Small ubiquitin-like modifier （SUMO） conjugation affects a broad range of processes in plants, including growth, flower initiation, pathogen defense, and responses to abiotic stress. Here, we investigate in vivo and in vitro a SUMO conjugating enzyme with a Cys to Ser change in the active site, and show that it has a dominant negative effect. In planta expression significantly perturbs normal development, leading to growth retardation, early flowering and gene expression changes. We suggest that the mutant protein can serve as a probe to investigate sumoylation, also in plants for which poor genetic infrastructure precludes analysis via loss-of-function mutants.

Diseases and health complications caused by mineral deficiencies afflict billions of people globally. Developing pulse crops with elevated seed mineral concentrations can contribute to reducing the incidence of these deficiencies. The objectives of this study were to estimate variance components conditioning seed mineral concentrations of chickpea and lentil grown in Washington and Idaho, determine correlations between different mineral concentrations and between mineral concentrations and yield, 100-seed weight, and days to flowering, and compare seed mineral concentrations between chickpeas and lentils grown in adjacent plots. Genotype effects, although significant in chickpea and lentil for all minerals except selenium, tended to be minimal compared to location, year, and their interaction effects. In both chickpeas and lentils high positive correlations were observed between seed concentrations of phosphorus and potassium, phosphorus and zinc, and potassium and zinc. Correlations between mineral concentration and yield, and mineral concentration and days to 50% flowering were similar for chickpeas and lentils across the majority of minerals. These results may reflect similarities between the two crops in physiological processes for mineral uptake and partitioning. Lentils had higher concentrations of iron and zinc than chickpea when the two crops were grown in adjacent plots,whereas chickpeas had higher concentrations of calcium and manganese. Plant genotypes that are more efficient at obtaining minerals from growing environments will be useful as parental materials to develop improved chickpea and lentil cultivars that have good yield potential coupled with high seed mineral concentrations.

Whole genome phylogeny of Gallus: introgression and data-type effects

Background: Previous phylogenetic studies that include the four recognized species of Gallus have resulted in a number of distinct topologies, with little agreement. Several factors could lead to the failure to converge on a consistent topology, including introgression, incomplete lineage sorting, different data types, or insufficient ***: We generated three novel whole genome assemblies for Gallus species, which we combined with data from the published genomes of Gallus gallus and Bambusicola thoracicus(a member of the sister genus to Gallus). To determine why previous studies have failed to converge on a single topology, we extracted large numbers of orthologous exons, introns, ultra-conserved elements, and conserved non-exonic elements from the genome assemblies. This provided more than 32 million base pairs of data that we used for concatenated maximum likelihood and multispecies coalescent analyses of ***: All of our analyses, regardless of data type, yielded a single, well-supported topology. We found some evidence for ancient introgression involving specific Gallus lineages as well as modest data type effects that had an impact on support and branch length estimates in specific analyses. However, the estimated gene tree spectra for all data types had a relatively good fit to their expectation given the multispecies ***: Overall, our data suggest that conflicts among previous studies probably reflect the use of smaller datasets(both in terms of number of sites and of loci) in those analyses. Our results demonstrate the importance of sampling large numbers of loci, each of which has a sufficient number of sites to provide robust estimates of gene trees. Low-coverage whole genome sequencing, as we did here, represents a cost-effective means to generate the very large data sets that include multiple data types that enabled us to obtain a robust estimate of Gallus phylogeny.

Knowledge of the higher-level phylogenetic relationships of birds has grown substantially during the past two decades due to the application of genomic ***,the nomenclature of higher-level taxa has not become more stable,due to the lack of regulation of taxon names above the level of superfamily by the ICZN,and the usage of rank-based nomenclature,which is not tied to clades in a *** of regulation and the instability of rank-based nomenclature impede effective communication among *** review support for higher-level avian clades using a set of 10 phylogenomic data sets,and identify clades that are supported by congruency of at least four of *** provide formal definitions of the names of these clades based on the rules of the recently published *** names of 25 clades are here defined using minimum-crown-clade(n=23),minimum-clade(n=1)and maximum-crown-clade(n=1)*** new names are introduced here:Dinocrypturi,Pteroclimesites,Musophagotides,Phaethoquornithes and *** also review diagnostic apomorphies of the relevant clades,and identify known synonyms and *** establishing a formal link between higher-level taxon names and well-supported phylogenetic hypotheses,our phylogenetic definitions will provide a solid basis for the stabilization of avian higher-level nomenclature.

Due to the need for energy conservation in buildings and the simultaneous benefit of cost savings, the development of a low firing rate load modulating residential oil burner is very desirable. One of the two main requirements of such a burner is the development of a burner nozzle that is able to maintain the particle size distribution of the fuel spray in the desirable (small) size range for efficient and stable combustion. The other being the ability to vary the air flow rate and air distribution around the fuel nozzle in the burner for optimal combustion at the current fuel firing rate. In this paper, which deals with the first requirement, we show that by using pulse width modulation in the bypass channel of a commercial off-the-shelf bypass nozzle, this objective can be met. Here we present results of spray patterns and particle size distribution for a range of fuel firing rates. The results show that a desirable fuel spray pattern can be maintained over a fuel firing rate turndown ratio (Maximum Fuel Flow Rate/Minimum Fuel Flow Rate) of 3.7. Thus here we successfully demonstrate the ability to electronically vary the fuel firing rate by more than a factor of 3 while simultaneously maintaining good atomization.

