Pseudomonas aeruginosa can cause persistent infections,such as biofilm infections,in cystic fibrosis patients,which are difficult to cure due to non-growing persister bacteria that are not effectively killed by the current *** antibiotic activity against growing *** is well documented,their activity against non-growing stationary phase cultures is less ***,we evaluated six major classes of antibiotics,including cell wall and cell membrane inhibitors,protein synthesis inhibitors,DNA synthesis inhibitors,RNA synthesis inhibitors,sulfa drugs and nitrofurantoin,for their activity against growing and non-growing *** foundthat cell wall and cell membrane inhibitors(cefuroxime and colistin),DNA synthesis inhibitors(clinafloxacin)and sulfa drugs(sulfamethoxazole)had good activity against stationary-phase bacteria,while protein synthesis inhibitors(gentamicin),RNA synthesis inhibitor(rifampin)and nitrofurantoin showed relatively poor *** was the only drug able to completely eradicate stationary-phase bacteria within four *** cefuroxime+gentamicin+clinafloxacin combination was able to kill all bacteria from a biofilm within two days,whereas the clinically used drug combination cefuroxime+gentamicin/colistin only partially killed the *** amurine persistent cystic fibrosis lung infectionmodel,only the cefuroxime+gentamicin+clinafloxacin drug combination eradicated all bacteria from the lungs,whereas clinafloxacin alone,cefuroxime+clinafloxacin or the currently recommended drug combination cefuroxime+gentamicin failed to do *** complete eradication is a property of the clinafloxacin combination,as the otherwise identical levofloxacin combination did not clear the bacterial loads from the *** findings offer new therapeutic options for more effective treatment of persistent *** infections,with possible implications for treating other persistent infections.

Targeted genotyping for the prediction of celiac disease autoimmunity development in patients with type 1 diabetes and their family members

BACKGROUND Patients with type 1 diabetes (T1D) and their first-degree relatives (FDRs) have an increased risk of developing celiac disease (CD) compared to the general population. This is largely explained by the shared association with major histocompatibility class II human leukocyte antigen (hLA) DQ2 and/or DQ8 between the two disease states. AIM To describe the frequency of CD autoimmunity (CDA) and the distribution of hLA and haptoglobin genotypes in patients with T1D and their FDRs. Additionally, we aimed at identifying predictors associated with an increased risk of developing CDA in patients with T1D and their family members. METhODS We obtained clinical information and blood samples from 1027 participants (302 with T1D and 725 FDRs) over a five-year period. Samples were tested for autoantibodies associated with CD, hLA-DQ alleles, and haptoglobin *** fit univariate and multiple logistic regression models for CDA separately for subjects with T1D and for FDRs of subjects with T1D. RESULTS Implementation of a screening program increased the frequency of CDA by 2- fold in participants with T1D and 2.8-fold in their FDRs. Multivariate analysis found that, in participants with T1D, having both DR7-DQ2 and DR4-DQ8 was associated with an increased frequency of CDA. In FDRs of T1D patients, reported CD in the family was associated with an increased frequency of CDA during screening. haptoglobin 2 genotype was not associated with developing CDA in the multivariate analysis. CONCLUSION Patients with T1D and their FDRs have a high frequency of CDA. Carrying both DR7-DQ2 and DR4-DQ8 was associated with development of CDA in patients with T1D.

Effect of Rye and Mix Cover Crops on Soil Water and Cotton Yield in a humid Environment

In recent years, the use of cover crops is becoming a popular technology among growers in many regions of the United States, which is expected to deliver various benefits such as improving soil health, increasing soil organic matter, controlling weeds, and helping conserve soil water and nutrients. Although expecting these benefits seems reasonable, it is challenging to know how much of these benefits to expect under specific situations. The potential effect of cover crops on soil water conservation is especially significant because of the documented impact of soil water on crop yield, especially for dryland cropping systems. Some researchers have found that planting a cover crop tended to increase soil water, while others have reported the opposite effect. Information on the impact of cover crops on soil water in cotton (Gossypium hirsutum L.) production systems in South Carolina is currently lacking. Therefore, the objective of this study was to quantify the effect of cover crops on soil water and cotton yield. A field experiment was conducted in South Carolina during winter, spring, and summer of 2015, with three cover crop treatments. The treatments included: 1) rye (Secale cereale L.), planted alone;2) a mix of six cover crop species;and 3) a control treatment with no-cover. The cover crop was established in the winter, terminated in the spring, and cotton was grown during the summer. Soil water was measured at different depths using capacitance probes and a neutron probe. Our results showed no significant differences in soil water and cotton yield among the cover crop treatments. These results suggest that under the humid conditions of this study, any short-term effect of the cover crop on soil water was masked by timely rain.

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor ILT2(synonyms:LIR1,MIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(MhC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of MhC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with MhC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTLs)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTL responses,the engineered high affinity ILT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTLs and could have potential for the development of novel autoimmunity therapies.

＜正＞The National Academies of Sciences 2007 report,"Toxicity Testing in the 21st Century:A Vision and A Strategy" envisions toxicity testing using human cells in vitro to evaluate perturbations of cellular response networks and coupling results from these assays with computational systems biology and in vitro-in vivo extrapolations （IVIVE） to complete human safety assessments. To support this transition in toxicity testing,our laboratory is evaluating key cell response networks to develop proof of concept safety assessments for a series of prototype compounds and networks,including the DNA damage response *** DNA damage toxicity pathway project studies the p53-mediated DNA damage stress response in human cells to determine the underlying response circuitry for the p53 pathway and the dose response behavior for pathway activation after chemical induced DNA damage. This research effort has three overarching goals:（1） map the key determinants of cellular fate following DNA damage induced by chemicals with different mechanisms of action（indirect vs. direct DNA-damage）,（2） identify dose-dependent thresholds associated with cellular adaptation and toxicity（mutation） and（3） perform a risk assessment for a prototype chemical based on predicted regions of safety determined from in vitro *** work involved validation of the cell model and collection of a dense data stream comprised of dose response data at the gene, protein（p53,p-p53,p-h2AX,MDM2,p21, WIP1）,and cellular（cell cycle arrest,apoptosis, micronucleus;MN）level using prototype chemicals for DNA damage:etoposide（topoisomeraseⅡinhibition）,methylmethane sulfonate（DNA alkylation） and quercetin（oxidative damage）.18 point dose-response curves were generated for protein and cell fate endpoints in a p53 competent cell line（hT1080）using flow cytometry and high content *** genome transcriptomic analysis was also performed for each prototype chemical at several doses ranging from concentrations with no e

Anti-hepatitis B virus(hBV)therapy leads to the emer- gence of mutant viral strains during the treatment of chronic hepatitis B with nucleos(t)ides analogues. The existence of hBV variants with primary antiviral resistance may be important for treatment choice. We studied two patients with chronic hBV infection by sequencing the hBV polymerase *** had adefovir-and tenofovir-related mutations in the viral polymerase,although they had never been treated.
