AIM: To investigate adenoviral transduction in mesenchymal stem cells(MSCs) and effects on stemness in vitro and function as a cell therapy in ***: Bone marrow-derived adult and fetal MSC were isolated from an equine source and expanded in monolayer tissue culture. Polyethylenimine(PeI)-mediated transfection of pc DNA3-e GFP or adenoviral transduction of green fluorescent protein(GFP) was evaluated in fetal MSCs. Adenoviral-mediated transduction was chosen for subsequent experiments. All experiments were carried out at least in triplicate unless otherwise noted. Outcome assessment was obtained by flow cytometry or immunohystochemistry and included transduction efficiency, cell viability, stemness(i.e., cell proliferation, osteogenic and chondrogenic cell differentiation), and quantification of GFP expression. Fetal and adult MSCs were then transduced with an adenoviral vector containing the gene for the bone morphogenic protein 2(BMP2). In vitro BMP2 expression was assessed by enzyme linked immunosorbent assay. In addition, MSC-mediated gene delivery of BMP2 was evaluated in vivo in an osteoinduction nude mouse quadriceps model. New bone formation was evaluated by microradiography and ***: PeI provided greater transfection and viability in fetal MSCs than other commercial chemical reagents. The thermal structure of subduction zones exerts a major influence on deep-seated mechanical and chemical processes controlling arc magmatism, seismicity, and global element cycles. Accretionary complexes exposed inland may comprise tectonic blocks with contrasting pressureetemperature(Pe T)histories, making it possible to investigate the dynamics and thermal evolution of former subduction interfaces. With this aim, we present new Lue Hf geochronological results for mafic rocks of the Halilbag?Complex(Anatolia) that evolved along different thermal gradients. Samples include a lawsoniteeepidote blueschist, a lawsoniteeepidote eclogite, and an epidote eclogite(all with counter-clockwise Pe T paths),a prograde lawsonite blueschist with a "hairpin"-type Pe T path, and a garnet amphibolite from the overlying sub-ophiolitic metamorphic sole. equilibrium phase diagrams suggest that the garnet amphibolite formed at w0.6 -0.7 GPa and 800 -850℃, whereas the prograde lawsonite blueschist records burial from 2.1 GPa and 420℃ to 2.6 GPa and 520℃. Well-defined Lue Hf isochrons were obtained for the epidote eclogite(92.38 ± 0.22 Ma) and the lawsoniteeepidote blueschist(90.19 ± 0.54 Ma),suggesting rapid garnet growth. The lawsoniteeepidote eclogite(87.30 ± 0.39 Ma) and the prograde lawsonite blueschist(ca. 86 Ma) are younger, whereas the garnet amphibolite(104.5 ± 3.5 Ma) is *** data reveal a consistent trend of progressively decreasing geothermal gradient from granulite-facies conditions at ~104 Ma to the epidote-eclogite facies around 92 Ma, and the lawsonite blueschist-facies between 90 Ma and 86 Ma. Three Lue Hf garnet dates(between 92 Ma and 87 Ma) weighted toward the growth of post-peak rims(as indicated by Lu distribution in garnet) suggest that the HP/LT rocks were exhumed continuously and not episodically. Lekking males aggregate to attract females and contribute solely to egg fer- tilization, without any further parental care. evolutionary theory therefore predicts them to be nonchoosy toward their mates, because any lost mating opportunities would out- weigh the benefits associated with such preferences. Nevertheless, due to time costs, the production of energetically costly sexual displays, and potential sperm limitation, the mat- ing effort of lekking males is often considerable. These factors, combined with the fact that many females of varying quality are likely to visit leks, could favor the evolution of male mate preferences. Here, we show that males of the lekking lesser wax moth, Achroia grisella, were indeed more likely to mate with heavier females in choice experiments, even at their virgin mating （i.e., when their reproductive resources have not yet been depleted by previous matings）. This differential female mating success could not be attributed to female behavior as heavy and light females showed similar motivation to mate （i.e., la- tency to approach the males） and time to copulate. Males seem to benefit from mating with heavier females, as fecundity positively correlated with female mass. This new em- pirical evidence shows that male mate choice may have been tmderestimated in lekking species

