Comparison of Biolistic and Agrobacterium-mediated Transformation Methods on Transgene Copy Number and Rearrangement Frequency in Rice

Transferring foreign DNA into plant cells by biolistic and Agrobacterium _mediated methods may result in random integration of different copy numbers of the transgene, and different proportions of intact vs. rearranged copies of the transgene. This may, in turn, affect transgene expression levels. To test the above hypothesis, we first introduced the same plasmid, pAc1PG_CAM, into rice (Oryza sativa L.) calli separately by the biolistic method and by the Agrobacterium _mediated method. To show whether different plasmids may affect the results, we also introduced pTOK233 by the Agrobacterium _mediated method and pJPM44 by the biolistic method. Transgene expression of R0 plants was monitored by histochemical analysis of GUS activity. Transgene copy number was determined by Southern blot analysis after digesting genomic DNA with an enzyme that has a unique cutting site within the input plasmid. The total genomic DNA was also digested by a two_cut enzyme (the cuts are located at two sides of a given transgene expression cassette), followed by Southern blotting analysis, for determining the number of intact transgene expression cassettes. Our data showed that Agrobacterium _mediated transformation resulted in lower transgene copy number (average between 2.1 and 2.3) in transgenic rice plants, compared with those plants obtained by the biolistic method (average between 4.2 and 5.6). The frequency of DNA rearrangement in expression cassettes is lower in transgenic rice plants obtained by the Agrobacterium _ mediated method than those obtained by the biolistic method. The average rearrangement frequency is 0.07 to 0.106 for the Agrobacterium _mediated method, and 0.57 to 0.66 for the biolistic method. Our results suggest that it is better to compare the number of intact expression cassettes instead of the total copy number of the transgene in demonstrating their influence on the level of transgene expression. This research was undertaken for the evaluation of soil erosion using the semi-distributed basin scale SWAT model for four subcatchments of the Dhrabi River Catchment(DRC),which is located in the Pothwar Plateau *** subcatchments(catchment-25 and-31)are characterized by gullies while the other two(catchment-27 and-32)are managed with terraced landuse *** performance of the model was satisfactory with coefficient of determination(R^(2))=0.67 to 0.91 and Nash-Sutcliffe efficiency(ENS)=0.54 to 0.85 for both surface runoff and sediment yield during the calibration(2009-2010)and validation(2011)*** PUSLE factor was found to be the most sensitive parameter during model *** was observed that all of the rainfall-runoff events occurred during the monsoon season(June to September).The estimated annual sediment loss ranged from 2.6 t/hm^(2) to 31.1 t/hm^(2) over the duration of the simulation period for the non-terraced catchments,in response to annual precipitation amounts that were between 194.8 mm to 579.3 *** contrast,the predicted annual sediment levels for the terraced catchments ranged from 0.52 t/hm^(2) to 10.10 t/hm^(2) due to similar precipitation *** terraced catchments resulted in 4 to 5 times lower sediment yield as compared to non-terraced *** results suggest that there is a huge potential for terraces to reduce soil erosion in the DRC specifically and Pothwar area generally,which have proven to be an efficient approach to establishing soil and water conservation structures in this region.

