Plant Phosphatidylcholine-Hydrolyzing Phospholipases C NPC3 and NPC4 with Roles in Root Development and Brassinolide Signaling in Arabidopsis thaliana

Phosphatidylcholine-hydrolyzing phospholipase C （PC-PLC） catalyzes the hydrolysis of phosphatidylcholine （PC） to generate phosphocholine and diacylglycerol （DAG）. PC-PLC has a long tradition in animal signal transduction to generate DAG as a second messenger besides the classical phosphatidylinositol splitting phospholipase C （PI-PLC）. Based on amino acid sequence similarity to bacterial PC-PLC, six putative PC-PLC genes （NPC1 to NPC6） were identified in the Arabidopsis genome. RT-PCR analysis revealed overlapping expression pattern of NPC genes in root, stem, leaf, flower, and silique. In auxin-treated PNPc3：GUS and PNPc4：GUS seedlings, strong increase of GUS activity was visible in roots, leaves, and shoots and, to a weaker extent, in brassinolide-treated （BL） seedlings. PNPc4：GUS seedlings also responded to cytokinin with increased GUS activity in young leaves. Compared to wild-type, T-DNA insertional knockouts npc3 and npc4 showed shorter primary roots and lower lateral root density at low BL concentrations but increased lateral root densities in response to exogenous 0.05-1.0 I^M BL BL-induced expression of TCH4 and LRX2, which are involved in cell expansion, was impaired but not impaired in repression of CPD, a BL biosynthesis gene, in BL-treated npc3 and npc4. These observations suggest NPC3 and NPC4 are important in BL-mediated signaling in root growth. When treated with 0.1 I^M BL, DAG accumulation was observed in tobacco BY-2 cell cultures labeled with fluorescent PC as early as 15 min after application. We hypothesize that at least one PC-PLC is a plant signaling enzyme in BL signal transduction and, as shown earlier, in elicitor signal transduction.

Objective:To determine the new M-superfamily conotoxins from molluscivorous snail Conus bandanus in ***:Conus bandanus venom was fractionated and purified on HPLC system with an analytical reversed-phase C18 column in order to screen small *** primary structure of peptide was analyzed by matrix-assisted laser desorption/ionization time of flight tandem mass spectrometry using collision-induced dissociation and confirmed by Edman’s degradation ***:Five new conotoxins were biochemically characterized from the crude venom of the mollusk-hunting cone snail Conus bandanus,which were collected at Ke Ga reef of the Nha Trang Bay(Vietnam).Each conotoxin had 15 or 16 amino acid residues and shared the same characteristic cysteine framework V as–CC–C–C–CC–.They were termed as Bn3 b,Bn3 c,Bn3 d,Bn3 e and Bn3 f following the conotoxins ***:The conotoxins Bn3 b,Bn3 e,and Bn3 f are categorized in the mini-M conotoxins of the M1 branch,while conotoxins Bn3 c and Bn3 d are categorized in the mini-M conotoxins of the M2 *** homological analysis reveals that these conotoxins could serve as promising probe compounds for voltage-gated sodium channels.

High tacrolimus intra-patient variability is associated with graft rejection,and de novo donor-specific antibodies occurrence after liver transplantation

AIM To investigate the role of tacrolimus intra-patient variability(IPV) in adult liver-transplant *** We retrospectively assessed tacrolimus variability in a cohort of liver-transplant recipients and analyzed its effect on the occurrence of graft rejection and de novo donor-specific antibodies(dn DSAs), as well as graft survival during the first 2 years posttransplantation. Between 02/08 and 06/2015, 116 patients that received tacrolimus plus mycophenolate mofetil(with or without steroids) were included. reSULTS Twenty-two patients(18.5%) experienced at least one acute-rejection episode(BPAR). Predictive factors for a BPAR were a tacrolimus IPV of > 35% [OR = 3.07 95%CI(1.14-8.24), P = 0.03] or > 40% [OR = 4.16(1.38-12.50), P = 0.01), and a tacrolimus trough level of 35% [OR = 4.83, 95%CI(1.39-16.72), P = 0.01] and > 40% [OR = 9.73, 95%CI(2.65-35.76), P = 0.001] were identified as predictors to detect dn DSAs. IPV did not impact on patient-or graft-survival rates during the follow-up. CONCLUSION Tacrolimus-IPV could be a useful tool to identify patients with a greater risk of graft rejection and of developing a de novo DSA after liver

A Posteriori Stabilized Sixth-Order Finite Volume Scheme with Adaptive Stencil Construction:Basics for the 1D Steady-State Hyperbolic Equations

We propose an adaptive stencil construction for high-order accurate finite volume schemes a posteriori stabilized devoted to solve one-dimensional steady-state hyperbolic *** accuracy(up to the sixth-order presently)is achieved,thanks to polynomial recon-structions while stability is provided with an a posteriori MOOD method which controls the cell polynomial degree for eliminating non-physical oscillations in the vicinity of *** supplemented this scheme with a stencil construction allowing to reduce even further the numerical *** stencil is shifted away from troubles(shocks,discontinuities,etc.)leading to less oscillating polynomial *** on linear,Burgers',and Euler equations,we demonstrate that the adaptive stencil technique manages to retrieve smooth solutions with optimal order of accuracy but also irregular ones without spurious ***,we numerically show that the approach allows to reduce the dissipation still maintaining the essentially non-oscillatory behavior.

