BACKGROUND Although the association of attention deficit hyperactivity disorder(ADHD)with psychiatric disorders is well known,its association with somatic diseases is *** few studies have investigated the gastrointestinal(GI)morbidity in adult patients with *** To measure gastrointestinal comorbidity and its burden on healthcare in young adults with *** The cohort included subjects aged 17-35 years recruited to the Israel Defense Forces in 2007-2013,33380 with ADHD and 355652 without(controls).The groups were compared for functional and inflammatory conditions of the gastrointestinal tract and clinic and specialist visits for gastrointestinal symptoms/disease during service(to 2016).Findings were analyzed by generalized linear models adjusted for background *** Compared to controls,the ADHD group had more diagnoses of functional gastrointestinal disorders(referred to as FGID),namely,dyspepsia[odds ratio(OR):1.48,95%confidence interval(CI):1.40-1.57,P<0.001],chronic constipation(OR:1.64,95%CI:1.48-1.81,P<0.001),and irritable bowel syndrome(OR:1.67,95%CI:1.56-1.80,P<0.001)but not of organic disorders(inflammatory bowel disease,celiac disease).They had more frequent primary care visits for gastrointestinal symptoms[rate ratio(RR):1.25,95%CI:1.24-1.26,P<0.001]and referrals to gastrointestinal specialists(RR:1.96,95%CI:1.88-2.03,P<0.001)and more episodes of recurrent gastrointestinal symptoms(RR:1.29,95%CI:1.21-1.38,P<0.001).Methylphenidate use increased the risk of dyspepsia(OR:1.49,95%CI:1.28-1.73,P<0.001)and constipation(OR:1.42,95%CI:1.09-1.84,P=0.009).CONCLUSION ADHD in young adults is associated with an excess of FGID and increased use of related health *** is needed to determine if an integrative approach treating both conditions will benefit these patients and cut costs.

Real-time Quantitative Study for Rhythmic Profiles of Dihydr...

In Southern China,chrono-chemotherapy has been tried to treat nasopharyngeal carcinoma (NPC) patients using 5-Fu combined with platinum complexes. However,there was no enough data about circadian profiles of the circadian profiles of dihydropyrimidine dehydrogenase (DPD), thymidylate synthase (TS) in mRNA level on Chinese NPC patients. In present study,total of 13 NPC patients were involved,9 males and 4 *** veinal blood was collected at 6 time points (12:00,16:00,20:00,and 24:00,04:00,08:00) during a single 24 hours from all the participants. The circadian gene expression profiles of DPD,TS in PBMC were measured using the real-time quantitative RT-PCR method. In addition, the whole-blood reduced Glutathione (GSH), Dihydrouracil and uracil ratios (UH2/U ratios)(an indirect method for measuring DPD activity, HPLC method), plasma cortisol level were also measured. In present study, Mesor (rhythm-adjust mean) of DPD gene copies was 6.21×107±7.76±106 copy/μg RNA (mesor±SEM), and it was 828×103±8.33×102 copy/ug RNA for TS gene. It was of no statistical significance for DPD mRNA expression according to sampling time (ANOVA for Repeated Measures,F= 1.067,P=0.369;Cosinor,P=0.078), and for TS (ANOVA for Repeated Measures, F= 0.505, P = 0.765, Cosinor, P = 0.1068). The plasma cortisol displayed a clearly circadian rhythm and the peak time located at 09:14±01:11 (Corsinor, P < 0.0001). The mean value of UH2/U was 4.80±0.20 and a significant circadian rhythm was not displayed (ANOVA for Repeated Measures, F= 0.352,P = 0.862;Corsinor, P = 03053). The mesor value of GSH was 19.60±1.11 (nmol/L) and GSH displayed a possible circadian rhythm and the acrophase located at 05:02 (ANOVA for Repeated Measures,F= 3.985, P = 0.050;Corsinor, P=0.0645). In conclusion, a clear circadian rhythm was not displayed for DPD gene and TS gene in mRNA level, also not found for DPD activity on Chinese NPC patients, although these patients still maintained an obvious circadian rhythm in