In accordance with the enhancement for luminous efficiency improving, LED (Light Emitting Diode) has been gradually developed by combining the characteristics of small volume, impact resistance, good reliability, long life, low power consumption with multiple purposes for energy saving and environmental protection. Therefore, the array LED has been widely applied in human livings nowadays. This study applies the finite element analysis software ANSYS to analyze the thermal behavior of the array power LED work lamp which is modeled by four same-size LED with MCPCB (Metal Core Print Circuit Board) mounted on a base heat-sink. The Flotran heat flow analysis is applied to obtain the natural convection of air coefficient, while the convection value can be confirmed by the iterative method since it is set as the boundary condition for ANSYS thermal analysis to obtain the temperature distribution, accordingly the chip junction temperature and the base heat-sink temperature were followed through experiments in order to check if the simulation results meet the design requirements and coincide with the power LED product design specification. Prior to the optimal design process for chip junction temperature, the most significant parameters were first chosen by the fractional factorial design. The regressive models were respectively setup by the dual response surface method (RSM) and the mixed response surface method. Furthermore, the genetic algorithm combined with response surface method was applied to acquire the optimal design parameters, and the results were obtained from both methods, which are reviewed for comparison. Afterwards, the mixed response surface method is adopted to investigate the effects of interactions among various factors on chip junction temperature. In conclusion, it is found that the thermal conductivity of MCPCB and the height of base heat-sink are the two major significant factors. 本研究中 ,构建了含有编码绿色荧光蛋白的改进型基因质粒pJPM5。用基因枪法分别把pJPM5和另一带有绿色荧光蛋白基因的质粒pSBG70 0转入水稻TNG6 7愈伤组织。用South ern杂交法证实了转基因的存在 ,而且表明多数转基因植株含有 1到 8个拷贝的转基因。取 2个月的转基因植株上的叶片用于分析绿色荧光蛋白基因表达。用SLM - 80 0 0荧光分析仪定量测定绿色荧光蛋白。多数转基因植株具有很高的绿色荧光蛋白信号。虽然水稻植株有少量自发荧光 ,但是绿色荧光蛋白基因表达出的绿色荧光蛋白信号比植株的自发荧光强得多 ,其测定不会受自发荧光的太大影响。在荧光显微镜下观察到了绿色荧光蛋白基因的表达。借助观察分析绿色荧光蛋白基因的瞬时表达 ,本研究还发现基因枪法转化中 ,如果两枪的气压为90 0psi& 135 0psi,比两枪的气压都为 90 0psi或者 135 0psi更好 ,因其能使质粒进入更多的细胞。研究结果表明 ,绿色荧光蛋白基因可以作为水稻 (甚至小麦、玉米 )转基因研究中的报告基因。研究还显示 ,MAR序列能明显增强绿色荧光蛋白基因的表达能力 (这一结果在另文讨论 ) .

In this paper,the numerical simulation method is used to study the ballistic performances of hourglass lattice sandwich structures with the same mass under the vertical incidence of *** is paid to elucidating the influences of rod cross-section dimensions,structure height,structure layer,and rod inclination angle on the deformation mode,ballistic performances,and ability to change the ballistic direction of *** results show that the ballistic performances of hourglass lattice sandwich structures are mainly affected by their structural *** this respect,structural parameters optimization of the hourglass lattice sandwich structures enable one to effectively improve their ballistic limit velocity and,consequently,ballistic performances.

A resilience-incorporated risk assessment framework is proposed and demonstrated in this study to manifest the advantageous seismic resilience of precast concrete frame(PCF)structures with“dry”connections in terms of their low damage and rapid *** framework integrates various uncertainties in the seismic hazard,fragility,capacity,demand,loss functions,and post-earthquake *** this study,the PCF structures are distinguished from ordinary reinforced concrete frame(RCF)structures by characterizing multiple limit states for the PCF based on its unique damage ***,probabilistic story-wise pushover analyses are performed to yield story-wise capacities for the predefined limit *** the seismic resilience analysis,a step-wise recovery model is proposed to idealize the functionality recovery process,with separate considerations of the repair and non-repair *** recovery model leverages the economic loss and downtime to delineate the stochastic post-earthquake recovery curves for the resilience loss *** such,contingencies in the probabilistic post-earthquake repairs are incorporated and the empirical judgments on the recovery parameters are largely *** proposed framework is demonstrated through a comparative study between two“dry”connected PCFs and one RCF designed as alternative structural systems for a prototype *** results from the risk quantification indicate that the PCFs show reduced loss hazards and lower expected losses relative to the ***,the PCF equipped with energy dissipation devices at the“dry”connections largely reduces the expected economic loss,downtime,and resilience loss by 29%,56%,and 60%,respectively,compared to the RCF.

Tropical cyclones typically form over warm tropical and subtropical ocean surfaces, as their formation and intensification rely primarily on the energy provided by the ocean. Pioneering research by Gray (1968) revealed that the formation of tropical cyclones requires the sea surface temperature (SST) to be no less than 26.5℃. Consequently, SST is commonly recognized as a crucial parameter among the environmental factors influencing tropical cyclone formation(Gray, 1979;Emanuel and Nolan, 2004).
