Walking is the most commonly chosen type of physical activity(PA)during pregnancy and provides several health benefits to both mother and *** initiatives have promoted the importance of walking in general,but little emphasis is directed toward pregnant women,the majority of whom are insufficiently *** women face a variety of dynamic barriers to a physically active lifestyle,some of which are more commonly experienced during specific times throughout the pregnancy *** is unique in that it appears resistant to a number of these barriers that limit other types of PA participation,and it can be meaningfully integrated into some transportation and occupational activities when leisure-time options are *** intervention work suggests that walking programs can be effectively adopted into a typical pregnancy ***,a great deal of work remains to administer successful pregnancy walking interventions,including developing and using validated methods of PA and walking *** narrative review discusses the unique advantages of walking during pregnancy,provides recommendations for future intervention work,and outlines the need for pregnancy-focused community walking *** search procedures were followed to determine sources from the literature specific to walking during pregnancy for use in each section of this review.

Cdc42 regulates the cellular localization of Cdc42ep1 in controlling neural crest cell migration

The member of Rho family of small GTPases Cdc42 plays important and conserved roles in cell polarity and motility. The Cdc42ep family proteins have been identified to bind to Cdc42, yet how they interact with Cdc42 to regulate cell migration remains to be elucidated. In this study, we focus on Cdc42epl, which is expressed predominantly in the highly migratory neural crest ceils in frog embryos. Through morpholino-mediated knockdown, we show that Cdc42epl is required for the migration of cranial neural crest cells. Loss of Cdc42epl leads to rounder cell shapes and the formation of membrane blebs, consistent with the observed disruption in actin organization and focal adhesion alignment. As a result, Cdc42ep1 is critical for neural crest cells to apply traction forces at the correct place to migrate efficiently. We further show that Cdc42ep1 is localized to two areas in neural crest celts： in membrane protrusions together with Cdc42 and in perinuciear patches where Cdc42 is absent. Cdc42 directly interacts with Cdc42epl （through the CRIB domain） and changes in Cdc42 level shift the distribution of Cdc42epl between these two subcellular locations, controlling the formation of membrane protrusions and directionality of migration as a consequence. These results suggest that Cdc42ep1 elaborates Cdc42 activity in neural crest cells to promote their efficient migration.

Involvement of CRF2 signaling in enterocyte differentiation

To determine the role of corticotropin releasing factor receptor (CRF2) in epithelial permeability and enterocyte cell *** this purpose, we used rat Sprague Dawley and various colon carcinoma cell lines (SW620, hCT8R, hT-29 and Caco-2 cell lines). Expression of CRF2 protein was analyzed by fluorescent immunolabeling in normal rat colon and then by western blot in dissociated colonic epithelial cells and in the lysates of colon carcinoma cell lines or during the early differentiation of hT-29 cells (ten first days). To assess the impact of CRF2 signaling on colonic cell differentiation, hT-29 and Caco-2 cells were exposed to Urocortin 3 recombinant proteins (Ucn3, 100 nmol/L). In some experiments, cells were pre-exposed to the astressin 2b (A2b) a CRF2 antagonist in order to inhibit the action of Ucn3. Intestinal cell differentiation was first analyzed by functional assays: the trans-cellular permeability and the para-cellular permeability were determined by Dextran-FITC intake and measure of the transepithelial electrical resistance respectively. Morphological modifications associated to epithelial dysfunction were analyzed by confocal microscopy after fluorescent labeling of actin (phaloidin-TRITC) and intercellular adhesion proteins such as E-cadherin, p120ctn, occludin and ZO-1. The establishment of mature adherens junctions (AJ) was monitored by following the distribution of AJ proteins in lipid raft fractions, after separation of cell lysates on sucrose gradients. Finally, the mRNA and the protein expression levels of characteristic markers of intestinal epithelial cell (IEC) differentiation such as the transcriptional factor krüppel-like factor 4 (KLF4) or the dipeptidyl peptidase IV (DPPIV) were performed by RT-PCR and western blot respectively. Kaposiform hemangioendothelioma in children: a benign vascular tumor with multiple treatment options

Background Kaposiform hemangioendothelioma (KhE) is a rare vascular tumor affecting infants and young children. Although benign, it can be associated with an aggressive locally growing tumor and/or a life-threatening Kasabach–Merritt phenomenon (KMP). To date, only reviews of limited cases have been performed. We, therefore, conducted a comprehensive literature search to collect relevant data and make recommendations for future treatment trials. Methods Review of the available literature between 1993 and 2017 revealed a total of 105 publications involving 215 patients of less than 21 years of age. To this, we added 12 from our department and 4 from the Cooperative Weichteilsarkomstudie database. Results We found that KMP was present in 79% of the infants, in 47% of the 1–5-year olds, in 43% of the 6–12-year olds, and in 10% of the 13–21-year-old patients. KMP was present in nearly all (94%) patients with retroperitoneal tumors and in all patients with extra-regional tumors. The median size of a KhE without KMP was 12 cm2 as compared to 49 cm2 when associated with a KMP. With complete (not further classifiable if R0 or R1) resection, all patients were cured. If inoperable, response regarding KMP/regression of tumor size was seen in 29/28% with steroid-, 47/39% with vincristine-, 44/43% with interferon alpha-, 65/61% with anti-platelet agents-, and in 97/100% with sirolimus-containing therapies. Conclusions Patients with progressive KhE should undergo resection whenever it is considered a safe option. If inoperable, sirolimus should be the first choice for treating KMP and reducing tumor size.
