NOD2 and ATG16L1 polymorphisms affect monocyte responses in Crohn's disease

AIM： TO assess whether polymorphisms in NOD2 and ATG16L1 affect cytokine responses and mycobacterium avium subspecies paratuberculosis （MAP） survival in monocytes from Crohn＇s disease （CD） patients METHODS： Monocytes were isolated from peripheral blood of CD patients of known genotype for common single nucleotide polymorphisms of NOD2 and ATG16L1, Monocytes were challenged with MAP and bacterial per- sistence assessed at subsequent time-points. Cytokine responses were assayed using a Milliplex multi-analyte profiling assay for 13 cytokines. RESULTS： Monooltes heterozygous for a NOD2 polymorphism （R702W, P268S, or 1007fs） were more permissive for growth of MAP （P = 0.045） than those without. There was no effect of NOD2 genotype on subsequent cytokine expression. The T300A polymorphism of ATG16L1 did not affect growth of MAP in our model （P = 0.175）, but did increase expression of cytokines interleukin （IL）-10 （P = 0.047） and IL-6 （P = 0.019）. CONCLUSION： CD-associated polymorphisms affected the eliminaUon of MAP from ex v/vo monooltes （NOD2）, or expression of certain oltokines （ATG16LI）, implying independent but contributory roles in the pathogenesis of CD.

Characterization of Cassava Root Husk Powder: Equilibrium, Kinetic and Modeling Studies as Bioadsorbent for Copper(II) and Lead(II)

This study was aimed to investigate Pb(II) and Cu(II) ions removal ability from aqueous solution by cassava root husks (CRH) as a cheap, sustainable and eco-friendly bioadsorbent. The CRH was characterized by Fourier Transform Infrared (FTIR) spectroscopy which indicated the availability of various functional groups for metal coordination and the result was supported by elemental analysis studies. UV-Visible spectral studies indicated the presence of oxalate () and it could possibly interact with metal ions to give rise to a stable chelated coordination complex which affects metal ions removal efficiency. Bioadsorption process was carried out as a function of metal concentration, contact time, pH of the solution, particle size, and dosage of the adsorbent. Experimental results indicated the optimal adsorption condition of pH 4 for both Pb(II) and Cu(II) ions, dosage of 0.1 g/0.1L and 1 g/0.1L for Pb(II) and Cu(II) ions respectively, adsorption equilibrium time of 2 and 25 minutes for Pb(II) and Cu(II) respectively, and concentration of 0.5 mg/L for both metal ions. Kinetic data best fitted pseudo-second-order model and not the pseudo-first-order model. Equilibrium data best fitted the Freundlich model than the Langmuir model. Specific surface area and pore volume studies indicated that CRH is non-porous and hence rapid adsorption kinetics is expected. The popularity of keeping reptiles such as snakes and lizards is ever increasing. The health and welfare of these animals depends on the knowledge and understanding of their environmental and nutritional needs. But where does a new owner of these species obtain such information? This study aims to investigate what information is available in pet shops and stores for the reptile owning public and how well this information is imparted. Our findings show that there is a lot of excellent advice on the husbandry of reptiles in some shops while others offer less advice. It is hoped that through this study areas of downfall on primary knowledge can be recognized so that all shops can aspire to the highest levels of information.

Seasonality of ER Admissions in Northwestern Pennsylvania: A Cross-Sectional Study

Seasonality, in the context of emergency room (ER) admissions, can be described as the periodic incidence of disease, corresponding to seasons, or other pre-established calendar periods. Respiratory diseases, in general, show a seasonal pattern with incidence peak at the winter season, however research still presents a considerable amount of inconsistency. Incidence of cardiovascular diseases (CVD) is also very well linked to the cold season. Gastrointestinal, genitourinary and neurological diseases are poorly studied in regards their seasonal patterns. This study aimed to assess seasonality of the five categories of diseases–respiratory, cardiovascular, gastrointestinal, genitourinary, and neurological-using data from a community hospital in northwestern PA. We analyzed 14 years (2000-2014) of data from the Meadville Medical Center (MMC) ER admissions. For each ER admission case, we had information about ICD-9 code, sex, insurance, race, age and date, time and year of admission. Statistical analyses were performed using SAS 9.4 University version software. We found significantly fewer cases of respiratory diseases in spring (OR = 0.757), summer (OR = 0.579), and fall (OR = 0.741), when comparing to the winter season;however, seasonal differences were not found for cardiovascular, genitourinary, and neurological diseases. The implications of these results will primarily be used to improve Meadville’s public health policies for cold seasons, and more specifically, implement programs that prepare the ER to receive and treat respiratory cases more efficiently in the cold season.

