Effect of age on the standardized ileal amino acid digestibility of soybean meal and canola meal in broilers

Standardized ileal digestibility coefficients (SIDC) of nitrogen (N) and amino acids (AA) in two protein sources (soybean meal [SBM] and canola meal [CM]) were investigated at six broiler ages (d 7, 14, 21, 28, 35, and 42). Two assay diets were formulated to contain either SBM (413 g/kg) or CM (553 g/kg) as the sole dietary AA source. Titanium dioxide (5 g/kg) was added as an indigestible marker. A total of 696 male broilers at 1 d old were allotted to 12 replicate cages per age group. Each assay diet was offered to birds for 4 d prior to the ileal digesta collection on d 7 (14 birds/cage), 14 (12 birds/cage), 21 (10 birds/cage), 28 (8 birds/cage), 35 (8 birds/cage) and 42 (6 birds/cage), respectively. The apparent digestibility coefficients were standardized using age-specific basal endogenous AA flows. In the SBM group, though the SIDC of N tended to be influenced (quadratic; P=0.075) by age, no linear or quadratic response of age effect was observed on the average SIDC of indispensable (IAA) and total AA (TAA). An age effect (quadratic; P<0.05) was observed on the average SIDC of dispensable AA (DAA) in SBM with the highest value recorded at d 7, followed by a decrease from d 14 to 28, which increased beyond d 35. The SIDC of some individual AA (Arg, Thr, Trp, Cys, Pro) were affected (P<0.05 or P<0.001) in a quadratic manner by age. In the CM, the SIDC of N, average SIDC of IAA, DAA and TAA were influenced (quadratic; P<0.05 or P<0.001) by age. The SIDC of N and average SIDC of DAA and TAA were higher from d 7 to 14, declined at d 21, and then increased beyond d 28. The average SIDC of IAA was low between d 7 and 28 and increased thereafter. The SIDC of individual AA were affected (linear or quadratic; P<0.05 or P<0.001) by different magnitudes by age. The age influence on the SIDC AA was variable, depending on the protein source and AA. The results demonstrate that age-specific SIDC AA data might need consideration in broiler feed formulations.

Objective: Prostate cancers(PCa) in Asian individuals are molecularly distinct from those found in their Caucasian counterparts.There is no risk stratification tool for Asian men with rapid biochemical recurrence(BCR) following radical prostatectomy(Rad P). This study aims to assess the detection rate of ^(68)Ga-prostate-specific membrane antigen-positron emission tomography/computed tomography(PSMA-PET/CT) for diagnosis of clinical recurrence and as a treatment decision making tool in Asian patients with BCR post-Rad P.Methods: ^(68)Ga PSMA-PET and CT body with/without bone scan [conventional workup(CWU)] were performed in 55 Asian patients with BCR within 36 months post-Rad P. Two blinded reviewers assessed the images. Detection rates of ^(68)Ga PSMAPET/CT were evaluated, and impact on management was reviewed by comparison with CWU.Results: Median time to BCR post-Rad P was 8.1 months. Detection rate for ^(68)Ga PSMA-PET/CT was 80%(44/55). A positive scan was significantly associated with increasing prostate-specific antigen(PSA) level [odds ratio(OR) = 1.13(95% CI 1.05–1.30), P =0.017], but not with higher Gleason grade or shorter PSA doubling time. Compared to CWU, ^(68)Ga PSMA-PET/CT detected an additional 106 lesions in 33/44 patients with a positive scan, resulting in a change in management in 25/44(56.8%) patients: 10 to hormonal therapy(HT) and whole pelvis radiotherapy(RT) in addition to bed RT, and 15 to palliative HT alone.Conclusions: In the present report, we demonstrated the diagnostic and treatment decision utility of ^(68)Ga PSMA-PET/CT in Asian men with rapid BCR. Detection of small volume nodal and systemic recurrences at low PSA levels(< 1.0 ng/mL) highlights the role of the tool in assigning patients to treatment intensification with HT-RT or palliative HT in polymetastatic disease.

