Androgen receptor(AR)is a major transcription factor that plays a role in inflammatory response including interleukin-6(IL6)signaling.1 While AR regulation through paracrine loop signaling in prostate tissue is well-studied,its impact through an IL6 autocrine loop in the lung has not been wellstudied despite the organ's response to respiratory viral *** inhibition and RNA knockdown of AR identified a bZIP transcription factor MAF to be a common target of inflammation using these perturbations in lung cells.

Aim: Emerging evidence implicates astrocyte/microglia dysregulation in a range of brain disorders, thereby making glial cells potential therapeutic targets. The novel anti-inflammatory actions of beta-funaltrexamine (β-FNA) are of particular interest. β-FNA is a derivative of naltrexone, and recognized as a selective, irreversible antagonist at the mu-opioid receptor (MOR). However, we discovered that β-FNA has novel anti-inflammatory actions that seem to be mediated through a MOR-independent mechanism. Thus far, we have focused on the acute effects of β-FNA on inflammatory ***: The effect of β-FNA treatment on interleukin-1β (IL-1β)-induced inflammatory signaling in normal human astrocytes (NHA) and C20 human microglial cells. Cytokine/chemokine expression was measured using ELISA, and nuclear factor-kappaB (NF-κB) p65 activation was evaluated by immunoblot. Results: IL-1β-induced interferon-γ inducible protein-10 (CXCL10) production in NHA was more sensitive to chronic (3 day) β-FNA as indicated by an approximately 3-fold lower EC50 compared to that observed in acutely treated cells. Chronic β-FNA did not affect IL-1β-induced monocyte chemoattractant protein-1 (CCL2) or IL-6 production in NHA. β-FNA inhibited phosphorylation of NF-κB p65, suggesting that the inhibitory effects may be due in part to reduced NF-κB activation. We showed for the first time that C20 human microglial cells were insensitive to the anti-inflammatory actions of acute β-FNA. Conclusion: β-FNA differentially affects inflammatory cytokine/chemokine expression in human astrocytes and microglia. These findings warrant further investigation into the novel anti-inflammatory actions of β-FNA, with a particular focus on astrocytes. These insights should contribute to the development of strategies to treat brain disorders that involve neuroinflammation.

利用人工气候室进行了双低冬油菜Canola（BrassicanapusL.）苗期的控流试验，以探讨供硫不足条件下对植株不同生长参数动态变化的影响效应。结果表明，单位重量叶面积（即A／W）的变化是反映植株硫营养状况的最敏感指示参数。根据其相对增产率的变化求得了对油菜叶片扩展的功能性需硫（S）量为0.32％。这一数值较传统方法，即采用全株或某一器官的干物重（DM）变化所求得的临界值，能更有效地反映植株（器官）的硫素丰缺状况。当再利用能力较弱的硫元素供应不足时，最先受到影响的是幼叶的扩展速率，而植株（或叶片）的干物量往往因净同化率（EA）和相对生长率（RW）在缺硫后的一定时间内相对增加而产生累积效应。因此，油菜叶片扩展的功能性需硫量可以作为油菜缺硫的诊断指标。

Improved capacity of genomics and biotechnology has greatly enhanced genetic studies in different areas. Genomic selection exploits the geno type-to-phe no type relationship at the whole-ge nome level and is being implemented in many crops? Here we show that design-thinking and data-mining techniques can be leveraged to optimize genomic prediction of hybrid performance? We phenotyped a set of 276 maize hybrids generated by crossing founder inbreds of nested association mapping populations for flowering time, ear height, and grain yield? With 10 296 310 SNPs available from the parental inbreds, we explored the pat? terns of genomic relati on ships and phenotypic variation to establish training samples based on clustering, graphic n etwork analysis, and genetic mating scheme ? Our analysis showed that training set desig ns outperformed random sampling and earlier methods that either minimize the mean of prediction error variance or maximize the mean of generalized coefficient of determination. Additional analyses of 2556 wheat hybrids from an early-stage hybrid breeding system and 1439 rice hybrids from an established hybrid breeding system validated the approaches. Together, we dem on strated that effective genomic predicti on models can be established with a training set 2%-13% of the size of the whole set, enabling an efficient exploration of enormous inf ere nee space of gen etic combi nations.

