Polyurethane-degrading fungi from soils contaminated with rocket propellant and their ability to decompose alkyne terminated polybutadiene with urethane

A large amount of propellant materials are produced every year,and storage and disposal of these propellant materials seriously contributes to environmental pollution.Alkyne terminated polybutadiene with urethane segments(PUPB)is the macromolecule backbone of these propellant materials,and degradation of PUPB is central to the eco-friendly treatment of propellant materials.In this study,we isolated a polyurethane(PU)-and PUPB-degrading fungus from soils contaminated with rocket propellant,and the fungus H14 was identified as Fusarium solani(Mart.)Sacc.based on macro-and micro-morphology as well as phylogenetic analyses.The ability of F.solani H14 to degrade PU film and PUPB patches was evaluated via mass loss,scanning electron microscopy(SEM)and enzyme production ability.Mass loss analyses revealed a 25.8%reduction in mass of PU and 1.3%reduction in mass of PUPB after F.solani H14 was incubated with PU and PUPB for 90 days,respectively.We found that F.solani H14 mycelia significantly colonized both PU and PUPB.SEM images showed that the surface of PU films and PUPB patches formed holes,underwent folding and experienced damage as well as irregular fissuring from the erosion of fungal hypha.Moreover,two possible degradative enzymes,lipase and esterase,were produced by F.solani.Our study opens a new avenue of research for eco-friendly treatments of explosive materials and propellants.This paper represents the first article on the degradation of PUPB patches.

Four hitherto unknown polysubstituted isofiavonoids,including three isofiavans:7,4'-dihydroxy-8,2',3'-trimethoxyisofiavan(1),7,2',4'-trihydroxy-8,3'-dimethoxyisofiavan(2),and 7,2',4'-trihydroxy-5-methoxyisofiavan(3),and one prenylated isofiavone cudraisofiavone M(4)were isolated from the ethanol extracts of Spatholobus suberectus(for 1 and 2),Flemingia macrophylla(for 3),and Cudrania cochinchinensis(for 4),respectively.Their structures were established on the basis of extensive spectroscopic analysis.Compounds 1 and 4 exhibited weak cytotoxic activity against five human cancer cell lines(HL-60,A-549,SMMC-7721,MCF-7,and SW-480).

Rural Tourism Development in China:Principles,Models and the Future

Rural tourism in China,has undergone a rapid development in the last three decades.It is an emerging and effective catalyst that promotes industrial restructuring,agricultural development and the upgrading of rural areas.However,there remains little understanding about the core issues of rural tourism in China:the exact connotation,the principles,the development models and the future directions.This review paper identifies the key issues based on literature analysis,national statistics and press reports to form a general picture of the past,the current and the future prospects of China's rural tourism.The paper firstly addresses the definition,content and principles of rural tourism,and then provides an overview and brief evaluation of the progress and the current development models in China.Finally,based on the analysis of the experiences and problems,the authors sum up with five pathways of the future development of China's rural tourism from the perspective of rural tourism resources,product,market and promotion.

Infectious Behavior in Photo-oxidation of Polymers

When a polymer is used together with others,its aging process will be affected by the adjacent polymers.This infectious behavior between polymers makes the aging process more complex than that of an individual material.In this study,infectious behavior in photooxidation of polymers was investigated.Polypropylenes(PPs),an unstabilized PP and a commercial PP,were chosen as the infection sources.Six typical polymers,high density polyethylene(HDPE),low density polyethylene(LDPE),polystyrene(PS),polycarbonate(PC),poly(ethylene terephthalate)(PET),and polyamide 6(PA6),were used as the targets.The degree of oxidation of the targets was evaluated by attenuated total reflection-Fourier transform infrared spectroscopy(ATR-FTIR).An accelerating effect of two infection sources on the photo-oxidation of the target polymers was observed.Potential infectious agents from the infection sources were detected by pyrolysis-gas chromatography-mass spectrometry(Py-gc-MS)and gas chromatography(gc).The acceleration effect of two main infectious agents,i.***.acetone and acetic acid,on the photo-oxidation of the commercial PP was verified.The infectious effect of the infection source on the target polymer was considered to be a comprehensive result of the effects of a variety of infectious agents.