Dear editor,Prostate cancer(PCa)remains a major healthcare burden in men globally[1].Most patients present with localized disease,and treatment is recommended based on risk classification systems like the National Comprehensive Cancer Network(NCCN)[2].However,these methods are imprecise for estimating metastasis-free survival and prostate cancer-specific mortality and thus biomarkers that can predict tumor aggression are needed[3–5].Several studies have since characterized the molecular landscape of localized PCa in White[4,5]and Black/African-American men[6],but data is lacking in Asian men.The Chinese Prostate Cancer Genome and Epigenome Atlas(CPGEA)reported on the genomic and epigenomic landscape of 208 PCa of men from China[7].Comparative analyses between the CPGEA cohort and data from The Cancer Genome Atlas(TCGA)revealed higher frequencies of Forkhead box A1(FOXA1)and chromodomain-helicase DNA-binding 1(CHD1)mutations,and lower frequencies of phosphatase and tensin homolog(PTEN)mutations and transmembrane protease serine 2-E26 transformationspecific related gene(TMPRSS2-ERG)fusion in Chinese compared with White men[7].These preliminary findings highlight the presence of race-specific differences in molecular phenotypes of PCa.

Sammon Quadratic Recurrent Multilayer Deep Classifier for Legal Document Analytics

In recent years,machine learning algorithms and in particular deep learning has shown promising results when used in the field of legal domain.The legal field is strongly affected by the problem of information overload,due to the large amount of legal material stored in textual form.Legal text processing is essential in the legal domain to analyze the texts of the court events to automatically predict smart decisions.With an increasing number of digitally available documents,legal text processing is essential to analyze documents which helps to automate various legal domain tasks.Legal document classification is a valuable tool in legal services for enhancing the quality and efficiency of legal document review.In this paper,we propose Sammon Keyword Mapping-based Quadratic Discriminant Recurrent Multilayer Perceptive Deep Neural Classifier(SKM-QDRMPDNC),a system that applies deep neural methods to the problem of legal document classification.The SKM-QDRMPDNC technique consists of many layers to perform the keyword extraction and classification.First,the set of legal documents are collected from the dataset.Then the keyword extraction is performed using SammonMapping technique based on the distance measure.With the extracted features,Quadratic Discriminant analysis is applied to performthe document classification based on the likelihood ratio test.Finally,the classified legal documents are obtained at the output layer.This process is repeated until minimum error is attained.The experimental assessment is carried out using various performance metrics such as accuracy,precision,recall,F-measure,and computational time based on several legal documents collected from the dataset.The observed results validated that the proposed SKM-QDRMPDNC technique provides improved performance in terms of achieving higher accuracy,precision,recall,and F-measure with minimum computation time when compared to existing methods.

Calorie restriction potentiates epigallocatechin-3-gallate-mediated Nrf2 activation in hepatocytes of aged rats

Objective:To explore the combinatorial effect of epigallocatechin-3-gallate(EGCG)and calorie restriction on activation of nuclear factor erythroid 2-related factor 2(Nrf2),a transcription factor involved in the antioxidant defense system of aged rats.Methods:Aged male Wistar rats were calorie-restricted and treated with EGCG orally for 45 days.The initial body weight of aged rats was recorded,and the final body weight was measured at the end of the experimental period.Serum lipid and lipoprotein status,oxidative stress markers such as free radicals and malondialdehyde levels,and reduced glutathione were assessed.In addition,RT-PCR and Western blotting analyses were performed.Results:Calorie restriction potentiated the effect of EGCG on enhancing antioxidant status,improving the levels of serum lipid and lipoproteins,upregulating Nrf2 and Bcl2,and downregulating Keap1,cullin3,Bax and cytochrome c in aged rats.Conclusions:Calorie restriction can promote EGCG-mediated Nrf2 activation in aged rats.This preliminary finding paves the way for a combinatory approach to replenishing the antioxidant status during aging,thereby reducing the risk for age-associated degenerative diseases.

