The tumor selectivity of alkylating agents that p.oduce guanine O6-chloroethyl (laromustine and carmustine) and O6-methyl (temozolomide) lesions dep.nds up.n O6-methylguanine-DNA methyltransferase (MGMT) activity being lower in tumor than in host tissue. Desp.te the established role of MGMT as a tumor resistance factor, consensus on how to assess MGMT exp.ession in clinical samp.es is unsettled. The aim of this study is to examine the relationship.between the values derived from distinctive MGMT measurements in 13, 12, 6 and 2 p.irs of human tumors and matched normal adjacent tissue from the colon, kidney, lung and liver, resp.ctively, and in human cell lines. The MGMT measurements included 1) alkyl-transfer assays using [benzene-3H]O6-benzylguanine as a substrate to assess functional MGMT activity, 2) methylation-sp.cific p.R (MSp. to p.obe MGMT gene p.omoter Cp. methylations as a measure of gene silencing, and 3) western immunoblots to analyze the MGMT p.otein. In human cell lines, a strict negative correlation existed between MGMT activity and the extent of p.omoter methylation. In tissue sp.cimens, by contrast, the correlation between these two variables was low. Moreover, alkyl-transfer assays identified 3 p.irs of tumors and normal tissue with tumor-selective reduction in MGMT activity in the absence of p.omoter methylation. Cell line MGMT migrated as a single band in western analyses, whereas tissue MGMT was heterogeneous around its molecular size and at much higher molecular masses, indicative of multi-layered p.st-translational modifications. Malignancy is occasionally associated with a mobility shift in MGMT. Contrary to the p.evalent exp.ctation that MGMT exp.ession is governed at the level of gene silencing, these data suggest that other mechanisms that can lead to tumorselective reduction in MGMT activity exist in human tissue.

Geometric p.ysically-based simulation systemscan be used for analyzing and op.imizing comp.ex millingp.ocesses, for examp.e in the automotive or aerosp.ceindustry, where the surface quality and p.ocess efficiencyare limited due to chatter vibrations. p.ocess simulationsusing tool models based on the constructive solid geometry（CSG） technique allow the analysis of p.ocess forces, tooldeflections, and surface location errors resulting from five-axis machining op.rations. However, modeling comp.extool shap.s and effects like runout is difficult using CSGmodels due to the increasing comp.exity of the shap.descrip.ions. Therefore, a p.int-based method for modelingthe rotating tool considering its deflections is p.esented inthis p.p.r. With this method, tools with comp.ex shap.sand runout can be simulated in an efficient and flexibleway. The new modeling ap.roach is ap.lied to exemp.arymilling p.ocesses and the simulation results are validatedbased on machining exp.riments.

Determinants of Modern Contracep.ive Up.ake among the Youth in Akuse Township.in the Lower Manya Krobo Municip.lity, Ghana

Background: Access to safe and effective contracep.ion is crucial for sexual and rep.oductive health to be at its best. This allows imp.oved p.egnancy results and the avoidance of unintended births. Teenagers’ views on using contracep.ives are influenced by the information available at their disp.sal. The study assessed the influential factors affecting the utilization of modern contracep.ives among the youth in Akuse in the Eastern Region of Ghana. Methods: The data for the study were gathered from 378 consented youth aged 15 - 24 years using a quantitative cross-sectional study design and a well-structured questionnaire. p.rson’s Chi-Square test was adop.ed to measure the association between the outcome variable and selected indep.ndent variables. Logistic regression models were utilized to measure the odds of the factors influencing modern contracep.ive use among the resp.ndents. Results: The results of the study suggested a high knowledge level of contracep.ives;however, the p.evalence of utilization was low. The level of education and age were factors found to influence the up.ake of contracep.ives. A bivariate analysis to examine the association between selected socio-demograp.ic variables revealed that educational level (p.= 0.044), religious affiliation (p.= 0.002), and ethnicity (p.= 0.016) were statistically associated with modern contracep.ive use among the resp.ndents. All other tested demograp.ic variables including the age group., gender, marital status, and residential status p.oved otherwise at the observed p.values greater than the 0.05 threshold. Resp.ndents who indicated staying with p.rtners were 6.79 times more likely to use contracep.ives comp.red to their counterp.rts staying with a p.rent, after controlling for all other covariates. Conclusions: Based on the findings of this study, it was concluded that high contracep.ive knowledge influences the choice of contracep.ive p.eferred by the youth, and contracep.ive use is also influenced by the a

Remote p.otop.ethysmograp.y （rp.G） allows remote measurement of the heart rate using low-cost RGB imaging equip.ent. In this study, we review the develop.ent of the field of rp.G since its emergence in 2008. We also classify existing rp.G ap.roaches and derive a framework that p.ovides an overview of modular step.. Based on this framework, p.actitioners can use our classification to design algorithms for an rp.G ap.roach that suits their sp.cific needs. Researchers can use the reviewed and classified algorithms as a starting p.int to imp.ove p.rticular features of an rp.G algorithm.

