Urea-induced interfacial engineering enabling highly reversible aqueous zinc-ion battery

Aqueous zinc-ion batteries（AZIBs） have been regarded as prospective rechargeable energy storage devices because of the high theoretical capacity and low redox potential of Zn metal.However,the uncontrollable formation of dendrites and the water-induced side reactions at the Zn/electrolyte interface,and the poor reversibility under a high current density(> 2 mA·cm-2) and large area capacity(>2 mAh·cm-2) still limit the practical applications of AZIBs.Therefore,a strategy that can overcome these difficulties is urgently needed.Here,we introduce an environmentally friendly and low-cost additive,namely urea,to the electrolyte of AZIBs to induce uniform Zn deposition and suppress the side reactions.Measurements of the adsorption behavior,electrochemical characterization,and observations of the morphology revealed the interfacial modification induced by urea on the Zn/electrolyte interface,demonstrating its huge potential in AZIBs.Consequently,the long-term cycling stability（over2100 h） of a Zn/Zn symmetric cell under a high current density of 5 mA·cm-2and a capacity of 5 mAh·cm-2was achieved with a 1 mol·L-1ZnSO4electrolyte with the urea additive.Additionally,the assembled Zn/NH4V4O10full cell with urea exhibited excellent cycling performance and an outstanding average Coulombic efficiency of 99.98%.These results indicate that this is a low-cost and effective additive strategy for realizing highly reversible AZIBs.

BACKGROUND The self-assembly of solid organs from stem cells has the potential to greatly expand the applicability of regenerative medicine.Stem cells can self-organise into microsized organ units,partially modelling tissue function and regeneration.Dental pulp organoids have been used to recapitulate the processes of tooth development and related diseases.However,the lack of vasculature limits the utility of dental pulp organoids.AIM To improve survival and aid in recovery after stem cell transplantation,we demonstrated the three-dimensional(3D)self-assembly of adult stem cell-human dental pulp stem cells(hDPSCs)and endothelial cells(ECs)into a novel type of spheroid-shaped dental pulp organoid in vitro under hypoxia and conditioned medium(CM).METHODS During culture,primary hDPSCs were induced to differentiate into ECs by exposing them to a hypoxic environment and CM.The hypoxic pretreated hDPSCs were then mixed with ECs at specific ratios and conditioned in a 3D environment to produce prevascularized dental pulp organoids.The biological characteristics of the organoids were analysed,and the regulatory pathways associated with angiogenesis were studied.RESULTS The combination of these two agents resulted in prevascularized human dental pulp organoids(Vorganoids)that more closely resembled dental pulp tissue in terms of morphology and function.Single-cell RNA sequencing of dental pulp tissue and RNA sequencing of Vorganoids were integrated to analyse key regulatory pathways associated with angiogenesis.The biomarkers forkhead box protein O1 and fibroblast growth factor 2 were identified to be involved in the regulation of Vorganoids.CONCLUSION In this innovative study,we effectively established an in vitro model of Vorganoids and used it to elucidate new mechanisms of angiogenesis during regeneration,facilitating the development of clinical treatment strategies.

Insights into the history and tendency of glycosylation and digestive system tumor:A bibliometric-based visual analysis

BACKGROUND Glycosylation,a commonly occurring post-translational modification,is highly expressed in several tumors,specifically in those of the digestive system,and plays a role in various cellular pathophysiological mechanisms.Although the importance and detection methods of glycosylation in digestive system tumors have garnered increasing attention in recent years,bibliometric analysis of this field remains scarce.The present study aims to identify the developmental trends and research hotspots of glycosylation in digestive system tumors.AIM To find and identify the developmental trends and research hotspots of glycosylation in digestive system tumors.METHODS We obtained relevant literature from the Web of Science Core Collection and employed VOSviewer 1.6.19 and CiteSpace(version 6.1.R6)to perform bibliometric analysis.RESULTS A total of 2042 documents spanning from 1978 to the present were analyzed,with the research process divided into three phases:the period of obscurity(1978-1990),continuous development period(1991-2006),and the rapid outbreak period(2007-2023).These documents were authored by researchers from 66 countries or regions,with the United States and China leading in terms of publication output.Reis Celso A had the highest number of publications,while Pinho SS was the most cited author.Co-occurrence analysis revealed the most popular keywords in this field are glycosylation,expression,cancer,colorectal cancer,and pancreatic cancer.Furthermore,the Journal of Proteome Research was the most prolific journal in terms of publications,while the Journal of Biological Chemistry had the most citations.CONCLUSION The bibliometric analysis shows current research focus is primarily on basic research in this field.However,future research should aim to utilize glycosylation as a target for treating tumor patients.

