RNAi-Mediated Silencing of ITPK Gene Reduces Phytic Acid Content,Alters Transcripts of Phytic Acid Biosynthetic Genes,and Modulates Mineral Distribution in Rice Seeds

Phytic acid is the principal storage form of phosphorus in plant seeds and an essential signalling molecule in several regulatory processes of plant ***,it is known as an anti-nutrient compound owing to its potent chelating ***,reducing the phytic acid content in crops is *** involving regulation of MIPS and IPK1 genes to generate low phytate rice have been reported ***,the functional significance of OsITPK and the effect of its down-regulation on phytic acid content and the associated pleiotropic effects on rice have not yet been *** this study,tissue specific RNA interference(RNAi)-mediated down-regulation of a major ITPK homolog(OsITP5/6K-1)resulted in 46.2%decrease in phytic acid content of T2 transgenic seeds with a subsequent 3-fold enhancement in the inorganic phosphorus *** of OsITP5/6K-1 altered the transcript levels of essential phytic acid pathway genes,without significantly affecting the transcript levels of other OsITPK ***,the mapping of elements through X-ray microfluorescence analysis revealed significant changes in the spatial distribution pattern and translocation of elements in low phytate ***,low phytate polished seeds exhibited 1.3-fold and 1.6-fold enhancement in iron and zinc content in the grain endosperm,*** of OsITP5/6K-1 also altered the amino acid and myo-inositol content of the transgenic *** results successfully established that RNAi-mediated silencing of OsITP5/6K-1 gene significantly reduced the phytate levels in seeds without hampering the germination potential of seeds and plant *** present study provided an insight into the mechanism of phytic acid biosynthesis pathway.

Dynamics of novel COVID-19 in the presence of Co-morbidity

A novel coronavirus(COVID-19)has emerged as a global serious public health issue from December *** having a weak immune system are more susceptible to coronavirus *** is a double challenge for people of any age with certain underlying medical conditions including cardiovascular disease,diabetes,high blood pressure and cancer ***-morbidity increases the probability of COVID-19 *** this paper a deterministic compartmental model is formulated to understand the transmission dynamics of *** mathematical analysis of the model shows that it exhibits backward bifurcation phenomenon when the basic reproduction number is less than *** the case of no re-infection it is shown that having the reproduction number less than one is necessary and sufficient for the effective control of COVID-19,that is,the disease free equilibrium is globally asymptotically stable when the reproduction threshold is less than ***,in the absence of reinfection,a unique endemic equilibrium of the model exists which is globally asymptotically stable whenever the reproduction number is greater than *** simulations of the model,using data relevant to COVID-19 transmission dynamics,show that the use of efficacious face masks publicly could lead to the elimination of COVID-19 up to a satisfactory *** study also shows that in the presence of co-morbidity,the disease increases significantly.

The effective study of hybrid machining processes(HMPs),in terms of modeling and optimization has always been a challenge to the *** combined approach of Artifcial Neural Network(ANN)and Non-Dominated Sorting Genetic Algorithm-II(NSGA-II)has attracted attention of researchers for modeling and optimization of the complex machining *** this paper,a hybrid machining process of Electrical Discharge Face Grinding(EDFG)and Diamond Face Grinding(DFG)named as Electrical Discharge Diamond face Grinding(EDDFG)have been studied using a hybrid methodology of *** this study,ANN has been used for modeling while NSGA-II is used to optimize the control parameters of the EDDFG *** observations of input-output relations,the experiments were conducted on a self developed face grinding setup,which is attached with the ram of EDM *** experimentation,the wheel speed,pulse current,pulse on-time and duty factor are taken as input parameters while output parameters are material removal rate(MRR)and average surface roughness(Ra).The results have shown that the developed ANN model is capable to predict the output responses within the acceptable limit for a given set of input *** has also been found that hybrid approach of ANN-NSGAII gives a set of optimal solutions for getting appropriate value of outputs with multiple objectives.

Efficient Microorganism Compost Benefits Plant Growth and Improves Soil Health in Calendula and Marigold

Organic farming needs to be promoted as a means of sustaining soil health and reducing the cost of farming, especially for small *** present study deals with the effect of organic compost, prepared using Efficient Microorganism(EM) consortium and applied along with full or half of the recommended dose of chemical fertilizers, on the growth of Calendula and Marigold plants, soil physico-chemical parameters and soil enzyme activities. Soil enzyme activities were improved with the increase in the rate of EM compost application in both Calendula and Marigold. Carotenoid pigment increased by 46.11% and 12.19% with application of EM compost over the control in Calendula and Marigold flowers respectively. Soil humus, available nitrogen and organic carbon content also increased due to the supplementation of EM compost resulting in better soil fertility. For Calendula, treatment T5(Half dose NPK + EM compost 20 000 kg·hm^(-2)) was found to be the most promising in terms of acid phosphatase(82.63 μg p-Nitrophenyl Phosphate·g^(-1)·h^(-1)), dehydrogenase(10.46 μg Triphenyl Formazan·g^(-1)·d^(-1)) and β-glucosidase(0.30 IU·g^(-1)) activities. In Marigold, treatment C(Half dose NPK + EM compost 5 000 kg·hm^(-2)) was the most promising in terms of amendment in soil enzyme activities.

