Interplay of RNA-binding proteins controls germ cell development in zebrafish

The specification of germ cells in zebrafish mostly relies on an inherited mechanism by which localized maternal determinants, called germ plasm, confer germline fate in the early embryo. Extensive studies have partially allowed the identification of key regulators governing germ plasm formation and subsequent germ cell development. RNA-binding proteins, acting in concert with other germ plasm components, play essential roles in the organization of the germ plasm and the specification, migration, maintenance, and differentiation of primordial germ cells. The loss of their functions impairs germ cell formation and causes sterility or sexual conversion. Evidence is emerging that they instruct germline development through differential regulation of mRNA fates in somatic and germ cells. However, the challenge remains to decipher the complex interplay of maternal germ plasm components in germ plasm compartmentalization and germ cell specification. Because failure to control the developmental outcome of germ cells disrupts the formation of gametes, it is important to gain a complete picture of regulatory mechanisms operating in the germ cell lineage. This review sheds light on the contributions of RNA-binding proteins to germ cell development in zebrafish and highlights intriguing questions that remain open for future investigation. Hole cleaning evaluation and installation spacing optimization of cuttings bed remover in extended-reach drilling

In extended-reach or long-horizontal drilling,cuttings usually deposit at the bottom of the *** cuttings accumulate to a certain thickness,complex problems such as excessive torque and drag,tubing buckling,and pipe stuck probably occur,which results in a lot of non-productive time and remedial *** bed remover can efficiently destroy deposited cuttings in time through hydraulic and mechanical stirring *** paper aims to build a method for hole cleaning evaluation and installation spacing optimization of cuttings bed remover to improve the wellbore cleaning ***,a Computational Fluid Dynamics approach with Eulerian—Eulerian multiphase model was utilized to investigate the mechanism of cuttings transportation,and a new type of cuttings bed remover was ***,an evaluation method of hole cleaning effect of remover was *** that,the effects of several drilling parameters on hole cleaning including flow rate of drilling fluid,rotational speed of drillpipe,rate of penetration,wellbore size,rheological property of drilling fluid,and remover eccentricity on the performance of cuttings bed remover were *** results demonstrate that the new type of remover with streamline blade performs better than conventional *** efficiency of hole cleaning is greatly improved by increasing the rotational speed of drillpipe,flow rate of drilling fluid,remover eccentricity,and 6 rpm Fann dial reading for drilling *** higher rate of penetration and large wellbore size result in worse hole *** findings can serve as an important guide for the structure optimization design of cuttings bed remover and installation spacing of removers.

The effect of abiotic stresses on plant C:N:P homeostasis and their mitigation by silicon

In crop plants, various environmental stresses affect the balance of carbon, nitrogen, and phosphorus(C:N:P), leading to biochemical and physiological alterations and reductions in yield. Silicon(Si) is a beneficial element that alleviates plant stress. most studies involving silicon have focused on physiological responses, such as improvements in photosynthetic processes, water use efficiency, and antioxidant defense systems. But recent research suggests that stressed plants facing either limited or excessive resources(water, light, nutrients, and toxic elements), strategically employ Si to maintain C:N:P homeostasis, thereby minimizing biomass losses. Understanding the role of Si in mitigating the impact of abiotic stresses on plants by regulating C:N:P homeostasis holds great potential for advancing sustainable agricultural practices in crop production. This review presents recent advances in characterizing the influence of environmental stresses on C:N:P homeostasis, as well as the role of Si in preserving C:N:P equilibrium and attenuating biological damage associated with abiotic stress. It underscores the beneficial effects of Si in sustaining C:N:P homeostasis and increasing yield via improved nutritional efficiency and stress mitigation.

A numerical model to simulate the transient frictional viscoelastic sliding contact

Sliding motion has always been one of the major concerns when it comes to the analysis of viscoelastic contact problems.A new model simulating the transient sliding contact of smooth viscoelastic surfaces is developed in this *** taking the dry contact friction and the coupling between shear tractions and normal pressure into account,the effect of the early partial slip period,which is often neglected in the study of viscoelastic sliding contact problems,is investigated *** with solutions based on the frictionless assumption,the steady-state pressure profile is found to be slightly different under the effect of the partial slip regime,including a lower peak pressure and the shift of the contacting region in the direction opposite to the sliding ***,the time required for the viscoelastic contact to reach its steady state is delayed owing to the partial slip period preceding the global sliding motion.