A Comparison Study of the Efficacy of Rapid Titration Quetiapine and Haloperidol in Agitated Adults in an Emergency Setting

Objective: Intramuscular (IM) Benzodiazepines and/or Haloperidol alone or with benzodiazepines are frequently used to treat agitation. Based on emerging literature regarding Quetiapine used for the control of anxiety we examined Quetiapine as a possible alternative in selected cases. Methods: This study was a single-blind randomized study comparing Quetiapine PO (300 mg) with a combination of Haloperidol (5 mg), Benztropine mesylate (2 mg) and lorazepam (2 mg) administered IM to treat agitated patients seeking care in a busy psychiatric emergency setting. Male or female patients (18 - 60), deemed by the attending (admitting) psychiatrist to be indicative of agitated and/or aggressive behavior and had a Positive and Negative Syndrome Score-Excited Component (PANSS-EC), as evaluated by the Research Psychiatrist, and total score equal to or greater than 15. Patients deemed competent were randomized into one of the following treatment groups: Quetiapine 300 or Haloperidol 5 mg, benztropine mesylate, or lorazepam given by the IM route. Two scales, PANSS-EC and CGI-C were used to assess patients in the trial. The primary outcome measure PANSS-EC at 2 hours after administration of the medication. Results: Sixty-eight patients were included in the study. There were no significant treatment group differences in baseline condition. There was no significant difference between the two conditions. There was, however, a significant within-group decrease from baseline condition. Conclusion: Finding no significant differences suggests that in general the two treatments were equivalent. To sum up, Quetiapine 300 mg as a single dose appeared safe and effective in agitated patients treated in an ER. The results were similar to a comparison group receiving an intra-muscular combination of Haloperidol, Lorazepam and Benztropine. This study has significant limitations. The study was single blind and the use of a placebo would have strengthened the design but would be considered unethical

Heavy metal pollution from both anthropogenic and natural processes can have significant effect on environmental quality of stream and river systems. However, in Ghana, heavy metal pollution of waterbodies is attributed mainly to mining activities but the role of natural mechanisms in altering stream water and sediment quality in relation to heavy metals has received little attention. Spatial and temporal variation in water quality parameters and heavy metal concentrations in water and sediments were studied comparatively in a river and two streams in a gold-rich watershed impacted by heavy mining activities. Samples were collected monthly over a twelve-month period from November 2010 to October 2011 from upstream (unmined) and downstream (mined) sections of the studied streams. Parameters measured include temperature, dissolved oxygen, conductivity, pH, turbidity, colour, mercury (Hg) and arsenic. High spatial variability of water quality parameters was found. Hg concentrations in water were extremely low in both upstream and downstream areas. Maximum geochemical background levels of Hg in unmined pristine areas were 2.45 mg/g whilst arsenic was 29.10 mg/g. By contrast, gold-mined downstream areas recorded Hg and arsenic concentrations of 8.75 mg/g and 82.53 mg/g in stream sediments respectively. Levels of Hg and arsenic in sediments were several orders of magnitude greater than concentrations in surface water in downstream sections and this may be explained by substances originating from mining activities, upstream transport or remobilized sedimented materials in the overlying water column. Our study showed that both natural and human activities may contribute to heavy metal pollution in the highly mineralized watershed of the Pra River Basin. Human factors are however likely to amplify the natural background levels of heavy metals.

Runoff is an important component of the water balance of agricultural fields. Accurate measurement or estimation of agricultural runoff is important due to its potential impact on water quantity and quality. Since runoff from agricultural fields is sporadic and is often associated with irrigation and/or intense rainfall events, manually measuring runoff and collecting water samples for water quality analysis during runoff events is inconvenient and impractical. In the fall of 2017, a field site was selected at the Clemson University Edisto Research and Education Center with the objective of developing, constructing, and testing an Internet of things (IoT) flume system to automatically measure runoff and collect water samples. In 2018, an automatic IoT system was developed and installed consisting of six stainless steel H-flumes (22.9-cm), which measured runoff from six adjacent research plots under two different cultural regimes (cover crop and no cover crop). An electronic eTape sensor was installed in the flume and used to measure the water level or the flume’s head. Open-source electronic (Arduino) devices and a cloud-based platform were then used to create a wireless sensor network and IoT system to automatically record the amount of runoff (hydrograph) coming from each section, collect water samples and transmit the data to a Cloud server (***) where the data can be viewed remotely in real-time. The IoT flume system has been operating successfully and reliably for more than two years.