C1M2:a universal algorithm for 3D instance segmentation,annotation,and quantification of irregular cells

Cell instance segmentation is a fundamental task for many biological applications,especially for packed cells in three-dimensional(3D)microscope images that can fully display cellular morphology.Image processing algorithms based on neural networks and feature engineering have enabled great progress in two-dimensional(2D)instance segmentation.However,current methods cannot achieve high segmentation accuracy for irregular cells in 3D images.In this study,we introduce a universal,morphology-based 3D instance segmentation algorithm called Crop Once Merge Twice(C1M2),which can segment cells from a wide range of image types and does not require nucleus images.C1M2 can be extended to quantify the fluorescence intensity of fluorescent proteins and antibodies and automatically annotate their expression levels in individual cells.Our results suggest that C1M2 can serve as a tissue cytometry for 3D histopathological assays by quantifying fluorescence intensity with spatial localization and morphological information.

Essential oil from Saussurea costus inhibits proliferation and migration of Eca109 cells via mitochondrial apoptosis and STAT3 signaling

Objective:To investigate the effect and its underlying molecular mechanisms of essential oil from Saussurea costus in esophageal cancer cell line Eca109.Methods:The chemical composition of essential oil from Saussurea costus was investigated by gas chromatography-mass spectrometry(GC-MS).The anti-proliferative,anti-migrative,and apoptotic effects of essential oil from Saussurea costus against Eca109 cells were analyzed.Moreover,the expression of proteins associated with cell cycle,metastasis,and apoptosis was determined.Results:GC-MS analysis showed that essential oil from Saussurea costus was predominantly comprised of sesquiterpenes.Saussurea costus essential oil inhibited the viability of Eca109 cells in a dose-and time-dependent manner with IC_(50) values of(24.29±1.49),(19.16±2.27)and(6.97±0.86)μg/mL at 12,24,and 48 h,respectively.The expression levels of target proteins in the cell cycle(phase G_(1)/S),including cyclin D1,p21,and p53,were affected by Saussurea costus essential oil.The essential oil also downregulated the expression of metastasis-related proteins MMP-9 and MMP-2.Moreover,it induced apoptosis of Eca109 cells through the mitochondrial pathway,as well as inhibition of STAT3 phosphorylation.Conclusions:The essential oil from Saussurea costus exhibited anti-proliferative,anti-migrative,and apoptotic effects on Eca109 cells,and could be further explored as a potential anti-esophageal cancer agent.

Regulation of TMEM100 expression by epigenetic modification,effects on proliferation and invasion of esophageal squamous carcinoma