Schizophrenia is a severe mental illness which can have a devastating impact onan individual’s quality of life. Comorbidities are high amongst patients and lifeexpectancy is approximately 15 years less than the general population. Despite thewell-known increased mortality, little is known about the impact of gastrointestinaland liver disease on patients with schizophrenia. We aimed to reviewthe literature and to make recommendations regarding future care. Literaturesearches were performed on PubMed to identify studies related to gastrointestinaland liver disease in patients with schizophrenia. High rates of chronic liverdisease were reported, with Non-Alcoholic Fatty Liver Disease being of particularconcern;antipsychotics and metabolic syndrome were contributing factors. Ratesof acute liver failure were low but have been associated with antipsychotic useand paracetamol overdose. Coeliac disease has historically been linked to schizophrenia;however, recent research suggests that a causal link is yet to be *** is emerging regarding the relationships between schizophrenia andpeptic ulcer disease, inflammatory bowel disease and irritable bowel syndrome;clinical vigilance regarding these conditions should be high. Patients with schizophreniapoorly engage with bowel cancer screening programmes, leading to latediagnosis and increased mortality. Clozapine induced constipation is a significantissue for many patients and requires close monitoring. There is a significantburden of gastrointestinal and liver disease amongst patients with *** levels of support from all members of the medical team are essential toensure that appropriate, timely care is provided.

Background： The broiler industry has undergone intense genetic selection over the past 50 yr. resulting in improvements for growth and feed efficiency, however, significant variation remains for performance and growth traits. Production improvements have been coupled with unfavourable metabolic consequences, including immunological trade-offs for growth, and excess fat deposition. To determine whether interactions between fatty acid（FA） metabolism and innate immunity may be associated with performance variations commonly seen within commercial broiler flocks, total carcass lipid %, carcass and blood FA composition, as wel as genes involved with FA metabolism, immunity and cel ular stress were investigated in male birds of a broiler strain, layer strain and F1 layer × broiler cross at d 14 post hatch. Heterophil：lymphocyte ratios, relative organ weights and bodyweight data were also ***： Broiler bodyweight（n = 12） was four times that of layers（n = 12） by d 14 and had significantly higher carcass fat percentage compared to the cross（n = 6; P = 0.002） and layers（P = 0.017） which were not significantly different from each other（P = 0.523）. The carcass and whole blood FA analysis revealed differences in the FA composition between the three groups indicating altered FA metabolism, despite al being raised on the same diet. Genes associated with FA synthesis andβ-oxidation were upregulated in the broilers compared to the layers indicating a net overal increase in FA metabolism,which may be driven by the larger relative liver size as a percentage of bodyweight in the broilers. Lentil(Lens culinaris Medik.), a diploid(2n = 14) with a genome size greater than 4000 Mbp, is an important cool season food legume grown worldwide. The availability of genomic resources is limited in this crop species. The objective of this study was to develop polymorphic markers in lentil using publicly available curated expressed sequence tag information(ESTs). In this study, 9513 ESTs were downloaded from the National Center for Biotechnology Information(NCBI) database to develop unigene-based simple sequence repeat(SSR) markers. The ESTs were assembled into 4053 unigenes and then analyzed to identify 374 SSRs using the MISA microsatellite identification tool. Among the 374 SSRs, 26 compound SSRs were *** pairs for these SSRs were designed using Primer3 version 1.14. To classify the functional annotation of ESTs and EST–SSRs, BLASTx searches(using E-value 1 × 10-5) against the public UniP rot(http://***/) and NCBI(http://***/) databases were performed. Further functional annotation was performed using PLAZA(version3.0) comparative genomics and GO annotation was summarized using the Plant GO slim category. Among the synthesized 312 primers, 219 successfully amplified Lens DNA. A diverse panel of 24 Lens genotypes was used to identify polymorphic markers. A polymorphic set of 57 markers successfully discriminated the test genotypes. This set of polymorphic markers with functional annotation data could be used as molecular tools in lentil breeding.
