Antimicrobial Activity of Purified Toxins from Crossopriza lyoni (Spider) against Certain Bacteria and Fungi

Toxins from spider venom Crossopriza lyoni were subjected to purify on a Sepharose CL-6B 200 column. These were investigated for its antibacterial and antifungal activity against 13 infectious microbial pathogenic strains. Antimicrobial susceptibility was determined by using paper disc diffusion and serial micro-dilution assays. Triton X-100 (0.1%) proved to be a good solubilizing agent for toxin/proteins. Higher protein solubilization was observed in the supernatant than in the residue, except TCA. The elution pattern of purified and homogenized sting glands displayed two major peaks at 280 nm. First one was eluted in fraction no. 43 - 51 while second one after fraction no. 61 - 90. From gel filtration chromatography total yield of protein obtained was 67.3%. From comparison of gel chromatographs eluted toxins peptide molecular weight was ranging from 6.2 - 64 kD. Toxin peptides have shown lower MIC values i.e. 7.5 - 15 μg/ml against K. pneumoniae, E. coli, L. acidophilus, B. cereus;against S. aureus and M. luteus that the broad spectrum antibiotics i.e. tetracycline and ampicillin. In tests, larger inhibition zone diameter was obtained in comparison to control. Diameter of inhibition zones obtained in spider toxins at a concentration range of 197.12 - 0.96 g/ml for E. coli was 17.86 ± 0.21, Bacillus cereus 19.13 ± 0.21, L. acidophilus 16.83 ± 0.25, Micrococcus luteus 18.46 ± 0.17, S. aeurus 16.23 ± 0.19, Klebsiella pneumoniae 21.83 ± 0.16, Salmonella typhi 16.16 ± 0.21, Vibrio cholera 18.66 ± 0.21, Pseudomonas aeruginosa 18.66 ± 0.21, Aspergillus niger 22.9 ± 0.24, Candida albicans 24.66 ± 0.28, Rhizopus stolonifer 21.1 ± 0.16. Spider toxins generate cytotoxic effect on bacterial cells that results in heavy cell death. No doubt spider toxins can be used as alternate of broad spectrum antibiotics.

In this paper, the analysis of crosstalk induced by stimulated Raman scattering has been discussed. The expression for SRS induced crosstalk has been observed at different fiber types, such as single mode fiber, DCF, NZDF and NZDSF with the variation of modulation frequency, transmission length and the input power. It has been observed that as an increase in the modulation frequency, the crosstalk decreases.

Plant pathogenic bacteria deliver effectors into plant cells to suppress immunity and promote pathogen survival;however, these effectors can be recognised by plant disease resistance (R) proteins to activate innate immunity. The bacterial acetyltransferase effectors HopZ5 and AvrBsT trigger immunity in Arabidopsis thaliana genotypes lacking SUPPRESSOR OF AVRBST-ELICITED RESISTANCE 1 (SOBER1). Using an Arabidopsis accession, Tscha-1, that naturally lacks functional SOBER1 but is unable to recognise HopZ5, we demonstrate that RESISTANCE TO P. SYRINGAE PV MACULICOLA 1 (RPM1) and RPM1-INTERACTING PROTEIN 4 (RIN4) are indispensable for HopZ5- or AvrBsT-triggered immunity. Remarkably, T166 of RIN4, the phosphorylation of which is induced by AvrB and AvrRpm1, was directly acetylated by HopZ5 and AvrBsT. Furthermore, we demonstrate that the acetylation of RIN4 T166 is required and sufficient for HopZ5- or AvrBsT-triggered RPM1-dependent defence activation. Finally, we show that SOBER1 interferes with HopZ5- or AvrBsT-triggered immunity by deacetylating RIN4 T166. We have thus elucidated detailed molecular mechanisms underlying the activation and suppression of plant innate immunity triggered by two bacterial acetyltransferases, HopZ5 and AvrBsT from different bacterial pathogens.

Principal component transformation is a standard technique for multi-dimensional data analysis. The purpose of the present article is to elucidate the procedure for interpreting PC images. The discussion focuses on logically explaining how the negative/positive PC eigenvectors (loadings) in combination with strong reflection/absorption spectral behavior at different pixels affect the DN values in the output PC images. It is an explanatory article so that fuller potential of the PCT applications can be realized.

