Folic acid protects against fluoride-induced oxidative stress and testicular damage in rats

Objective:To investigate the effects of folic acid on testicular oxidative damage in sodium fluoride-induced male Wistar rats.Methods:A total of 24 male Wistar rats were divided into 4 groups:the control,sodium fluoride(fed with 100 mg/L sodium fluoride through drinking water orally for 21 days),folic acid(36μg/kg body weight/day,orally),and sodium fluoride plus folic acid(received similar dose orally)groups.At the end of 21 days,epididymal sperm parameters,biochemical analysis of testicular tissue,and serum hormonal levels were performed along with histopathological studies.Results:Sodium fluoride intoxication resulted in marked reduction in gonado somatic index,serum luteinizing hormone,and testosterone level along with 3β-hydroxysteroid dehydrogenase and 17β-hydroxysteroid dehydrogenase activities.In addition,reduction in sperm density,as well as loss of sperm motility and sperm viability,were also observed.Besides,increased levels of testicular malondialdehyde,nitrite,interleukin-6 and tumor necrosis factor-α as well as decreased levels of superoxide dismutase and catalase activities and reduced glutathione content were found to be associated with this toxicity.Folic acid co-treatment,on the other hand,could prevent all the sodium fluoride-induced testicular pathophysiology and oxidative stress related parameters.Histological examinations of testicular sections from the experimental rats supported these results.Conclusions:Combining all,this study suggests that being an antioxidant,folic acid plays a beneficial role against fluoride-induced adverse effects on the male reproductive system.

The leaf extracts of Camellia sinensis (green tea) ameliorate sodium fluoride-induced oxidative stress and testicular dysfunction in rats

Objective:To investigate the effect of Camellia(C.)sinensis in mitigating oxidative damage and reproductive toxicity in testis induced by sodium fluoride in a rat model.Methods:Twenty-four adult male Wister rats were divided into 4 groups,with 6 rats in each group.Group 1 orally received distilled water(1 mL/100 g body weight)daily and served as the control group,while group 2 received drinking water with 100 ppm sodium fluoride per day for 21 consecutive days,group 3 was administered with only C.sinensis extract by gavage at a dose of 100 mg/kg body weight and group 4 received drinking water with 100 ppm sodium fluoride and 100 mg/kg body weight C.sinensis leaf extract per day for 21 consecutive days.At the end of the treatment,the rats were sacrificed under light ether anesthesia.The gonado-somatic index,sperm count and motility,serum level of luteinizing hormone and testosterone were assayed.Lipid peroxidation[malondialdehyde(MDA)level],nitric oxide(NO)production,and activities of antioxidant enzymes-superoxide dismutase(SOD),catalase,and reduced glutathione level(GSH)were also analysed.Results:Sodium fluoride treatment significantly decreased gonado-somatic index,sperm count and motility as well as the serum level of luteinizing hormone and testosterone(P<0.05).The histological examination of testes revealed atrophy and degenerative changes in several seminiferous tubules,along with enhanced interstitial space and a reduced number of Leydig cells.There was a highly significant increase in NO and MDA production(P<0.05),while SOD,catalase activities and GSH level decreased significantly(P<0.05).However,C.sinensis significantly restored testicular weight,sperm parameter,hormonal level(P<0.05),and also reversed MDA and NO generation and antioxidant enzymes activities in the testicular tissue(P<0.05).Conclusions:C.sinensis may have an ameliorative role against sodium fluoride-induced oxidative damage in the testis probably because of its antioxidant property.

Numerical simulation of square shaped particle sedimentation

Direct numerical simulations of square particle sedimentation in a viscous incompressible fluid are presented to examine the effects of sharp edges for various particle-to-fluid density ratios and different initial inclination angles.The settling process exhibits four different motion regimes;non-oscillatory,non-uniform centered oscillatory,uniform off-centered oscillatory,and non-uniform off-centered(irregular)oscillatory.At moderate density ratios,we observe that the rotational motion of settling square particle varies with the initial inclination angle.We employ Dynamic Time Warping and Fast Fourier Transform to analyze translational and angular velocities.Oscillating systems are described using phase-space plots,and a chaotic phenomenon is observed at a higher density ratio.

TALYS calculation and a short review of the experimental status of proton capture studies on p-nuclei:A guide to future investigation

TALYS calculations were performed to obtain the theoretical proton capture cross-sections on the p-nuclei.A short review on the status of related experimental studies was also conducted.Some basic properties such as Q-values,Coulomb barrier,Gamow peak,Gamow Window,and decay properties of the parent and daughter nuclei were studied.Various experimental parameters,e.g.,beam energy,beam current,targets,and detectors,used in experimental investigations reported in the literature,were tabulated.The results of the TALYS calculations in the Gamow region were compared with the corresponding experimental values wherever available.This study is expected to facilitate the planning of future experiments.

We consider a one-dimensional reaction-diffusion equation describing single-and two-species population dynamics in an advective environment,based on the modeling frameworks proposed by Lutscher et al.in 2006.We analyze the effect of rate of loss of individuals at both the upstream and downstream boundaries.In the single-species case,we prove the existence of the critical domain size and provide explicit formulas in terms of model parameters.We further derive qualitative properties of the critical domain size and show that,in some cases,the critical domain size is either strictly decreasing over all diffusion rates,or monotonically increasing after first decreasing to a minimum.We also consider competition between species differing only in their diffusion rates.For two species having large diffusion rates,we give a sufficient condition to determine whether the faster or slower diffuser wins the competition.We also briefly discuss applications of these results to competition in species whose spatial niche is affected by shifting isotherms caused by climate change.

