Introducing PCR products into pla.mids vectors is key for molecula. techniques. Idea.ly cloning vectors a.e ea.y to construct, modify a.d propa.a.e, neither require a.va.ced techniques nor specia. equipment or rea.ents a.d efficiently incorpora.e PCR products a. close to zero empty vector ba.kground. We provide a. ea.y to engineer self-ma.e cloning vector, neither requiring sophistica.ed tools or techniques nor a.va.ced cloning knowledge. Through recombina.ion we obta.ned the pUC18ccdB vector, ca.rying the ccdB suicide gene within the pUC18 ba.kbone. When Sma. clea.ed (within the ccdB) vector wa. T4 liga.ed with sma.l (0.2 kbp) a.d intermedia.e (1.3 to 2.2 kbp) blunt end PCR-products a.d tra.sformed into E. coli, the a.ount of clones with incorpora.ed PCR product wa. compa.a.le to commercia. PCR-cloning kits a.d a. a.close to zero PCR product nega.ive ba.kground. In conclusion we present a.simple, versa.ile a.d chea. a.proa.h to a. efficient “home ma.e” PCR-cloning vector tha. a.lows integra.ion of crude blunt end PCR products a. close to zero ba.kground.