Long-term mental health outcomes after unintentional burns sustained during childhood:a retrospective cohort study

Background:Burns are a devastating injury that can cause physical and psychological issues. Limited data exist on long-term mental health (MH) after unintentional burns sustained during childhood. This study assessed long-term MH admissions after paediatric burns. Methods:This retrospective cohort study included all children (<18 years) hospitalised for a first burn (n=11,967) in Western Australia, 1980–2012, and a frequency matched uninjured comparison cohort (n=46,548). Linked hospital, MH and death data were examined. Multivariable negative binomial regression modelling was used to generate incidence rate ratios (IRR) and 95%confidence intervals (CI). Results:The burn cohort had a significantly higher adjusted rate of post-burn MH admissions compared to the uninjured cohort (IRR, 95%CI:2.55, 2.07–3.15). Post-burn MH admission rates were twice as high for those younger than 5 years at index burn (IRR, 95%CI 2.06, 1.54–2.74), three times higher for those 5–9 years and 15–18 years (IRR, 95%CI:3.21, 1.92–5.37 and 3.37, 2.13–5.33, respectively) and almost five times higher for those aged 10–14 (IRR, 95%CI:4.90, 3.10–7.76), when compared with respective ages of uninjured children. The burn cohort had higher admission rates for mood and anxiety disorders (IRR, 95%CI:2.79, 2.20–3.53), psychotic disorders (IRR, 95%CI:2.82, 1.97–4.03) and mental and behavioural conditions relating to drug and alcohol abuse (IRR, 95%CI:4.25, 3.39–5.32). Conclusions:Ongoing MH support is indicated for paediatric burn patients for a prolonged period after discharge to potentially prevent psychiatric morbidity and associated academic, social and psychological issues.

Aim: Increased inflammatory signaling in microglia is implicated in the pathogenesis of neurodegenerative diseases, trauma, psychiatric disorders, and anxiety/depression. Understanding inflammatory signaling in microglia is critical for advancing treatment options. Studying rodent-derived microglia has yielded substantial information, yet, much remains to better understand inflammatory signaling in human microglia. Hence, there is great interest in developing immortalized human microglial cell lines. The C20 human microglial cell line was recently developed and our primary objective was to advance our knowledge of inflammatory signaling in these cells. Methods: Expression of the microglia specific marker transmembrane protein 119 (TMEM119) was assessed by western blot analysis. Lipopolysaccharide (LPS)- and interleukin-1β (IL-1β)-induced cytokine/chemokine expression was determined by ELISA. Phosphorylation of inhibitory kappa B alpha (IκBα), nuclear factor (NF)-κB p65, and p38 mitogen-activated protein kinase (p38 MAPK) was measured by western blot analysis. Results: TMEM119 was expressed in unstimulated C20 cells, and to a greater extent in IL-1β-stimulated cells. IL-1βsignificantly induced IL-6, monocyte chemoattractant protein-1/CCL2, and interferon-γ inducible protein 10/CXCL10 expression. LPS induced CCL2 expression, but not IL-6 or CXCL10 expression. IL-1β induced inflammatory signaling as indicated by increased phosphorylation of IκBα, NF-κB p65 and p38 MAPK. Conclusion: We provide the first evidence that C20 microglia express TMEM119. This is the initial report of IL-1β-induced activation of IκBα, NF-κB p65, and p38 MAPK and subsequent CXCL10, CCL2 and IL-6 secretion in C20cells. These findings advance our understanding of inflammatory signaling in C20 cells and support the value of this cell line as a research tool.