The member of Rho family of small GTPases Cdc42 plays important and conserved roles in cell polarity and motility. The Cdc42ep family proteins have been identified to bind to Cdc42, yet how they interact with Cdc42 to regulate cell migration remains to be elucidated. In this study, we focus on Cdc42epl, which is expressed predominantly in the highly migratory neural crest ceils in frog embryos. Through morpholino-mediated knockdown, we show that Cdc42epl is required for the migration of cranial neural crest cells. Loss of Cdc42epl leads to rounder cell shapes and the formation of membrane blebs, consistent with the observed disruption in actin organization and focal adhesion alignment. As a result, Cdc42ep1 is critical for neural crest cells to apply traction forces at the correct place to migrate efficiently. We further show that Cdc42ep1 is localized to two areas in neural crest celts： in membrane protrusions together with Cdc42 and in perinuciear patches where Cdc42 is absent. Cdc42 directly interacts with Cdc42epl （through the CRIB domain） and changes in Cdc42 level shift the distribution of Cdc42epl between these two subcellular locations, controlling the formation of membrane protrusions and directionality of migration as a consequence. These results suggest that Cdc42ep1 elaborates Cdc42 activity in neural crest cells to promote their efficient migration.

Decidual natural killer （dNK） cells express an array of activation receptors to regulate placental immunity and development during early pregnancy. We investigated the functional character of human dNK cells during the first and second trimester of gestation and the interaction between dNK and trophoblast cells. Although the frequency of CD56＋CD16-dNK among the total CD45＋ leukocytes did not change over this period, the expression of the activating receptors, NKp80 and NKG2D, was greatly upregulated. We observed a significantly higher number of extravillous trophoblast cells in proximity to the dNK cells in the first trimester in comparison with the second trimester decidua. NKG2D expression by first trimester dNK cells was decreased when co-cultured with the HTR-8 trophoblast cell line. In the second trimester, functional markers of dNK activation, i.e., angiogenic factor production （e.g., vascular endothelial growth factor, interleukin-8, interferon-gamma）, remained stable despite an increase in NKp80 or NKG2D surface expression. Furthermore, the degranulation capacity of dNK cells, as assessed by CD107a, was decreased in the second trimester. We suggest that in the first trimester, trophoblast-dNK interactions generate a population of dNK cells with a suppressed activating phenotype. In the second trimester, the loss of trophoblast-dNK interactions led to the inhibition of dNK cell function, although their activating receptor expression was increased. We speculate that during pregnancy, two mechanisms operate to modulate the dNK cell activation：suppression of activating receptor levels in the first trimester by trophoblasts and disengagement of receptor-ligand coupling in the second trimester.

Frontiers of Literary Studies in China-Selected Publications from Chinese Universities 2012年第4期6卷 472-489页

This article explores propaganda and self-portrayals among women rulers in seventh and eighth century Tang China, a unique era in which court politics were dominated by female leaders. I analyze the way in which these leaders themselves wished to be rhetorically constructed, the images and allusions with which they desired to be figured, and the way in which they were rhetorically reconstructed by later writers after their deaths. I focus on the theme of auspiciousness--in particular, the definition of the ＂natural＂ in relation to gender identity and power. Female imagery is deployed in late seventh- and early eighth-century works to create the image of a particular brand of far-reaching, generative power possessed and/or desired by the leaders of the time. Beyond revealing the images and allusions with which the female power-holders wished to hear themselves be described and exalted, and what occasions were deemed worthy of exalting, these works offer a fascinating counterpoint to materials which retroactively defame this image. The rhetorical strategies and images later used to delegitimize and denigrate the power of these women often represent opposite treatments of themes present in the court literature from the Zhou-Jinglong era. This paper argues that reconstructions of these women＇s identities as female power-holders indicate the prerogative of later writers to reshape their images in accordance with their own judgments, conceptualizations, and fears of female power.