Over the past 20 years,the incidence and prevalence of type 2 diabetes mellitus(T2DM)in children and adolescents have increased,particularly in racial and ethnic minorities.Despite the rise in T2DM in children and adolescents,the pathophysiology and progression of disease in this population are not clearly understood.Youth-onset T2DM has a more adverse clinical course than is seen in those who develop T2DM in adulthood or those with T1DM.Furthermore,the available therapeutic options are more limited for children and adolescents with T2DM compared to adult patients,mostly due to the challenges of implementing clinical trials.A better understanding of the mechanisms underlying the development and aggressive disease phenotype of T2DM in youth is important to finding effective prevention and management strategies.This review highlights the key evidence about T2DM in children and adolescents and its current burden and challenges both in clinical care and research activities.

Geochemistry of volcanic glass from Mahanadi offshore region,eastern continental margin of India:Constraints on the contribution of latest Toba super-eruption

The tephra layers in multiple sediment cores from the offshore region of the Mahanadi basin in the northern Bay of Bengal were investigated for possible volcanic sources. The glass shards from those tephra layers were studied for size distribution, texture, and elemental geochemistry to establish chronostratigraphic markers for regional and global Quaternary correlation. The textural features of fine-grained(silty) volcanic glasses suggest the distal source of these tephra deposits. Major element composition with elevated SiO_(2) contents ranging between75%–76% and dominance of K_(2)O(> 4.5%) over CaO(< 0.9%) suggest ashes have originated from siliceous rhyolitic melts, similar to the petrographic composition of tephra from the Toba volcano. The bulk trace element compositions of the same glass shards were comparable with those reported in the youngest Toba tephra reported elsewhere. Likewise, the LREE-dominated chondrite normalized REE profiles of tephra from the Mahanadi basin closely resemble the characteristic REE patterns in Toba ash from other parts of the Indian Ocean and thus confirmed the contribution of the youngest Toba super-eruption for this ash layers.

Novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan induces robust apoptosis on human acute lymphoblastic leukemia cells through a RONS-subsisted mitochondria-dependent pathway

Objective:To explore the therapeutic effects of the novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan(A.hygrometricus)on human acute lymphoblastic leukemia cells.Methods:Extensive screening of the antiproliferative and chemopreventive potential of different extracts from 5 wild mushrooms,A.hygrometricus,Phallus sp.,Lentinus sp.,Tricholoma sp.,and Serpula sp.was performed against a panel of 6 cancer cell lines and normal cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Apoptosis determination,cell cycle profiling,intracellular reactive oxygen species(ROS)and reactive nitrogen species(RNS),and mitochondrial membrane potential were analyzed by flow cytometry.The activity of caspases was measured colorimetrically,and the expression pattern of mitochondrial proteins was analyzed.Results:The methanol extract of A.hygrometricus and MOLT-4 cells were identified as the most potent extract exhibiting antiproliferative activity and most sensitive cell line,respectively.The mushroom extract induced robust selective apoptosis in MOLT-4 cells and arrested cell cycle progression at the G0/G1 stage.The extract disrupted the mitochondrial membrane potential and enhanced ROS production in MOLT-4 cells.The methanol extract induced apoptosis by downregulating the expression of Bcl-2,increasing the expression of Bax,and activating the caspase cascade.Conclusion:The novel wild edible mushroom is a potential repository of biomolecules for the development of antileukemic drugs.

Non-alcoholic fatty liver disease in type 2 diabetes:Emerging evidence of benefit of peroxisome proliferator-activated receptors agonists and incretin-based therapies

Nonalcoholic fatty liver disease(NAFLD)is a global epidemic,affecting more than half of the people living with type 2 diabetes(T2D).The relationship between NAFLD and T2D is bidirectional and the presence of one perpetuates the other,which significantly increases the hepatic as well as extrahepatic complications.Until recently,there was no approved pharmacological treatment for NAFLD/nonalcoholic steatohepatitits(NASH).However,there is evidence that drugs used for diabetes may have beneficial effects on NAFLD.Insulin sensitizers acting through peroxisome proliferator-activated receptor(PPAR)modulation act on multiple levels of NAFLD pathogenesis.Pioglitazone(PPARγ agonist)and saroglitazar(PPARα/γagonist)are particularly beneficial and recommended by several authoritative bodies for treating NAFLD in T2D,although data on biopsyproven NASH are lacking with the latter.Initial data on elafibanor(PPARα/δ agonist)and Lanifibranor(pan PPAR agonist)are promising.On the other hand,incretin therapies based on glucagon-like peptide-1(GLP-1)receptor agonists(GLP-1RA)and dual-and triple-hormone receptor co-agonists reported impressive weight loss and may have anti-inflammatory and antifibrotic properties.GLP-1 RAs have shown beneficial effects on NAFLD/NASH and more studies on potential direct effects on liver function by dual-and triple-agonists are required.Furthermore,the long-term safety of these therapies in NAFLD needs to be established.Collaborative efforts among healthcare providers such as primary care doctors,hepatologists,and endocrinologists are warranted for selecting patients for the best possible management of NAFLD in T2D.
