Morphological alterations and redox changes associated with hepatic warm ischemia-reperfusion injury

AIM To study the effects of warm ischemia-reperfusion(I/R) injury on hepatic morphology at the ultrastructural level and to analyze the expression of the thioredoxin(TRX)and glutaredoxin(GRX) *** Eleven patients undergoing liver resection were subjected to portal triad clamping(PTC). Liver biopsies were collected at three time points; first prior to PTC(baseline), 20 min after PTC(post-ischemia) and 20 min after reperfusion(post-reperfusion). Electron microscopy and morphometry were used to study and quantify ultrastructural changes, respectively. Additionally, gene expression analysis of TRX and GRX isoforms was performed by quantitative PCR. For further validation of redox protein status, immunogold staining was performed for the isoforms GRX1 and *** Post-ischemia, a significant loss of the liver sinusoidal endothelial cell(LSEC) lining was observed(P = 0.0003) accompanied by a decrease of hepatocyte microvilli in the space of disse. Hepatocellular morphology was well preserved apart from the appearance of crystalline mitochondrial inclusions in 7 out of 11 patients. Postreperfusion biopsies had similar features as post-ischemia with the exception of signs of a reactivation of the LSECs. No changes in the expression of redox-regulatory genes could be observed at mR NA level of the isoforms of the TRX family but immunoelectron microscopy indicated a redistribution of TRX1 within the *** At the ultrastructural level, the major impact of hepatic warm I/R injury after PTC was borne by the LSECs with detachment and reactivation at ischemia and reperfusion, respectively. Hepatocytes morphology were well preserved. Crystalline inclusions in mitochondria were observed in the hepatocyte after ischemia.

The corpus cavernosum after treatment with dutasteride or finasteride： a histomorphometric study in a benign prostatic hyperplasia rodent model

Erectile dysfunction is a common side effect of finasteride and dutasteride treatments. The objective of this study was to investigate the structural changes in the penis using a benign prostatic hyperplasia （BPH） rodent model treated with dutasteride or finasteride. Sixty male rats were divided into the following groups： C, untreated control rats; C ＋ d, control rats receiving dutasteride; C ＋ F, control rats receiving finasteride; H, untreated spontaneously hypertensive rats （SHRs）; H ＋ d, SHRs treated with dutasteride; and H ＋ F, SHRs treated with finasteride. Treatments were performed for 40 days, and penises were collected immediately thereafter. The organs were analyzed using histomorphometric methods to determine the cross-sectional penile area, as well as the surface density （Sv） of smooth muscle fibers, connective tissue, elastic system fibers, and sinusoidal spaces of the corpus cavernosum. The results were compared using a one-way ANOVA with Bonferroni＇s posttest. Groups C ＋ d and C ＋ F had a significantly smaller penile cross-sectional area, but more elastic system fiber Sv compared to Group C. Group C ＋ d showed less smooth muscle Sv, and Group H showed more connective tissue but a smaller sinusoidal space Sv in the corpus cavernosum compared to Group C. Groups H ＋ d and H ＋ F had less smooth muscle Sv than Group H. Group H ＋ d also had more connective tissue and elastic system fiber Sv than Group H. Both dutasteride and finasteride promoted penile modifications in the control rat penis, although this affect was ~reater in Group H animals. In this rodent model, dutasteride was the drug that most affected the corpus cavernosum.

In vitro Safety Evaluation and Anticlastogenic Effect of BacoMind^(TM) on Human Lymphocytes

Objective BacoMind^TM （BM） is a standardized extract of Bacopa monnieri, which belongs to the family Scrophulariaceae and is a creeping annual plant found throughout the Indian subcontinent. It has been used by Ayurvedic medicinal practitioners in India for almost 3000 years and is classified as a medharasayana, a substance which improves memory and intellect. With the widespread traditional use as well as scientific validation of Bacopa monnieri for nootropic activity, a bioactive-rich unique phytochemical composition-BacoMind^TM was developed from B. monnieri for use as a cognition and memory enhancing agent. The present study aimed to investigate the in vitro toxicity of this formulation of BacoMind^TM on human lymphocytes and to rule out its possible contribution to mutagenicity. Methods In the present investigation the active ingredients present in BM were identified and quantified by high performance liquid chromatography （HPLC） and high performance thin-layer chromatography （HPTLC）. Antioxidant and anticlastogenic properties of BM were studied in vitro with and without metabolic activation. doses of BM were chosen on the basis of mitotic index （MI） and cytokinesis-block proliferation index （CBPI）. Clastogenicity assays were performed at 31.2 μg/mL, 62.5 μg/mL, and 125 μg/mL, while the Salmonella reverse mutation assay （Ames test） was performed at doses of 61.72, 185.18, 555.55, 1666.67, and 5000.00 μg/plate. Results HPLC and HPTLC analysis of BM revealed the presence of bacoside A3. bacopaside Ⅰ, bacopaside Ⅱ, jujubogenin isomer of bacopasaponin C, bacosine, luteolin, apigenin, bacosine, and β-sitosterol d glucoside. BM demonstrated significant antioxidant activity. The number of chromosomal aberrations and the frequency of micronuclei induced by BM were not statistically significant up to a dose of 62.5 μg/mL. Numerical modeling of adhesion and adhesive failure during unidirectional contact between metallic surfaces

