Ascorbic acid and prunasin,two candidate biomarkers for endodormancy release in almond flower buds identified by a nontargeted metabolomic study

Temperate fruit trees belonging to prunus species have the ability to suspend(induce dormancy)and resume growth periodically in response to environmental and seasonal *** release requires the long-term accumulation of *** accumulation of cultivar-specific chill requirements,plants enter the state of ecodormancy,which means the ability to grow has been restored,depending on the fulfilment of heat *** many different metabolic pathways are implicated in endodormancy release,we have performed a metabolomic analysis,using the ultra-high-performance liquid chromatography–quadrupole time-of-flying(UpLC–QToF)*** assayed flower buds in different stages of endodormancy in four almond cultivars with different flowering times:the extra-early Desmayo Largueta,the late Antoñeta,the extra-late penta,and the ultra-late *** orthogonal projection to latent-structure discriminant-analysis model was created to observe differences between endodormant and ecodormant flower *** metabolites showing the most significant variation were searched against the Metlin,HMDB,and KEGG libraries,which allowed us to identify 87 *** metabolites were subsequently assigned to specific pathways,such as abscisic acid biosynthesis,phenylpropanoid biosynthesis,and D-sorbitol metabolism,among *** two metabolites that exhibited the most significant variations in all the cultivars studied with fold changes of up to 6.49 were ascorbic acid and *** the first time,these two metabolites have been proposed as potential biomarkers for endodormancy release in *** the high synteny present between the Rosaceae species,these results could be extrapolated to other important crops like peach,plum,cherry,or apricot,among others.

Mitochondrial protection by low doses of insulin-like growth factor-Ⅰin experimental cirrhosis

AIM： To characterize the mitochondrial dysfunction in experimental cirrhosis and to study whether insulin-like growth factor-Ⅰ （IGF-Ⅰ ） therapy （4 wk） is able to induce beneficial effects on damaged mitochondria leading to cellular protection. METHODS： Wistar rats were divided into three groups： Control group, untreated cirrhotic rats and cirrhotic rats treated with IGF-Ⅰ treatment （2 μg/1O0 g bw/d）. Mitochondrial function was analyzed by flow cytometry in isolated hepatic mitochondria, caspase 3 activation was assessed by Western blot and apoptosis by TUNEL in the three expedmental groups. RESULTS： Untreated cirrhotic rats showed a mitochondrial dysfunction characterized by a significant reduction of mitochondrial membrane potential （in status 4 and 3）; an increase of intramitochondrial reactive oxigen species （ROS） generation and a significant reduction of ATpase activity. IGF-Ⅰ therapy normalized mitochondrial function by increasing the membrane potential and ATpase activity and reducing the intramitochondrial free radical production. Activity of the electron transport complexes Ⅰ and Ⅲ was increased in both cirrhotic groups. In addition, untreated cirrhotic rats showed an increase of caspase 3 activation and apoptosis. IGF- Ⅰ therapy reduced the expression of the active peptide of caspase 3 and resulted in reduced apoptosis. CONCLUSION： These results show that IGF- Ⅰ exerts a mitochondrial protection in experimental cirrhosis leading to reduced apoptosis and increased ATp production.

Natural killer(NK)cell-mediated antibody-dependent cellular cytotoxicity(ADCC)through CD16 plays a critical role in antihuman immunodeficiency virus(HIV)responses.1–3 CD300a is a surface receptor highly expressed on NK cells that has the capacity to inhibit NK cell-mediated cytotoxicity in healthy donors.4 The CD300a molecule has been related to several viral infections and is able to diminish the NK cell killing of pseudorabies-infected cells through interactions with its ligands phosphatidylserine and phosphatidylethanolamine.5 In addition,CD300a expression on B and CD4+T lymphocytes is altered during HIV-1 infection,and combined antiretroviral therapy(cART)does not restore nonpathological expression levels.5,6 However,the expression and function of CD300a on NK cells during HIV-1 infection is still *** have determined the surface expression of CD300a on different NK cell subsets and the capacity of this receptor to inhibit CD16-induced NK cell effector functions in healthy and HIV-1 infected individuals.