Superpenumbral chromospheric flare

We observed a C-class flare at the outer boundary of the superpenumbra of a sunspot. The flare was triggered by an emerging magnetic bipolar region that was obliquely oriented with respect to the superpenumbral fibrils. The flare started due to the low height magnetic reconnection of emerging magnetic flux with a superpenumbral field resulting in hot multi-temperature plasma flows in the inverse Evershed flow channel and its overlying atmosphere. Inverse Evershed flows in the chromosphere start from the superpenumbra towards the sunspot that end at the outer boundary of the penumbra. The hot plasma flow towards the sunspot in the inverse Evershed channels show about 10 km s^-1 higher velocity in Ha wavelengths compared to the plasma emissions at various temperatures as seen in different AIA filters. Even though these velocities are about seven times higher than the typical inverse Evershed flow speeds, the flow is diminished at the outer boundary of the sunspot＇s penumbra. This suggests that the superpenumbral field lines that carry inverse Evershed flows are discontinued at the boundary where the penumbral field lines dive into the sun and these two sets of field lines are completely distinct. The discontinuity in the typical magnetic field and plasma properties at the adjoining of these two sets of field lines further leads to discontinuity in the characteristic magnetoacoustic and Alfvrn speeds, thereby stopping the plasma flows further on. The multi-temperature plasma in the inverse Evershed channels exhibits possible longitudinal oscillations initially during the onset of the flare, and later flows towards the sunspot. In the multi-temperature view, the different layers above the flare region have a mixture of supersonic as well as subsonic flows.

Design, synthesis and MAO inhibitory activity of 2-(arylmethylidene)-2,3-dihydro-1-benzofuran-3-one derivatives

A series of 2-（arylmethylidene）-2,3-dihydro-1-benzofuran-3-one derivatives（aurones, 1–20） were synthesized and screened for their inhibitory activity against h MAO. Seventeen compounds（1–5, 7–17,19） were found to be selective towards h MAO-B, while two were non-selective（6 and 20） and one（18）selective towards h MAO-A. Compound 17（Ki = 0.10 0.01 mmol/L） was found to be equally potent and selective towards h MAO-B, when compared with the standard drug Selegiline（Ki = 0.12 0.01 mmol/L）.Nature and position of substitution in aryl ring at 2nd position of benzofuranone influences h MAO-B inhibitory potency, while their structural bulkiness influences selectivity between h MAO-A and h *** docking simulation was also carried out to understand the interaction of inhibitor with the enzyme at molecular level, and we found the docking results were in good agreement with the experimental values. Comparison of the activity profile of the aurones with their corresponding flavones reported earlier by our group revealed that there exists no difference in potency as well as selectivity.

Diagnosis of Helicobacter pylori : What should be the gold standard?

Since the discovery of Helicobacter pylori (H. pylori) in 1983, numerous detection methods for the presence of the bacterium have been developed. Each one of them has been associated with advantages and disadvantages. Noninvasive tests such as serology, 13C urea breath test (UBT) and stool antigen tests are usually preferred by the clinicians. Serology has its own limitation especially in endemic areas while 13C UBT is technically very demanding. The stool antigen detection method, although specific, is usually associated with poor sensitivity. The 13C UBT is believed to be specific, but with present revelation of the fact that stomach is colonized by many other urease producing bacteria makes it questionable. Histology, culture, rapid urease test and polymerase chain reaction (PCR) are the tests which are carried out on antral biopsies collected by invasive means. Histology has been proposed to be very sensitive and specific but the question is how by simply looking the morphology of the bacteria in the microscope, one can claim that the curved bacterium is exclusively H. pylori. Rapid urease test (RUT), the doctor’s test, is also challenged because the presence of other urease producing bacteria in the stomach cannot be denied. Moreover, RUT has been reported with poor sensitivity specially, when density of the bacterium is low. Isolation of H. pylori is essential to investigate its growth requirements, antibiotic susceptibility testing, studying virulence factor to develop vaccine and many more explorations. It has also got several disadvantages i.e., special condition for transporting, media, incubation and few days waiting for the colonies to appear, apart from the speed essentially needed to process the specimens. Till date, majority of the microbiological laboratories in the world are not equipped and trained to isolate such fastidious bacterium. Kinetic approach to the study of froth flotation applied to a lepidolite ore

The number of published studies related to the optimization of lithium extraction from low-grade ores has increased as the demand for lithium has grown. However, no study related to the kinetics of the concentration stage of lithium-containing minerals by froth flotation has yet been reported. To establish a factorial design of batch flotation experiments, we conducted a set of kinetic tests to determine the most selective alternative collector, define a range of pulp p H values, and estimate a near-optimum flotation time. Both collectors（Aeromine 3000 C and Armeen 12D） provided the required flotation selectivity, although this selectivity was lost in the case of pulp p H values outside the range between 2 and 4. Cumulative mineral recovery curves were used to adjust a classical kinetic model that was modified with a non-negative parameter representing a delay time. The computation of the near-optimum flotation time as the maximizer of a separation efficiency（SE） function must be performed with caution. We instead propose to define the near-optimum flotation time as the time interval required to achieve 95%–99% of the maximum value of the SE function.