In this work, we developed a finite element modeling approach to study adhesion during unidirectional contact between a two-dimensional plane-strain square and a flat slab. The surfaces were metallic or ceramic, and we analyzed different pairs of materials and their adhesion intensity using a FORTRAN subroutine (dLOAd) connected to a commercial finite element code Abaqus, which provided the surface attractive forces based on the Lennard-Jones interatomic potential using Hamaker constants. We considered adhesive loads during both the approach and separation of the surfaces. during the separation step, we modeled the material transfer between surfaces due to adhesion with respect to damage initiation and propagation at the flat slab. The parameters considered in the simulations include normal load, chemical affinity, and system size, and we analyzed different conditions by comparing the interaction forces during approach and withdrawal. This work also presents: (i) a description of the evolution of energy dissipation due to adhesion hysteresis, (ii) the formation–growth–breakage process of the adhesive junctions and the material transfer between surfaces, and (iii) an adhesive wear map based on a proposed novel equation that correlates the material parameters and material loss due to adhesion. The results indicate that the chemical affinity between bodies in contact is more related to adhesion than the applied load. In addition, the ratio between the material strength and elastic modulus seems to be an important factor in reducing adhesive wear.

Engagement in clinical learning environment among nursing students: Role of nurse educators

Student engagement in a clinical learning environment is a vital component in the curricula of pre-licensure nursing students, providing an opportunity to combine cognitive, psychomotor, and affective skills. This paper is significant in Arab world as there is a lack of knowledge, attitude and practice of student involvement in the new clinical learning environment. The purpose of this review article is to describe the experiences and perspectives of the nurse educator in facilitating pre-licensure nursing students’ engagement in the new clinical learning environment. The review suggests that novice students prefer actual engagement in clinical learning facilitated through diversity experiences, shared learning opportunities, student-faculty interaction and active learning. They expressed continuous supervision, ongoing feedback, interpersonal relationship and personal support from nurse educators useful in the clinical practice. However, the value of this review lies in a better understanding of what constitutes quality clinical learning environment from the students’ perspective of engagement in evidence-based nursing, reflective practice, e-learning and simulated case scenarios facilitated by the nurse educators. This review is valuable in planning and implementing innovative clinical and educational experiences for improving the quality of the clinical teaching-learning environment.

The Current Status of Stem Cell Regeneration in Intra Oral Applications—A Systematic Review

Aim: 1) To review the literature of various applications of stem cell regeneration in dentistry from 2010 to 2016. 2) To review these studies and to summarize the current status of stem cell regeneration in intra oral applications. 3) To present the available successful data with regard to stem cell regeneration in dentistry and to highlight the future trends. Materials and Methods: Search Protocol: A systematic search was made in the PubMed database with the key words gingiva, papillary reconstruction, periodontal ligament, dental pulp, salivary gland, enamel re-mineralization, dentin, cementum, bone, whole tooth, cleft palate, regeneration and stem cells. All articles from 2010 to 2016, relevant to the topic were included. After the search a total of 1826 articles were obtained which were screened and categorized by three independent reviewers as review papers, in vitro, animal and human studies, pertaining to stem cell regeneration in intra oral applications. On the basis of the extracted data and outcome measures, conclusions were drawn. Results: Maximum number of animal studies has been done to regenerate periodontal tissues, bone, dentin and pulp tissues. Few human studies have been done till date. Although clinically, researchers have been able to regenerate periodontal-like tissue, bone and pulp-like tissue, they still haven’t been able to regenerate tissues that completely resemble tissues in their natural form. Conclusion: The future of stem cell therapy in dental applications looks promising. The predictability and efficacy of outcomes, as well as safety concerns of stem cell therapy is yet to be fully established. Ongoing research and development of newer scaffolds, understanding various signaling molecules and their cues, understanding gene expression and proteomics of stem cells are the future directions that will take us a step forward to achieving successful regeneration.