With the size of the biopharmaceutical market exponentially increasing,there is an aligned growth in the importance of data-rich analyses,not only to assess drug product safety but also to assist drug development driven by the deeper understanding of structure/function *** monoclonal antibodies,many functions are regulated by N-glycans present in the constant region of the heavy chains and their mechanisms of action are not completely *** importance of their function focuses analytical research efforts on the development of robust,accurate and fast methods to support drug development and quality *** N-glycan analysis is considered as the gold standard for glycosylation characterisation;however,it is not the only method for quantitative analysis of glycoform *** this study,ten different analytical workflows for N-glycan analysis were compared using four monoclonal *** observing good comparability between the quantitative results generated,it was possible to appreciate the advantages and disadvantages of each technique and to summarise all the observations to guide the choice of the most appropriate analytical workflow according to application and the desired depth of data generated.

Non-participation of asymptomatic candidates in screening protocols reduces early diagnosis and worsens prognosis of colorectal cancer

The Agatsuma et al’s study shows that despite the evidence of the benefits of an early colorectal cancer(CRC)diagnosis,through screening in asymptomatic subjects,up to 50%of candidates reject this option and many of those affected are diagnosed later,in advanced *** efficacy of screening programs has been well-established for several years,which reduces the risk of CRC morbidity and mortality,without taking into account the test used for screening,or other ***,a significant proportion of patients remain unscreened,so understanding the factors involved,as well as the barriers of the population to adherence is the first step to possibly modify the participation *** barriers could include a full range of social and political aspects,especially the type of financial provision of each health *** Japan,health services are universal,and this advantageous situation makes it easier for citizens to access to these services,contributing to the detection of various diseases,including ***,the symptomatic CRC group had a lower early-stage diagnosis rate than the patients detected during follow-up for other comorbidities,and symptomatic and cancer screening groups showed similar early-stage diagnosis.

Calibration of the DEM sliding friction and rolling friction parameters of a cohesionless bulk material

The discrete element method(DEM)has become a valuable tool for understanding the mechanical behaviour of granular assemblies,however,the accuracy of the DEM simulations depends on several interaction parameters such as the sliding friction and rolling friction ***,these parameters are estimated using bulk calibration approach(BCA)where the draw down test has been suggested as an effective way to approach these *** test provides up to four bulk criteria,the angle of repose,shear angle,mass flow rate and the mass loss which are used to narrow down the possible ***,there is still more research needed around it to fully understand how this methodology *** experimental and numerical study was carried out using the draw down test to assess the influence of different mass flow rates and particle shapes on DEM parameters for a cohesionless bulk *** is concluded that use of multi-sphere particles and three aperture sizes in the draw down test to calibrate the sliding and rolling friction parameters of a cohesionless bulk material can converge to a small feasible region in which a single combination of the friction coefficients can be *** calibrated sliding and rolling friction coefficients were validated using multi-sphere particles,where the maximum deviation was 5.9%from the experimental values.

Colombia aims to boost the utilization of mass transportation systems in its major cities while simultaneously reducing greenhouse gas emissions by 20%,in alignment with the commitments of the COp21 *** 2020,the transport sector in Colombia accounted for 34.4%of the country’s energy demand and was responsible for~49%of its total CO_(2) *** article presents an assessment of energy consumption,environmental effects and the fuel costs of Bogotá’s bus rapid transit system based on the Activity,Share,Intensity,Fuel methodology.A long-term analysis spanning from 2021 to 2040 was developed using the long-range energy alternatives planning *** conduct this assessment,the tool was calibrated using data from 2019 and *** distinct scenarios based on energy policies implemented in Bogotáwere examined:Business as Usual,Fast Transition,High Growth and Low *** energy consumption and environmental effects,the results underscore the pivotal role of diversifying energy sources and reducing reliance on fossil fuels such as ***,the analysis emphasizes the urgent need to accelerate the transition to alternative energy sources such as natural gas and electricity.
