Comparative metabolomics reveal histamine biodegradation mechanism by salt stressed Bacillus subtilis JZXJ-7

Bacillus subtilis (B. subtilis) JZXJ-7 isolated from shrimp paste can significantly degrade histamine under salt stress but the mechanism is unclear. This study aims to evaluate the effect of 170 and 340 mmol/L NaCl on B. subtilis JZXJ-7 growth， histamine degradation， antioxidant enzymes [catalase (CAT)， superoxide dismutase (SOD) and glutathione S-transferase (GST)] activities and Na+， K+-ATPase activity. Furthermore， comparative metabolomics was used to investigate histamine biodegradation mechanism by *** JZXJ-7 subjected to salt stress. Both 170 and 340 mmol/L NaCl promoted B. subtilis JZXJ-7 growth in late stages of reproduction (32 - 48 h)， increased histamine degradation rate by 64.85% and 79.87% (P < 0.05)， Na+， K+-ATPase activity to 6.28 U/mg (P < 0.05) and 11.63 U/mg (P < 0.01) respectively. NaCl treatment significantly increased the activities of CAT， GST and SOD (P < 0.05)， amino acids and its metabolites (33.39%)， benzene and substituted derivatives (12.05%)， heterocyclic compounds (10.62%)， organic acids and derivatives (9.75%)， aldehydes， ketones， esters (5.59%) and nucleotides and its metabolites (4.58%). Metabolite set enrichment analysis revealed NaCl induced differential metabolic pathways of D-glutamine， D-glutamate， L-arginine， L-proline， histidine and glycerophospholipids， L-lysine degradation， and aminoacyl-tRNA biosynthesis. Exposure to 340 mmol/L NaCl up-regulated carbohydrate， glutathione and glycerophospholipid metabolism. The new insights into the mechanism of salt stress to promote B. subtilis JZJX-7 growth， energy metabolic pathways and to degrade histamine provide the theoretical basis for application of B. subtilis JZXJ-7 in food fermentation industry.

【目的】探究湛江市麻章区红树林沉积物中微生物对聚乙烯(PE)塑料的降解特性,丰富红树林塑料降解菌种类。【方法】在湛江市麻章区红树林的高、中、低潮带随机选取9个样点,并采用五点采样法从红树林表层2~5 cm的沉积物中取样。将采集到的沉积物样本在以聚乙烯为唯一碳源的无机盐培养基中进行富集、分离、纯化,以筛选出能够降解PE的菌株,并通过显微形态分析、分子鉴定等方法确定其种属;通过测定生长曲线、失重率、以及分析官能团变化来验证菌株对PE塑料的降解性能。【结果与结论】从沉积物中共筛选出K1-1、K1-2和K1-3三株PE降解能力较强的菌,分别为威尼斯不动杆菌(Acinetobacter venetianus)、Pseudomonas juntendi和维德曼芽孢杆菌(Bacillus wiedmannii)。在120 d的培养周期中,这些菌株的生长曲线均呈抛物线形态。经三株菌作用后,PE表面生成羟基,并形成氨基和羟基化合物,羟基取代羰基,导致PE表观和内部结构明显改变。经120 d处理后,PE膜的失重率分别为6.42%、5.38%和5.73%,其中K1-1菌株处理后PE塑料的失重率相较于目前报道的红树林沉积物中最高降解效率菌株Bacillus gottheilii处理后塑料的失重率提升3.55%。威尼斯不动杆菌(***)在PE塑料降解上具有一定潜力,这丰富了湛江红树林沉积物中PE降解菌的种类,为PE塑料的生物降解提供理论支持。

Seasonal Change of Loline Alkaloids in Endophyte-lnfected Meadow Fescue

Lolines are a group of saturated pyrrolizidine alkaloids that possess broad bioactivity against a wide array of herbivorous insects. However, they do not exhibit toxicity to ruminants such as cattle and sheep. In order to study the direct and potential physiological effects on ruminants and the mechanism of insecticide/insectifuge, the distribution of loline alkaloids in endophyte-infected meadow fescue and the seasonal change of the distribution were analyzed. The crowns, roots and leaves of endophyte-infected meadow fescue at its four different growth periods, i.e., spring, summer, early autumn and late autumn, in New Zealand were colleted. After powdering, organic solvent extraction and purification by column chromatography, all loline alkaloid samples were analyzed by capillary gas chromatography with 4-phenylmorpholine （PM） as an internal standard. The analytic results showed that the loline contents in the roots, crowns and leaves of endophyte-infected meadow fescue vary with seasons. Even within the same season, the distribution of lolines in endophyte-infected meadow fescue varies. During summer, lolines mainly existed in the leaves and roots, but in early autumn, they are produced in the crowns. It was concluded that, lolines were mainly produced in the leaves and roots of endophyte-infected meadow fescue. In gas chromatographic analysis, N-formylloline, the major component of loline alkaloid in the plant, was employed to assay the alkaloids.

Study of Bacillus subtilis on growth performance, nutrition metabolism and intestinal microflora of 1 to 42 d broiler chickens

To study the influence of different levels of Bacillus subtilis on growth performance, nutrition metabolism and intestinal microflora of 1 to 42 d Arbor Acres(AA) broilers, a total of 800 one-day-old healthy broilers were randomly divided into 5 groups with 4 replicates per group and 40 broilers per replicate. Broilers were fed a basic diet(group 1) which acted as the control group, and 4 other groups(2 to 5) were fed the basal diet with B. subtilis added at concentrations of 100, 150, 200 and 250 mg/kg, respectively for 42 days. The results showed as follow: the average daily gain(ADG) of group 4 was significantly higher than(P 0.05). The feed to gain ratio(F/G) of all the experimental groups was lower than that of the control and the difference was significant in group 4(P 0.05), crude fat(P > 0.05), dry matter(P > 0.05) and organic matter(P < 0.05). B. subtilis decreased the Escherichia coli and Salmonella populations in the cecum. This shows that adding B. subtilis to the broiler diet can improve the growth performance, increase feed efficiency, regulate serum index and reduce harmful bacteria in the intestinal tract. Based on our study, it could be recommended that addition of B. subtilis at 200 mg/kg could improve the growth performance of broilers.

为探究湛江近海域海洋沉积物中类二噁英多氯联苯（dioxin-like polychlorinated biphenyls,Dl PCBs）的生物学毒性效应,选取湛江近海域2个地点（近工业区TS和近生活区JSW）采集沉积物样品,制备近海域沉积物Dl PCBs提取物,将斑马鱼胚胎暴露于不同浓度的Dl PCBs提取物,测定斑马鱼胚胎7-乙氧基异吩恶唑脱乙基酶（ethoxyresorufin-O-deethylase,EROD）活性和cyp1a m RNA相对表达量。结果发现,JSW采样点Dl PCBs提取物染毒组EROD酶活性变化与TS采样点Dl PCBs提取物一致,在各染毒浓度下,斑马鱼仔鱼EROD酶活性为对照组的1.1～1.8倍。TS和JSW采样点不同浓度Dl PCBs提取物暴露斑马鱼胚胎96 h后,使斑马鱼仔鱼cyp1a m RNA相对表达量是对照组的3.36～19.45倍。说明一定浓度的近海沉积物Dl PCBs能诱导斑马鱼仔鱼EROD酶活性和cyp1a m RNA表达量升高,且呈现浓度-效应关系。
