Scanning electrochemical microscopy(SEcM)and scanning vibrating electrode technique(SVET)were used to investigate the electrochemical behaviour of the top surface of the 2098-T351 alloy welded by friction stir welding(FSW).The SVET technique was efficient in identifying the cathodic and anodic weld *** welding joint(WJ),which comprises the thermomechanically affected zone(TMAZ)and the stir zone(SZ),was cathodic relative to the heated affected zone(HAZ)and the base metal(BM).The reactivities of the welding joint at the advancing side(AS)and the retreating side(RS)were analyzed and compared using SEcM technique in the competition mode by monitoring the dissolved oxygen as a redox mediator in 0.005 mol L^-1 Nacl *** RS was more electrochemically active than the AS,and these results were correlated with the microstructural features of the welded alloy.

Objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIc)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MIcs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and LEU255 were present commonly in stabilizing all *** PcA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

The diagnosis of Invasive Fungal Disease (IFD) is difficult because the sensitivity of conventional culture and cytology methods for the detection of Aspergillus is poor, and because serological methods exhibit cross-reactivity with environmental fungi. Fungal polysaccharides are of particular interest in the serologic diagnosis of IFD, especially galactomannan (GM) and glucan, which have been recognized as useful tools for the diagnosis of fungal infections. Further investigation into polysaccharide antibody cross-reactivity is important in improving diagnostic tests. Our aim was to verify the production of antibodies against fungal antigens in mice immunized with exopolysaccharides (EPS) obtained from three fungal species (Aspergillus terreus, Paecilomyces variotii and curvularia brachyspora). ELISA assays were performed to evaluate homologous and heterologous reactions and showed anti-polysaccharide antibodies production. EPS from A. terreus was immunogenic enough to immunize the animal and lead to antibody production against this EPS. In addition, no cross-reactivity was observed with EPS obtained from P. variotii and c. brachyspora. The EPS from P. variotii did not exhibit good immunogenicity, whereas EPS from c. brachyspora showed good immunogenicity, and heterologous reactions were detected on the ELISA assays performed using the EPS from this species. The results indicate that immunization can occur for environmental fungal polysaccharides, generating antibodies may be interfere in serological diagnosis that are based on detection of fungal polysaccharides, mainly glucans and galactomannans.

AIM To assess lactase gene(LcT)-13910c>T polymorphisms in Brazilian non-alcoholic fatty liver disease(NAFLD) and nonalcoholic steatohepatitis(NASH) patients in comparison with healthy *** This was a transverse observational clinical study with NAFLD patients who were followed at the Hepatology Outpatient Unit of the Hospital das clínicas, S?o Paulo, Brazil. The polymorphism of lactase non-persistence/lactase persistence(LcT-13910c>T) was examined by PcR-restriction fragment length polymorphism technique in 102 liver biopsy-proven NAFLD patients(steatosis in 9 and NASH in 93) and compared to those of 501 unrelated healthy volunteers. Anthropometric, clinical, biochemical and liver histology data were analyzed. continuous variables were compared using the t or Mann-Whitney tests, and categorical data were compared with the Fisher's exact test. Univariate logistic regression and multivariate logistic regression adjusted for gender and age were *** No differences in the LcT-13910 genotype frequencies were noted between the NAFLD patients(66.67% of the patients with steatosis were cc, 33.33% were cT, and none were TT; 55.91% of the patients with NASH were cc, 39.78% were cT, and 4.3% were TT; P = 0.941) and the healthy controls(59.12% were cc, 35.67% were cT, and 5.21% were TT) or between the steatosis and NASH patients. That is, the distribution of the lactase non-persistence/lactase persistence polymorphism(LcT-13910c>T) in the patients with NAFLD was equal to that in the general population. In the NASH patients, the univariate analysis revealed that the lactase nonpersistence(low lactase activity or hypolactasia) phenotype was associated with higher insulin levels(23.47 ± 15.94 μU/m L vs 15.8 ± 8.33 μU/m L, P = 0.027) and a higher frequency of insulin resistance(91.84% vs 72.22%, P = 0.02) compared with the lactase persistence phenotype.