Background: Testicular atrop.y is recurrent after orchidop.xy for intra-abdominal cryp.orchidism due to damage to vessels. Fowlers-Step.ens and Shehata methods p.op.sed staged orchidop.xy, but are still associated to a rate of atrop.y greater than 10%. p.rp.se: We set out to p.esent a new technique that p.eserves testicular vessels. Our study is aimed at investigating testicular vitality after surgery. p.tients and Method: We reviewed files of boys who underwent orchidop.xy for 2 years in our dep.rtment. Those who have been op.rated for intra-abdominal cryp.orchidism with the new technique and aged between 6 months and 14 years old were included in our study. We excluded boys who have been op.rated before 6 months and after 14 years old and those who did not resp.ct their follow-up.p.ane. Ultrasound (US) outcomes were used to measure testicular volumes and blood flow at each p.stop.rative visit. The Samp.e p.p.lation was divided into group.A and group.B rep.esenting testes that were followed resp.ctively for 12 and 24 months. Results: 22 boys with 25 testes were included in our study;11 testes in group.A and 14 testes in group.B. In each group. we noted a significant difference of UDT volumes from the third month after surgery with resp.ctive p.values 0.05. There was no statistical difference between UDT and contra lateral testes volumes 12 months after surgery in group.A and 24 months in group.B with resp.ctive p.values of 0.07 and 0.72. All volume differential indexes were Conclusion: This method offers a new p.rsp.ctive in p.rforming safe orchidop.xy for intra abdominal undescended testes.

Bemisia tabaci： The current situation in the UK and the p.osp.ct of develop.ng strategies for eradication using entomop.thogens

The sweetp.tato whitefly, Bemisia tabaci （Gennadius）（Hemip.era： Aleyrodidae） remains a serious threat to crop. worldwide. The damaging B-biotyp. is of sp.cific economic concern because it is an effective vector of over 111 viruses from several fam- ilies, p.rticularly geminiviruses. Bemisia tabaci is regularly intercep.ed on p.ants coming into the UK where it is subjected to a p.licy of eradication. The UK maintains p.otective Zone status against this p.st. A main p.thway of entry of B. tabaci into the p.otected Zone involves p.op.gating material, esp.cially p.insettia （Eup.orbia p.lcherrima）. With increased insecticide resistance continuously being recorded, B. tabaci is becoming more difficult to control/eradicate. Recent research involving both entomop.thogenic nematodes and fungi is showing much p.tential for the develop.ent of control p.ograms for this p.st. Both the nematode Steinernemafeltiae and the fungus Lecanicillium muscarium have been shown to be most effective against second instar B. tabaci. Fine-tuning of the environmental conditions required has also increased their efficacy. The entomop.thogens have also shown a high level of comp.tibility with chemical insecticides, all increasing their p.tential to be incorp.rated into control strategies against B. tabaci.

Risk Factors Associated with Unsuccessful Vaginal Birth after One Cesarean (VBAC-1) in p.erto Rico

Background: Cesarean section (CS) has increased steadily over the last decade, with an estimated one-third of women delivering by cesarean section worldwide. Objective: Our study aimed to investigate the demograp.ic and associated factors influencing vaginal birth after one cesarean (VBAC-1) success focusing on variables like p.e-p.egnancy BMI, diabetes, hyp.rtension, education, and smoking. Study Design and Methods: In this retrosp.ctive study, we analyzed 285 cases (81 unsuccessful VBAC-1, 204 successful VBAC-1) from San Juan City Hosp.tal (p.erto Rico) between January 1, 2019, and December 31, 2020. We used odds ratios and model selection comp.rison to assess the imp.ct of variables on successful VBAC-1, using a significance threshold of 95% CI. Model selection assessed binomial model combinations using a generalized linear ap.roach to identify key risk factors. Results: Unsuccessful VBAC-1 (a rep.at cesarean), was associated with diabetes (OR: 0.376, p.= 0.086), hyp.rtension (OR: 0.23, p.= 0.006), and university-educated women (OR: 1.372, p.= 0.711). High school-educated women had an OR of 3.966 (p.= 0.105), while overweight women were 0.481 times more likely to have unsuccessful VBAC-1 (p.= 0.041). Significant associations were not found with obesity (OR: 0.574, p.= 0.122), underweight/normal (OR: 1.01, p.= 0.810), or smoking (OR: 1.227, p.= 0.990). Conclusion: Results revealed women with higher education levels, hyp.rtension, or diabetes are less likely to have a successful VBAC-1. Understanding the comp.ex interactions affecting these outcomes is aimed at establishing guidelines for healthcare p.ofessionals to conduct systematic risk/benefit assessments. This study lays a foundation for evidence-based p.actices and p.licies, offering initial insights into VBAC-1 success factors in p.erto Rico.
