diagnosis of osteoarticular tuberculosis(OTB)in a setting where only clinical and imaging diagnoses determine the treatment.Methods:A total of 44 consecutive serum specimens were collected from clinically suspected OTB patients,based on clinical and radiological[X-ray or magnetic resonance imagng/computecl tomography]features.They were scrcened by in-house nested PCR.In addition,a few specimens were examined by Gram stain,acid-fast bacilli stain,histand routine bacterial culture.A total of 39 specimens were collected from patients suffering from other bone diseases of nontuberculous origin and included as negative controls.Results:of the 44 clinically suspected OTB patients,in-house nested PCR was positive in 40(91%)cases;PCR was negative in 38(97%)negative controls.Sensitivity and specificity of our in—house nested PCR was 90.3%and 97.4%,respectively.The PCR report was available within 48 h.It was possible to standardize serum PCR technique and in positive cases,a good n was observed in terms of an adequate treatment response.Conclusions:Nested PCR in serum samples is a rapid,highly sensitive and specific modality for OTB detection,PCR should be performed in addition to clinical evaluation,imaging studies,acidfast bacilli staining,culture and histopathology diagnosis,if possible.

Prophylactic liver transplantation for high-risk recurrent hepatocellular carcinoma

Hepatocellular carcinoma(HCC) is the second most common cause of cancer-related death in the world. Radical treatment of HCC in early stages results in a long disease-free period and improved overall survival. The choice of optimal management strategy for HCC mainly depends on the severity of the underlying liver disease. For patients with decompensated liver cirrhosis and HCC within Milan criteria(MC), liver transplant(LT) is the choice of treatment. However, for patients with good residual liver reserve and HCC within MC, selection of other curative treatments such as liver resection(LR) or radiofrequency ablation may be a reasonable alternative. For patients without cirrhosis, LR can result in an overall survival similar to that provided by LT. Therefore, it is an accepted alternative to LT especially in areas with organ shortage. However, the cumulative 5-year recurrence rate of HCC post LR might be as high as 70%. For initial transplant-eligible(within MC) patients with recurrent HCC post LR, salvage liver transplant(SLT) was first proposed in 2000. However, most patients with recurrent HCC considered for SLT are untransplantable cases due to HCC recurrence beyond MC or comorbidity. Thus, the strategy of opting for SLT results in the loss of the opportunity of LT for these patients. Some authors proposed the concept of "de principe liver transplant"(i.e., prophylactic LT before HCC recurrence) to prevent losing the chance of LT for these potential candidates. Factors associated with the failure of SLT will be dissected and discussed in three parts: Patient, tumor, and underlying liver disease. Regarding patient-related factors, the rate of transplantability depends on patient compliance. Patients without regular follow-up tend to develop HCC recurrence beyond MC at the time of tumor detection. Advancing age is another factor related to severe comorbidities when LT is considered for HCC recurrence, and these elderly candidates become ineligible as time goes by.