Prediction models of burst strength degradation for casing with considerations of both wear and corrosion

Casing wear and casing corrosion are serious problems affecting casing integrity failure in deep and ultra-deep *** paper aims to predict the casing burst strength with considerations of both wear and ***,the crescent wear shape is simplified into three categories according to common mathematical ***,based on the mechano-electrochemical(m-E)interaction,the prediction model of corrosion depth is built with worn depth as the initial condition,and the prediction models of burst strength of the worn casing and corroded casing are ***,the accuracy of different prediction models is validated by numerical simulation,and the main influence factors on casing strength are *** last,the theoretical models are applied to an ultra-deep well in Northwest China,and the dangerous well sections caused by wear and corrosion are predicted,and the corrosion rate threshold to ensure the safety of casing is *** results show that the existence of wear defects results in a stress concentration and enhanced m-E interaction on corrosion depth *** accuracy of different mathematical models is different:the slot ring model is most accurate for predicting corrosion depth,and the eccentric model is most accurate for predicting the burst strength of corroded *** burst strength of the casing will be overestimated by more than one-third if the m-E interaction is neglected,so the coupling effect of wear and corrosion should be sufficiently considered in casing integrity evaluation.

methane cracking is considered a bridge technology between gray and green hydrogen production *** this work an experimental study of methane cracking in molten tin is *** tests were conducted in a quartz reactor(i.d.=1.5 cm,L=20 cm)with capillary injection,varying temperature(950–1070℃),inlet methane flow rate(30–60 mL·min^(-1))and tin height(0–20 cm).The influence of the residence time in the tin and in the headspace on methane conversion and on carbon morphology was *** conversions obtained in tin and in the empty reactor were measured and compared with results of detailed kinetic simulations(CRECK).Furthermore,an expression of a global kinetic constant for methane conversion in tin was also *** highest conversion(65%at Q_(0)=30 mL·min^(-1) and t=1070℃)is obtained for homogeneous gas phase reaction due to the long residence time(70 s),the presence of tin leads to a sharp decrease of residence time(1 s),obtaining a conversion of 35%at 1070℃,thus meaning that tin owns a role in the *** characterization(scanning electron microscopy,Raman)reported a change in carbon toward sheet-like structures and an increase of the carbon structural order in the presence of molten tin media.

Hiding secret data in digital multimedia has been essential to protect the ***,attackers with a steganalysis technique may break *** steganalysis methods have good results with conventional machine Learning(mL)techniques;however,the introduction of Convolutional Neural Network(CNN),a deep learning paradigm,achieved better performance over the previously proposed mL-based *** the existing CNN-based approaches yield good results,they present performance issues in classification accuracy and stability in the network training *** research proposes a new method with a CNN architecture to improve the hidden data detection accuracy and the training phase stability in spatial domain *** proposed method comprises three phases:pre-processing,feature extraction,and ***,in the pre-processing phase,we use spatial rich model filters to enhance the noise within images altered by data hiding;secondly,in the feature extraction phase,we use two-dimensional depthwise separable convolutions to improve the signal-to-noise and regular convolutions to model local features;and finally,in the classification,we use multi-scale average pooling for local features aggregation and representability enhancement regardless of the input size variation,followed by three fully connected layers to form the final feature maps that we transform into class probabilities using the softmax *** results identify an improvement in the accuracy of the considered recent scheme ranging between 4.6 and 10.2%with reduced training time up to 30.81%.

Community batteries(CBs)are emerging to support and even enable energy communities and generally help consumers,especially space-constrained ones,to access potential techno-economic benefits from storage and support local grid ***,the economic viability of CB projects is often *** this regard,typical feasibility studies assess CB value for behind-the-meter(BTm)operation or whole-sale market participation,i.e.,front-of-meter(FOm).This work proposes a novel techno-economic operational framework that allows systematic assessment of the different options and introduces a two-meter architecture that co-optimizes both BTm and FOm benefits.A real CB project application in Australia is used to demonstrate the significant two-meter co-optimization opportunities that could enhance the business case of CB and energy communities by multi-service provision and value stacking.

Cardiovascular diseases are the main cause of morbidity and mortality in the world, and obesity and the metabolic syndrome are risk factors for its development. One of the therapies to reduce cardiovascular risk is the use of polyunsaturated fatty acids. In Brazil, a source of such acid is the oil extracted from the fat of the capybara. The objective of this work is to study the effects of the capybara oil on lipid and glucose metabolism, as well as its effects on the adipose tissue and cardiac remodeling. We assessed the effects of capybara oil treatment on body mass, lipid and carbohydrate metabolism, systolic blood pressure, adipose tissue and cardiac remodeling, and performed an ultrastructural evaluation of the myocardium in C57Bl/6 mice treated with high-fat diet. Treatment with capybara oil reduced total cholesterol and triglyceride levels, systolic blood pressure, visceral and subcutaneous adipose tissue, and adipocyte diameter. In addition, cardiac remodeling was attenuated, preserving cardiomyocytes, increasing vascularization, reducing cardiomyocyte hypertrophy and the extracellular matrix, and preserving the morphological integrity of mitochondria. Capybara oil has several beneficial effects on the cardiovascular and metabolic system, and further studies are needed to better understand its role in the prevention or treatment of cardiovascular diseases.