Effects of sonication amplitude on the microstructure and mechanical properties of AZ91E magnesium alloy

Light metals are gaining increased attention due to ecological sustainability concerns and strict emission regulations. Magnesium(Mg)is one such metal that has the potential to replace high density components, which can reduce emissions through lightweighting. However,the mechanical properties of Mg alloys must be improved for them to become viable candidates for structural applications. To this end, the current study examines the effect of sonication vibrational amplitude on the microstructure and mechanical properties of AZ91E Mg alloy.The molten alloys were subjected to ultrasonic treatment at a frequency of 20 kHz, 180 s of processing time and vibrational amplitudes ranging from 1.25 to 15 μm. The resultant castings were characterized using optical microscopy, scanning electron microscopy and tensile testing. It was found that sonication with amplitudes up to 7.5 μm was able to effectively refine the secondary phases of the alloy. Similar trends were observed for grain size and yield strength. The refinement in microstructure was likely caused by the finer grain size and cavitation induced undercooling of the liquid metal. In addition, it was also noted that even the lowest level of amplitude(1.25 μm) was able to increase the density, improve the ultimate tensile strength and ductility of the castings. The tensile strength and ductility were thought to have been enhanced by ultrasonic degassing and refinement in microstructure, while the yield strength was improved through the Hall-Petch effect. The results from this study provided a basis for optimizing the sonication process and promoting its use in industry. As a result, Mg alloys improved through ultrasonic processing have the potential to replace higher density components, with consequent energy efficiency and environmental and ecological benefits.

Progress in ileal endogenous amino acid flow research in poultry

The progress in our understanding of the endogenous protein concept over the past century is reviewed.Nondietary proteins found in the digesta at the terminal ileum of poultry,known as endogenous protein loss,are comprised of digestive secretions,mucus and sloughed gut epithelial cells.The measurement of this loss is of fundamental importance because it is an indicator of gut metabolism and is essential to adjust apparent estimates of ileal amino acid digestibility.The ileal endogenous amino acid losses comprise of two components,namely basal and specific losses.The basal losses are fixed and associated with feed dry matter intake,whereas the specific losses are variable and induced by the presence of dietary components such as fibre and anti-nutrients.Currently there is no methodology available to directly measure the specific endogenous losses and these losses are calculated by determining the basal and total(basal plus specific)losses and,then subtracting the basal losses from total losses.The seminal features,specific applications and shortcomings of available methodologies are briefly outlined as well as the practical challenges faced in using the published endogenous amino acid loss values for true digestibility corrections.The relevance of taurine as a component of endogenous protein flow in poultry is identified for the first time.

Colorectal cancer accounts for 10%of diagnosed cancers globally and often presents as advanced disease,necessitating aggressive treatment.With both younger and healthier elderly patients being diagnosed,as well as potentially the need for salvage therapy post total neoadjuvant treatment,surgical options for cure include pelvic exenteration.Whilst typically performed via an open approach,there has been an increased utilisation of minimally invasive techniques including robotic surgery.Offering smaller in-cisions,reduced postoperative pain,and quicker recovery time than open surgery,robotic techniques have demonstrated lower blood loss,shorter hospital stays,and reduced morbidity.Moreover,the er-gonomic design of robotic systems provides surgeons with comfort during long procedures and increased precision.It also offers an increased opportunity for organ preservation and reconstruction whilst maintaining adequate oncological outcomes.As robotic technology continues to evolve and combines with artificial intelligence,it is poised to play an even more significant role in the management of complex colorectal cancer cases,improving survival and long-term outcomes.

Influence of barley inclusion method and protease supplementation on growth performance,nutrient utilisation,and gastrointestinal tract development in broiler starters

The influence of the method of barley inclusion(fine,coarse and whole barley)in a wheat-based diet and protease supplementation(0 and 0.20 g/kg)on growth performance,nutrient utilisation and gastrointestinal tract development of broilers(d 1 to 21)was evaluated in a 32 factorial arrangement.Whole barley(WB)grains were ground in a hammer mill to pass through the screen sizes of 2.5 and 8.0 mm to achieve fine(FB)and coarse(CB)barley particle sizes,respectively.A total of 288,one-day-old male broilers were allotted to 36 cages(6 cages/treatment;8 birds/cage).There was no significant(P>0.05)interaction between barley inclusion method and protease for any growth performance or nutrient utilisation parameters.Birds fed diets containing CB and WB showed higher(P0.05)on growth performance and nutrient utilisation,most likely due to the well balanced digestible amino acids and high inherent digestibility of protein in the basal diet,and/or the presence of exogenous carbohydrase and phytase.In conclusion,the present results showed that the inclusion of coarsely ground and whole barley in a wheat-based diet can enhance nutrient and energy utilisation and is beneficial to the growth performance of young broilers.