Adriamycin(doxorubicin),a common cancer chemotherapeutic drug,can be used to induce a model of chronic progressive glomerular disease in *** our studies,we evahuated renal changes in a rat model after Adriamydin injection using two photon microscopy(TPM),optical coherence tomography(OCT)and Doppler OCT(DOCT).Taking advantage of deep penetra-tion and fast scanning speed for three dimensional(3D)label-free imaging,OCT/DOCT system was able to reveal glomerular and tubular pathology noninvasively and in real *** imaging renal pathology following the infusion of fAuorophore-labeled dextrans of different molecular weights,TPM can provide direct views of glomerular and tubular flow dynamics with the onset and progression of renal ***,glomerular permeability and filtration,proximal and distal tubular flow dynamics can be revealed.6-8 weeks after injection of Adriamycin,TPM and OCT/DOCT imaging revealed glomerular sclerosis,compromised flow across the glomerular wall,tubular atrophy,tubular dilation,and variable intra-tubular flow *** results indicate that TPM and OCT/DOCT provide real-time imaging of renal pathology in vivo that has not been previously available using conventional microscopic procedures.

User-friendly tools for robust transcriptional activation of endogenous genes are highly demanded in plants. We previously showed that a dCas9-VP64 system consisting of the deactivated CRISPR- associated protein 9 （dCasg） fused with four tandem repeats of the transcriptional activator VP16 0/1=64） could be used for transcriptional activation of endogenous genes in plants. In this study, we developed a second generation of vector systems for enhanced transcriptional activation in plants. We tested multiple strategies for dCasg-based transcriptional activation, and found that simultaneous recruitment of VP64 by dCas9 and a modified guide RNA scaffold gRNA2.0 （designated CRISPR-Act2.0） yielded stronger transcrip- tional activation than the dCas9-VP64 system. Moreover, we developed a multiplex transcription activator- likeeffector activation （mTALE-Act） system for simultaneous activation of up to four genes in plants. Our results suggest that mTALE-Act is even more effective than CRISPR-Act2.0 in most cases tested. In addition, we explored tissue-specific gene activation using positive feedback loops. Interestingly, our study revealed that certain endogenous genes are more amenable than others to transcriptional activation, and tightly regulated genes may cause target gene silencing when perturbed by activation probes. Hence, these new tools could be used to investigate gene regulatory networks and their control mechanisms. Assembly of multiplex CRISPR-Act2.0 and mTALE-Act systems are both based on streamlined and PCR-independent Golden Gate and Gateway cloning strategies, which will facilitate transcriptional activation applications in both dicots and monocots.

AIM:To evaluate the quality of life(QOL)and level of depression among participants with high myopia in Nigeria and the demographic factors associated with these ***:This cross-sectional study was conducted on 100 adult participants with high myopia(defined as refractive error≤-5.00 D or worse,and uncorrected visual acuity worse than 6/18 in the better seeing eye)attending ophthalmology centres in Nigeria from 2 October 2021 to 30 August *** means and standard deviations were calculated for each of the four domains of World Health Organization Quality of life scale(WHOQOL-BREF)using the transformed *** Beck Depression Inventory(BDI)scale was used to assess the level of ***:The highest and the lowest mean scores of WHOQOL-BREF domains were found for the psychological and physical health domains(mean percentage scores were 67.0[95% confidence intervals(CI)64.1-68.9]and 55.3(95%CI 51.8-58.8,P<0.001),***-way analysis of variance(ANOVA)revealed significant differences in physical health with educational status(higher among those with tertiary education:mean difference 0.9,95%CI-0.2-2.1;P=0.049),differences in psychological health with working status(higher among those who were working 1.2,95%CI 0.3-2.1;P=0.012).Also,the result showed a statistically significant association between environmental health and marital status(higher among non-married:1.7,95%CI-0.9-2.3;P=0.012)while overall health was associated with place of residence(higher in urban areas:2.3,95%CI 1.2-3.5;P=0.024).For depression,one in every nine participants reported major depressive symptom,mostly younger people(aged 16-29 vs 30-49y:17.0%vs 0,P=0.019),and slightly more women than men(14.3%vs 0,P=0.064).There were significant negative correlations between the depression scores and psychological health(r=-0.48,P<0.001),physical health(r=-0.29,P=0.002),social and relationship(r=-0.49,P<0.001),environmental(r=-0.48,P<0.001)and overall health(r=-0.49,P<0.001)C
