The liver is a complex organ that performs several functions to maintain *** functions are modulated by calcium,a second messenger that regulates several intracellular *** hepatocytes and cholangiocytes,which are the epithelial cell types in the liver,inositol 1,4,5-trisphosphate(InsP3)receptors(ITPR)are the only intracellular calcium release *** isoforms of the ITPR have been described,named type 1,type 2 and type *** ITPR isoforms are differentially expressed in liver cells where they regulate distinct physiological *** in the expression level of these receptors correlate with several liver diseases and hepatic *** this review,we highlight how the expression level,modulation,and localization of ITPR isoforms in hepatocytes and cholangiocytes play a role in hepatic homeostasis and liver pathology.

Loss of DNA mismatch repair(MMR) function, due to somatic or germline epi/genetic alterations of MMR genes leads to the accumulation of numerous mutations across the genome, creating a molecular phenotype known as microsatellite instability(MSI). In gastric cancer(g C), MSI occurs in about 15% to 30% of the cases. This review summarizes the current knowledge on the molecular mechanisms underlying the acquisition of MSI in g C as well as on the clinic, pathologic and molecular consequences of the MSI phenotype. Additionally, current therapeutic strategies for g C and their applicability in the MSI subset are also discussed.

Arboviral diseases are viral infections transmitted to humans through the bites of arthropods,such as mosquitoes,often causing a variety of pathologies associated with high levels of morbidity and *** the past decades,these infections have proven to be a significant challenge to health systems worldwide,particularly following the considerable geographic expansion of the dengue virus(deNV)and its most recent outbreak in Latin America as well as the difficult-tocontrol outbreaks of yellow fever virus(YFV),chikungunya virus(CHIKV),and Zika virus(ZIKV),leaving behind a substantial portion of the population with complications related to these ***,the world is experiencing a period of intense globalization,which,combined with global warming,directly contributes to wider dissemination of arbovirus vectors across the ***,all continents remain on high alert for potential new ***,this review aims to provide a comprehensive understanding of the pathogenesis of the four main arboviruses today(deNV,ZIKV,YFV,and CHIKV)discussing their viral characteristics,immune responses,and mechanisms of viral evasion,as well as important clinical aspects for patient *** includes associated symptoms,laboratory tests,treatments,existing or developing vaccines and the main associated complications,thus integrating a broad historical,scientific and clinical approach.

Objective: To evaluate gene polymorphisms and their association with susceptibility to ***: A retrospective case-control study was performed with 262 subjects,comprising 78 dengue fever(DF) patients, 49 dengue hemorrhagic fever(DHF) patients and 135 healthy controls. Genotypic and allelic profiles were identified using polymerase chain reaction based in real time and amplification-refractory mutation ***: We observed a protective association of IL-10(-819 C/T) C allele(P = 0.028,OR = 0.56, CI = 0.34–0.91) against DHF, while the C/T(P = 0.047, OR = 2.10,CI = 1.01–4.38) and T/T(P = 0.008, OR = 3.82, CI = 1.38–10.59) genotypes were associated with DHF and DF, respectively. The dominant model TNFA-308 GA + AA(P = 0.043, OR = 0.45, CI = 0.20–1.00) genotypes were found to have protective effect against dengue infection. A protective association among the IFNG(+874 A/T) A/T genotype against DF(P = 0.02, OR = 0.46, CI = 0.24–0.89) and DHF(P = 0.034,OR = 0.43, CI = 0.19–0.95) was observed. When the studied single-nucleotide polymorphism was analyzed in combination, the combination GTA(P = 0.022, OR = 2.95,CI = 1.18–7.41) was statistically significantly associated with susceptibility to DF and the combination GCT(P = 0.035, OR = 0.28, CI = 0.08–0.90) with protection against the development of ***: This research identifies the association of the IFNG(+874 A/T), TNFA(-308 G/A), IL-10(-819 C/T) genotypes as a factor for protection, susceptibility and severity to dengue.

Mimosa tenuiflora and Piptadenia stipulacea are commonly accepted as drought-tolerant species but little is known about their response to drought followed by ***,the interplay between leaf water potential and osmotic adjustment on photo synthetic and growth parameters of these species was examined.A greenhouse study was conducted in a split-plot design with two water conditions in the main plots(control;drought followed by rehydration),and eight sampling times in the subplots(1,4and 7 days of drought,and 1,3,6,12,and 17 days of rehydration).Plant water status and biochemical changes were assessed as well as leaf gas exchange and subsequent *** drought stress,both species maintained a low leaf water potential throughout the day by accumulating compatible solutes,thus allowing a rapid and full recovery of water status when *** these plants minimized water loss by closing their stomata,neither showed stomatal limitations to *** inhibition of this process during drought was possibly related to mesophyll limitations as well as to a reversible downregulation of photo systems,along with adjustments of their *** deficits also triggered morphological adaptations at the whole plant level,leading to reduced growth,mainly of the shoots in *** and the roots in ***.

