Reducing phosphorylation of nitrate reductase improves nitrate assimilation in rice

Nitrate reductase(NR) is an important enzyme for nitrate assimilation in plants, and post-translational phosphorylation regulates NR activity. To evaluate the impact of the dephosphorylation of nitrate reductase 1(NIA1) protein on NR activity,nitrogen metabolism and plant growth, NIA1 phosphorylation site directed mutant lines(S532 D and S532 A) and an OsNia1 over-expression line(OE) were constructed, and the phenotype, NIA1 protein and its phosphorylation level, NR activity,nitrate metabolism and reactive oxygen metabolism of the transgenic lines were analysed. Exogenous NIA1 protein was not phosphorylated in S532 D and S532 A mutant lines, and their NR activities, activity states of NR and assimilation efficiencies of NO3–-N were higher than those in Kitaake(WT) and OE. The changes in these physiological and biochemical indexes in the OE line were less than in S532 D and S532 A compared to WT. These results suggest that the removal of transcriptional level control had little effect on nitrogen metabolism, but the removal of post-translational modification had a profound effect on it. With the removal of NIA1 phosphorylation and the improvement in the nitrate assimilation efficiency, the plant height and chlorophyll content of S532 D and S532 A decreased and the hydrogen peroxide and malondialdehyde contents of rice seedlings increased, which may be related to the excessive accumulation of nitrite as an intermediate metabolite. These results indicated that the phosphorylation of NR may be a self-protection mechanism of rice. The reduced phosphorylation level of nitrate reductase improved the assimilation of nitrate, and the increased phosphorylation level reduced the accumulation of nitrite and prevented the toxic effects of reactive oxygen species in rice.

Emotion Recognition in Conversations(ERC)is fundamental in creating emotionally ***-BasedNetwork(GBN)models have gained popularity in detecting conversational contexts for ERC ***,their limited ability to collect and acquire contextual information hinders their *** propose a Text Augmentation-based computational model for recognizing emotions using transformers(TA-MERT)to address *** proposed model uses the Multimodal Emotion Lines Dataset(MELD),which ensures a balanced representation for recognizing human *** used text augmentation techniques to producemore training data,improving the proposed model’s *** encoders train the deep neural network(DNN)model,especially Bidirectional Encoder(BE)representations that capture both forward and backward contextual *** integration improves the accuracy and robustness of the proposed ***,we present a method for balancing the training dataset by creating enhanced samples from the original *** balancing the dataset across all emotion categories,we can lessen the adverse effects of data imbalance on the accuracy of the proposed *** results on the MELD dataset show that TA-MERT outperforms earlier methods,achieving a weighted F1 score of 62.60%and an accuracy of 64.36%.Overall,the proposed TA-MERT model solves the GBN models’weaknesses in obtaining contextual data for ***-MERT model recognizes human emotions more accurately by employing text augmentation and transformer-based *** balanced dataset and the additional training samples also enhance its *** findings highlight the significance of transformer-based approaches for special emotion recognition in conversations.

Porous pre-sintered zirconia is subject to white machining during which its elasticity, plasticity and resistance to machining-induced damage determine its machinability and final quality. This study used nanoindentation techniques and the Sakai＇s series elastic and plastic deformation model to extract the resistance to plastic deformation from the plane strain modulus and the contact hardness for presintered zirconia. The modulus and the resistance to plasticity were used to calculate the relative amount of elasticity and plasticity. The fracture energy and the normalized indentation absorbed energy were used to deconvolute the resistance to machining-induced cracking based on the Sakai-Nowak model. All properties were extracted at a 10 mN peak load and loading rates of 0.1-2 mN/s to determine the loading rate effects on these properties. We found that the resistance to plasticity and the resistance to machining-induced cracking were independent of the loading rate （ANOVA, p 〉 0.05）. The elastic and plastic displacements depended on the loading rate through power laws. This loading rate-dependent deformation behaviour was explained by the maximum shear stress generated underneath the indenter and the indentation energy. The plastic deformation components and the indentation absorbed energy at all loading rates were higher than the elastic deformation components and the elastic strain energy, respectively. Finally, we established the linkage among the pore structure, indentation behaviour and machinability of pre-sintered zirconia.

Machines are growth engines of the economy;each sector of the economy achieves its demand by the use of the machine. They are installed in various establishments for the purpose of using them to perform certain functions or others. However, as a result of the kind of forces, dynamic and static loads, they transmitted to their adjoining surroundings when used, they are often mounted on supporting structures, foundations or a combination of them to achieve adequate or appropriate safe operation and stability. When a machine is operating, it is subjected to several time-varying forces and as a result of which it tends to exhibit vibrations. In such a situation or process, a certain quantity of this force is transmitted to the foundation, which could undermine the life of the foundation and also affect its performance and the operation of any other machines on the same foundation. Hence, it makes sense to minimize this force transmission. This research aimed at developing an adjustable steel-framed structure for supporting the major components of a 5.0 kW micro-steam power unit (steam, turbine and alternator) and evaluating the performance of the unit with or without the vibration isolator when they are axially connected with flexible flange coupling or transversely connected with sets of belts and pulleys, in succession, respectively. The results showed that reduction in the force transmitted to the supporting structure occurred when the vibration produced by the unit is isolated from its base by the use of a vibration isolator, maximum reduction of 99.95% achieved when axially coupled and 99.91% when transversely connected with belt and pulley system. The results also showed that better performance would be attained when the steam turbine is axially coupled to the alternator than when connected with belt and pulley;The maximum voltage of 52 V and speed of 1000 rpm at 77 dB sound level attained with coupling connection, and voltage of 20 V and speed of 752 rpm at 75 dB

Objectives: The study’s aims to determine and assess the application of problem-based learning to undergraduate nursing students. Background: Nursing students are the upcoming health care delivery system;according to their standard of learning, it will affect their clinical training. Method: The study design is a case study review, the data was collected using many articles related to problem-based learning collected from E-books and E-journals websites like CINAHEL, Google Scholar, etc. After that, the data was analyzed and evaluated related to the application of problem-based learning on undergraduate nursing students. Result: The result appeared that most of the research proved and supported that the application of problem-based learning is effective for undergraduate nursing students, and students can solve patients’ problems in a better way. Conclusion: In conclusion, problem-based learning is an essential part of the nursing diagnosis process that will increase knowledge, and performance, and merge it with the nursing concepts.

An adjustable bed is a supporting structure designed to provide a seating base or platform, for a 5.0 KW micro steam turbine generator plant prior to its mounting on a block-type concrete foundation. The design of the bed frame and flanges was carried out by considering the predetermined weights of the turbine and generator (alternator) [1]. On this basis, steel materials of U-channels and angle irons were used in the fabrication of the generator bed. The bed was designed to be adjustable by accommodating direct coupling of the turbine with the generator, and the belt drive. Fabrication was carried out by welding, machining, and assembly. During assembly, the bed was made to accommodate damping materials in order to reduce the vibration of the plant [2]. The performance of the unit with or without vibration isolator when they are axially connected with flexible flange coupling or transversely connected with sets of belt and pulley, in succession respectively. The results showed that a reduction in the force transmitted to the supporting structure occurred when the vibration produced by the unit is isolated from its base by the use of a vibration isolator, maximum reduction of 99.95% achieved when axially coupled and 99.91% when transversely connected with belt and pulley system [3].