Expression of nitric oxide synthase and guanylate cyclase in the human ciliary body and trabecular meshwork

Background The role played by the nitric oxide （NO） signaling pathway in the aqueous humor dynamics is still unclear.This study was designed to investigate the expression and distribution of NO synthase （NOS） isoforms and guanylate cyclase （gc） in human ciliary body,trabecular meshwork and the Schlemm＇s canal.Methods Twelve eyes after corneal transplantation were used.Expression of three NOS isoforms （i.***.neuronal NOS （nNOS）,inducible NOS （iNOS） and endothelial NOS （eNOS）） and gc were assessed in 10 eyes by immunohistochemical staining using monoclonal or polyclonal antibody of NOS and gc.Ciliary bodies were dissected free and the total proteins were extracted.Western blotting was performed to confirm the protein expression of 3 NOS isoforms and gc.Results Expression of 3 NOS isoforms and gc were observed in the ciliary epithelium,ciliary muscle,trabecular meshwork and the endothelium of the Schlemm＇s canal.Immunoreactivity of nNOS was detected mainly along the apical cytoplasmic junction of the non-pigmented epithelium （NPE） and pigmented epithelial （PE） cells.Protein expressions of 3 NOS isoforms and gc were confirmed in isolated human ciliary body by Western blotting.Conclusions The expression of NOS isoforms and gc in human ciliary body suggest the possible involvement of NO and cyclic guanosine monophosphate （cyclic GMP,cGMP） signaling pathway in the ciliary body,and may play a role in both processes of aqueous humor formation and drainage.

Degradation analysis of A^2/O combined with AgNO3 + K2FeO4 on coking wastewater

In this work, a coking wastewater was selected and a biochemical A^2/O treatment device for fractional degradation was designed and employed. After each stage of the treatment, the products were analyzed through gas chromatography–mass spectroscopy(gc–MS) to determine their composition. Finally, AgNO_3+ K_2 FeO_4 was used as an advanced deep catalytic oxidation treatment. It was concluded from the analysis that cyclic organics could be degraded and the chemical oxygen demand(COD) was controlled within 50 mg·L^(-1), in line with the target value. Meanwhile, the spectra obtained from the gc–MS were in accordance with the conclusions reached based on the COD. The research results showed that all hard-degradable organics in coking wastewater could be eliminated through the A^2/O bio-membrane treatment and the advanced treatment of making use of K_2FeO_4 as an oxidant and Ag+as a catalyst, the catalytic efficiency with Ag+as a catalyst of K+2FeO_4 was very high. Agcould evidently improve the oxidation capacity of K_2FeO_4 to wastewater in its short stability time, and this is an important innovation.

Kinetic and product studies of Cl atoms reactions with a series of branched Ketones

The rate constants for the C1 atom reaction with three branched ketones have been measured at 298 ± 2 K and 760 Torr using the relative rate method in the absence of NO. The rate constants values obtained （in units of 10^-10 cm^3/（molecule.sec）） are： k（2-methyl-3-pentanone） = 1.07 ± 0.26, k（3-methyl-2-pentanone） =1.21 ± 0.26, and k（4-methyl-2-pentanone） = 1.35 ± 0.27. Combining the chemical kinetic data obtained by this study with those reported for other ketones, a revised Structure Activity Relationship （SAR） parameter and R group reactivity （kR of R（O）R＇ and CHx （x = 1, 2, 3） group reactivity （kCHx） toward C1 atoms were proposed. In addition, the products from the three reactions in the presence of NO were also identified and quantified by using PTR-ToF-MS and gc-FID, and the yields of the identified products are： acetone （39% ± 8%）＋ ethanal （78% ± 12%）, 2-butanone （22% ± 2%）＋ ethanal （75% ± 10%）＋ propanal （14% ± 1%） and acetone （26% ± 3%）＋ 2-methylpmpanal （24% ± 2%）, for C1 atoms reaction with 2-methyl-3-pentanone, 3-methyl-2-pentanone and 4-methyl-2-pentanone, respectively. Based on the obtained results, the reaction mechanisms of C1 atoms with these three ketones are proposed.