Objective: To evaluate the synergic effects of a novel oral supplement formulation, containing prebiotics,yeast b-glucans, minerals and silymarin(Silybum marianum), on lipid and glycidic metabolism, inflammatory and mitochondrial proteins of the liver, in control and high-fat diet-induced obese ***: After an acclimation period, 32 male C57 BL/6 mice were divided into the following groups:nonfat diet(NFD) vehicle, NFD supplemented, high-fat diet(HFD) vehicle and HFD supplemented. The vehicle and experimental formulation were administered orally by gavage once a day during the last four weeks of the diet(28 consecutive days). We then evaluated energy homeostasis, inflammation, and mitochondrial protein expression in these groups of ***: After four weeks of supplementation, study groups experienced reduced glycemia, dyslipidemia,fat, and hepatic fibrosis levels. Additionally, proliferator-activated receptor-α, AMP-activated protein kinase-1α, peroxisome proliferator-activated receptor γ co-activator-1α, and mitochondrial transcription factor A expression levels were augmented;however, levels of inhibitor of nuclear factor-κB kinase subunit a and p65 nuclear factor-κB expression, and oxidative markers were reduced. Notably, the cortisol/C-reactive protein ratio, a well-characterized marker of the hypothalamic–pituitary–adrenal axis immune interface status, was found to be modulated by the ***: We discovered that the novel supplement was able to modify different antioxidant, metabolic and inflammatory pathways, improving the energy homeostasis and inflammatory status, and consequently alleviated hepatic steatosis.

Objective: To evaluate the antibacterial activity and neuroprotective capacity of the ethanolic and aqueous extracts of Tarenaya spinosa(T. spinosa) as well as to determine and quantify some of its polyphenols by high performance liquid chromatography with diode-array detection(HPLC-DAD). Methods: The bacterial Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa strains, grown in Heart Agar Infusion, were tested. The drugs gentamicin, norfloxacin and imipenem were used to evaluate the modulating or antagonistic capacity of the T. spinosa extracts. The extract was analysed by HPLC-DAD to determine the main phenolic compounds. For the cell viability tests, individual heads of the Nauphoeta cinerea arthropod model were removed, homogenized in Trifluoromethyl ketone and centrifuged afterwards. Subsequently, 20 μL of NaN O2 were added to the biological material, except in the control group, to evaluate the protection capacity of the extracts. The homogenate of the insect heads was incubated for 2 h in tubes containing tetrazolium bromide. Results: HPLC-DAD demonstrated that the ethanolic extract of T. spinosa presented caffeic acid as the major compound. The ethanolic extract also showed neuroprotective effects at concentrations ≥ 10 μg/mL, while aqueous extract was shown to have a protective effect only at the concentration of 100 μg/mL. The aqueous extract demonstrated a clinically relevant antibacterial activity against the Staphylococcus aureus multidrug resistant strain-MDR, with MIC 512 μg/m L. However, when the extracts were associated with gentamicin and imipenem, a synergism was detected against Staphylococcus aureus and Escherichia coli MDR strains. Conclusions: Although it does not present an antibacterial action, the extracts of T. spinosa can be used in the pharmaceutical industries since its extracts show modulating action of drugs. Besides, these natural products have neuroprotective capacity.

Current antiepileptic drugs can inhibit seizure occurrence but are not effective in preventing its ***,they produce several side effects,which may impact the efficacy of the *** addition,it has been stimulating the prospection of new molecules isolated from aromatic plants,with potential anticonvulsant and neuroprotective activities and less side *** study aimed to evaluate,though behavioural and neuro-chemical methodologies,the anticonvulsant,and neuroprotective effects of eucalyptol on mice subjected to the pilocarpine-induced seizure ***(100,200 and 400 mg/kg,p.o.)was administered in mice prior to pilocarpine(350 mg/kg,i.p.)and the following behavioural parameters were assessed:Latency to First Seizure(LFS),Seizure Intensity(SI)and Latency to death(LD).In addition,an oxotremorine-induced tremors test was performed to evaluate the cholinergic system involvement on the eucalyptol *** tests were also performed,including determination of hippocampal(HC)concentration of thiobarbituric acid reactive substances(TBARS)and nitrite/nitrate and striatal(ST)concentration of noradrenaline,dopamine,and ***,histopathological,and morphometric hippocampal analysis were *** increased the latency to first seizure and latency to death,inhibited oxotremorine-induced tremors,decreased hippocampal TBARS and nitrite/nitrate overproduction,increased striatal noradrenaline and dopamine levels and prevented hippocampal *** results demonstrate the potential anticonvulsant,neuroprotective and antioxidant effects of eucalyptol,probably through a conjunction of mechanisms including muscarinic cholin-ergic antagonism,oxidative stress mitigation and the monoaminergic system modulation,which appears to effectively control the seizure onset.

Stochastic optimization of mine production scheduling with uncertain ore/metal/waste supply

Optimization of long-term mine production scheduling in open pit mines deals with the management of cash flows, typically in the order of hundreds of millions of dollars. Conventional mine scheduling utilizes optimization methods that are not capable of accounting for inherent technical uncertainties such as uncertainty in the expected ore/metal supply from the underground, acknowledged to be the most critical factor. To integrate ore/metal uncertainty into the optimization of mine production scheduling a stochastic integer programming(SIP) formulation is tested at a copper deposit. The stochastic solution maximizes the economic value of a project and minimizes deviations from production targets in the presence of ore/metal uncertainty. Unlike the conventional approach, the SIP model accounts and manages risk in ore supply, leading to a mine production schedule with a 29% higher net present value than the schedule obtained from the conventional, industry-standard optimization approach, thus contributing to improving the management and sustainable utilization of mineral resources.