Konno Procedure for Managing Small Aortic Root during Aortic Valve Replacement Surgery: An Experience of 12 Cases

Background: Small aortic annulus during aortic valve replacement can lead to implanting a smaller sized valve compared to the body surface area thereby causing patient prosthesis mismatch. Various aortic root enlargement techniques have been described depending on anterior or posterior approach. Konno procedure uses anterior approach for aortic root enlargement. In this study, we reviewed results of Konno procedure done from 2011 to 2019 by a single surgeon. Methods: 12 adult patients who underwent aortic valve replacement along with Konno procedure for small aortic root by a single surgeon at a single center between 2011 and 2019 were reviewed. Echocardiographic and demographic data and post-operative data were obtained from medical records. Symptomatic profile was assessed as per New York Heart Association Classification. Intraoperative findings and post-operative period findings were noted. Follow up symptom profile was assessed for these patients. Results: 12 patients underwent Konno procedure between 2011 and 2019 for small aortic root along with valve replacement. The main indication for surgery was aortic stenosis with small aortic annulus, with or without involvement of the mitral valve. Preoperatively, 3 patients had NYHA class II and 9 patients had NYHA class III symptoms. Mean age at operation was 26.42 years, minimum age 10 years, and maximum age 39 years. 3 were females and 9 were males. Mean bypass time was 106.4 minutes and aortic cross clamp time was 80.67 minutes. Mechanical aortic valves were implanted in all patients. Mean post-operative blood loss was 134.2 ml and duration of ventilation before extubation was 14.5 hours. Mean duration of intensive care unit (ICU) stay was 2.83 days and hospital stay was 9.1 days. Mean gradient in the post-operative period was 10.75 mm Hg. There was no mortality in these 12 patients and no reoperation was needed in the follow up period. Follow up in the outpatient department suggested all patients had NYHA class I

Background: Spontaneous gallbladder perforation(GBP) is an uncommon diagnosis. This study presented the experience of managing spontaneous GBP over nine years at a large, tertiary care university hospital in north India and investigated the outcomes and treatment strategies. Methods: A retrospective review of prospectively maintained digital database of consecutive patients was performed. All patients received medical and/or surgical treatment for spontaneous GBP in our department between January 2010 and June 2018. Results: We identifed 151 patients(81 females and 70 males) with mean age of 53 years. Most common presenting features were pain(96.7%), fever(54.3%) and jaundice(31.1%). Most common cause was gallbladder stones(84.8%) followed by common bile duct stones(30.5%), xanthogranulomatous cholecystitis(17.9%) and malignancy(11.9%). As per Niemeier classifcation, 8.6% had type 1 GBP(free perforation in peritoneal cavity), 76.2% had type 2 GBP(localized perforation) and 13.2% had type 3 GBP(cholecystoenteric fstula). About 60% of the perforations were diagnosed preoperatively. Type 1 was more common in patients with diabetes and also had the worst prognosis. Surgery was performed in 109 patients(72.2%). Seven patients(4.6%) had a postoperative morbidity of Clavien-Dindo Ⅲ or higher. There were three mortalities in patients who underwent surgery. Conclusions: High index of suspicion is required for preoperative diagnosis of GBP, especially in types 2 and 3. Laparoscopic cholecystectomy can be difficult in these patients and patients may require open or partial cholecystectomy. Early diagnosis and step-up approach for the treatment of GBP is critical.

The rise of automation with Machine-Type Communication(MTC)holds great potential in developing Industrial Internet of Things(IIoT)-based applications such as smart cities,Intelligent Transportation Systems(ITS),supply chains,and smart industries without any human ***,MTC has to cope with significant security challenges due to heterogeneous data,public network connectivity,and inadequate security *** overcome the aforementioned issues,we have proposed a blockchain and garlic-routing-based secure data exchange framework,i.e.,GRADE,which alleviates the security constraints and maintains the stable connection in ***,the Long-Short-Term Memory(LSTM)-based Nadam optimizer efficiently predicts the class label,i.e.,malicious and non-malicious,and forwards the non-malicious data requests of MTC to the Garlic Routing(GR)*** GR network assigns a unique ElGamal encrypted session tag to each machine partaking in ***,an Advanced Encryption Standard(AES)is applied to encrypt the MTC data ***,the InterPlanetary File System(IPFS)-based blockchain is employed to store the machine's session tags,which increases the scalability of the proposed GRADE ***,the proposed framework has utilized the indispensable benefits of the 6G network to enhance the network performance of ***,the proposed GRADE framework is evaluated against different performance metrics such as scalability,packet loss,accuracy,and compromised rate of the MTC data *** results show that the GRADE framework outperforms the baseline methods in terms of accuracy,i.e.,98.9%,compromised rate,i.e.,18.5%,scalability,i.e.,47.2%,and packet loss ratio,i.e.,24.3%.