Potential Use of a Poultry Manure Digestate as a Biofertiliser:Evaluation of Soil Properties and Lactuca sativa Growth

Anaerobic digestion is a process that is widely used for the treatment of organic wastes. The digestate can be used as a soil amendment or crop fertiliser. The aims of our work were to evaluate 1) the physicochemical composition and pathogen content in a digestate from poultry manure, according to international regulations, and 2) the effect of its soil application on the major chemical and biological soil properties and on the growth of Lactuca sativa. The experiment consisted of two groups of pots(with and without crop). Treatments applied to each group were as follows: low and high doses of digestate and inorganic fertiliser, and no application(control)(low dose: 70 kg nitrogen(N) ha^(-1) and 21 kg phosphorus(P) ha^(-1); high dose: 210 kg N ha^(-1) and 63 kg P ha^(-1)). Soil samples were taken 7 and 34 d(harvest) after treatment applications. Heavy metal and pathogen contents in the digestate were below the upper limit values. Despite the high pH and electrical conductivity values of the digestate, both soil parameters presented acceptable values for crop growth. Although there were no initial increases in total inorganic N and available P in soil with digestate application, an increase in the fresh weight of crop was observed with the high dose application. This is probably associated with the slow nutrient release from the digestate during the development of the crop. Changes in the microbial community were temporary and occurred at the initial sampling stage of the experiment.

The mission of nuclear safety authorities in national radioactive waste disposal programmes is to ensure that people and the environment are protected against the hazards of ionising radiations emitted by the *** implies the establishment of safety requirements and the oversight of the activities of the waste management organisation in charge of implementing the *** Belgium,the safety requirements for geological disposal rest on the following principles:defence-in-depth,demonstrability and the radiation protection principles elaborated by the International Commission on Radiological Protection(ICRP).Applying these principles requires notably an appropriate identification and characterisation of the processes upon which the safety functions fulfilled by the disposal system rely and of the processes that may affect the system ***,research and development(R&D)on safety-relevant thermo-hydro-mechanical-chemical(THMC)issues is important to build confidence in the safety *** paper points out the key THMC processes that might influence radionuclide transport in a disposal system and its surrounding environment,considering the dynamic nature of these *** nature and significance are expected to change according to prevailing internal and external conditions,which evolve from the repository construction phase to the whole heatingecooling cycle of decaying waste after *** these processes have a potential impact on safety,it is essential to identify and to understand them properly when developing a disposal concept to ensure compliance with relevant safety *** particular,the investigation of THMC processes is needed to manage *** includes the identification and characterisation of uncertainties as well as for the understanding of their safety-relevance.R&D may also be necessary to reduce uncertainties of which the magnitude does not allow demonstrating the safety of the disposal system.

In order to assess the dynamics of rivers,a reliable characterization of bedload transport particularly during unsteady flow regimes is *** contrast to highly energetic cases in hillslope areas,we aim to answer the question whether the usage of acoustic measurements can improve the characterization of bedioad in small rivers draining low land mountains with comparatively low water discharge and *** addition to the investigation of natural flood events,controlled floods were generated by releasing water from a reservoir into a small gravel-bed *** controlled releases allow for an evaluation of bedioad solely from channel storage or bank *** acoustical in-situ characterization of bedioad transport,hydrophones were mounted onto the bottom side of steel plates,thus recording the impacts of sediments via the acoustic vibrations on the surface of the plates while at the same time minimizing the disturbing noise resulting from water *** bedioad traps are removable boxes with open lids fixed in the riverbed so that bedioad material registered by the hydrophone is *** acoustic signals correlate well with the quantity of the transported *** summer flood events the highest transport rates occur at the beginning of the rising limb featuring clockwise *** is due to the rising transport energy of the flow and the presence of loose,unconsolidated *** typical winter flood events bedioad shows anticlockwise *** intensification of bedioad conveyance after the runoff peak can be explained by a decreasing stability of the bed material from the beginning to the end of a transport *** behavior also results from a combination of bedioad exhaustion in the vicinity of the monitoring station with a delayed arrival of new material from distal sources later in the hydrograph.

Effect of radial heat conduction on effective thermal conductivity of carbon nanotube bundles

The effect of the radial heat conduction on the effective thermal conductivity of carbon nanotube(CNT) bundles is studied by the nonequilibrium molecular dynamics(NEMD) method. The hexagonal CNT bundle consists of seven(10, 10) single-walled carbon nanotubes(SWCNTs). The radial heat conduction is induced by creating the vacancy defects in some segments of the constituent CNTs. Combined with the temperature differences and the inter-tube thermal resistances at the different segments,the radial heat flow in the CNT bundle is calculated. The maximum percentage of the radial heat flow is less than 7% with the presence of four defective CNTs, while the resultant decrement of the effective thermal conductivity of the bundle is about 18%.The present results indicate that the radial heat flow can significantly diminish the axial heat conduction in the CNT bundles,which probably explains the smaller effective thermal conductivity in the CNT assemblies compared to that of the individual CNTs.