BACKGROUND Esophageal squamous cell carcinoma(ESCC)is a prevalent malignancy with a high morbidity and mortality rate.TMEM100 has been shown to be suppressor gene in a variety of tumors,but there are no reports on the role of TMEM100 in esophageal cancer(EC).AIM To investigate epigenetic regulation of TMEM100 expression in ESCC and the effect of TMEM100 on ESCC proliferation and invasion.METHODS Firstly,we found the expression of TMEM100 in EC through The Cancer Genome Atlas database.The correlation between TMEM100 gene expression and the survival of patients with EC was further confirmed through Kaplan-Meier analysis.We then added the demethylating agent 5-AZA to ESCC cell lines to explore the regulation of TMEM100 expression by epigenetic modification.To observe the effect of TMEM100 expression on tumor proliferation and invasion by overexpressing TMEM100.Finally,we performed gene set enrichment analysis using the Kyoto Encyclopaedia of Genes and Genomes Orthology-Based Annotation System database to look for pathways that might be affected by TMEM100 and verified the effect of TMEM100 expression on the mitogen-activated protein kinases(MAPK)pathway.RESULTS In the present study,by bioinformatic analysis we found that TMEM100 was lowly expressed in EC patients compared to normal subjects.Kaplan-meier survival analysis showed that low expression of TMEM100 was associated with poor prognosis in patients with EC.Then,we found that the demethylating agent 5-AZA resulted in increased expression of TMEM100 in ESCC cells[quantitative real-time PCR(qRT-PCR)and western blotting].Subsequently,we confirmed that overexpression of TMEM100 leads to its increased expression in ESCC cells(qRT-PCR and western blotting).Overexpression of TMEM100 also inhibited proliferation,invasion and migration of ESCC cells(cell counting kit-8 and clone formation assays).Next,by enrichment analysis,we found that the gene set was significantly enriched in the MAPK signaling pathway.The involvement of TMEM100 in the regulation of MAPK signaling p

Analogue Black Holes in Reactive Molecules

We show that reactive molecules with a unit probability of reaction naturally provide a simulator of some intriguing black hole physics.The unit reaction at the short distance acts as an event horizon and delivers a one-way traffic for matter waves passing through the potential barrier when two molecules interact by high partialwave scatterings or dipole-dipole interactions.In particular,the scattering rate as a function of the incident energy exhibits a thermal-like distribution near the maximum of the interaction energy in the same manner as a scalar field scatters with the potential barrier outside the event horizon of a black hole.Such a thermal-like scattering can be extracted from the temperature-dependent two-body loss rate measured in experiments on KRb and other molecules.

1.引言《构式语法:英语的结构》(Construction Grammar:The Structure of English)是Thomas Hoffmann 2022年在剑桥大学出版社出版的著作。本书是继Hilpert(2019[2014])之后第二本专门的构式语法教材。Hilpert(2014)打破了构式语法长期没有教材的窘境①,而本书则体现了构式语法近期发展的一些新特点。因此,本文将在与Hilpert(2019[2014])进行对比的基础上重点评介本书的几个突出特点,同时指出其不足之处。

The MYB transcription factor RcMYB1 plays a central role in rose anthocyanin biosynthesis

Rose(Rosa hybrida)is one of most famous ornamental plants in the world,and its commodity value largely depends on its flower color.However,the regulatory mechanism underlying rose flower color is still unclear.In this study,we found that a key R2R3-MYB transcription factor,RcMYB1,plays a central role in rose anthocyanin biosynthesis.Overexpression of RcMYB1 significantly promoted anthocyanin accumulation in both white rose petals and tobacco leaves.In 35S:RcMYB1 transgenic lines,a significant accumulation of anthocyanins occurred in leaves and petioles.We further identified two MBW complexes(RcMYB1-RcBHLH42-RcTTG1;RcMYB1-RcEGL1-RcTTG1)associated with anthocyanin accumulation.Yeast one-hybrid and luciferase assays showed that RcMYB1 could active its own gene promoter and those of other EBGs(early anthocyanin biosynthesis genes)and LBGs(late anthocyanin biosynthesis genes).In addition,both of the MBW complexes enhanced the transcriptional activity of RcMYB1 and LBGs.Interestingly,our results also indicate that RcMYB1 is involved in the metabolic regulation of carotenoids and volatile aroma.In summary,we found that RcMYB1 widely participates in the transcriptional regulation of ABGs(anthocyanin biosynthesis genes),indicative of its central role in the regulation of anthocyanin accumulation in rose.Our results provide a theoretical basis for the further improvement of the flower color trait in rose by breeding or genetic modification.