Ligation of CD180 inhibits IFN-α signaling in a Lyn-PI3K-BTK-dependent manner in B cells

A hallmark of systemic lupus erythematosus （SLE） is the consistent production of various auto-antibodies by auto-reactive B cells. Interferon-α（IFN-α） signaling is highly activated in SLE B cells and plays a vital role in the antibody response by B cells. Previous studies have shown that CD180-negative B cells, which are dramatically increased in SLE patients, are responsible for the production of auto-antibodies. However, the association between CD180 and IFN-αsignaling remains unknown. In the present study, we explored the effect of CD180 on regulating the activation of IFN-α signaling in B cells. We found that the number of CD180-negative B cells was increased in MRIJMp-Fas（Ipr/Ipr） lupus-prone mice compared with wild-type mice. Phenotypic analysis showed that CD180-negative B cells comprised CD138＋ plasmablast/plasma cells and GL-7＋ germinal center （gc） B cells. Notably, ligation of CD180 significantly inhibited the I FN-α-induced phosphorylation of signal transducer and activator of transcription 2 （STAT-2） and expression of IFN-stimulated genes （ISGs） in a Lyn-PI3K-BTK-dependent manner in vitro. Moreover, ligation of CD180 could also inhibit IFN-α-induced ISG expression in B cells in vivo. Furthermore, the Toll-like receptor 7 and Toll-like receptor 9 signaling pathways could significantly downregulate CD180 expression and modulate the inhibitory effect of CD180 signaling on the activation of I FN-a signaling. Collectively, our results highlight the close association between the increased proportion of CD180-negative B cells and the activation of IFN-α signaling in SLE. Our data provide molecular insight into the mechanism of IFN-α signaling activation in SLE B cells and a potential therapeutic approach for SLE treatment.

Maize MS2 encodes an ATP-binding cassette transporter that is essential for anther development

The anther cuticle and pollen exine play a critical role in male gametophyte development. The sporopollenin precursors and cuticular lipid monomers are transported to the surface of the microspores and the epidermis by lipid transport proteins(LTPs) and ATP-binding cassette G(ABCG) transporters for the formation of the pollen wall and anther cuticle, respectively. However, the function of ABCG transporters in maize anther development is unclear. Here, we cloned the MS2 gene from the maize male sterile2 mutant using map-based cloning and determined that it encodes an ABCG transporter. MS2 protein was experimentally confirmed to be located on the cell membrane. The quantitative real-time PCR(qRT-PCR)results showed that MS2 was ubiquitously expressed in all vegetative and reproductive tissues, whereas a high transcriptional level of MS2 was observed in anthers, especially at the young microspore stage. Gas chromatography-mass spectrometry(gc–MS) analysis showed decreased accumulation of cutin and wax components in ms2 anthers, indicating that MS2 plays a role in the transport of lipid molecules to anther cuticle and pollen exine. To our knowledge, MS2 is the first reported ABCG transporter gene that participates in anther development in maize.

Effect of KIF22 on promoting proliferation and migration of gastric cancer cells via MAPK-ERK pathways

Background:Gastric cancer (gc) is one of the most globally prevalent cancers in the world. The pathogenesis of gc has not been fully elucidated, and there still lacks effective targeted therapeutics. The influence of altered kinesin superfamily protein 22 (KIF22) expression in gc progression is still unclearly. The aim of this study was to investigate the KIF22 effects on gc and related mechanisms.Methods:Gastric carcinoma tissues and matching non-cancerous tissues were collected from patients with gc who have accepted a radical gastrectomy in Lanzhou University Second Hospital from May 2013 to December 2014. The expression of KIF22 was examined in gc of 67 patients and 20 para-carcinoma tissues by immunochemical staining. The relationship between the expression of KIF22 and clinicopathologic characteristics was next investigated in the remaining 52 patients except for 15 patients who did not complete follow-up for 5 years. Cell viability was performed via 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test and colony formation assay in the Mgc-803 and Bgc-823 gc cells. Cell scratch and trans-well invasion assay was performed to assess migration ability in the Mgc-803 and Bgc-823 gc cells. Gene set enrichment analysis (GSEA) pathway enrichment analysis was performed to explore the potential functions. Cell cycle was detected by flow cytometry. In addition, the two gc cell lines were used to elucidate the underlying mechanism of KIF22 in gc in vitro via assessing the effects on mitogenactivated protein kinase and extracellular regulated protein kinases (MAPK/ERK) signal transduction pathway-related expressions by Western blotting assays. The differences were compared by t tests, one-way analysis of variance, and Chi-squared tests. Results:The study showed that KIF22 was up-regulated in gc, and KIF22 high expression was significantly related to differentiation degree ( χ2 = 12.842, P = 0.002) and poorly overall survivals.
